Effect of Plant and Animal Proteins on Biomarkers of Colorectal Cancer and Type 2 Diabetes in Healthy Adults (ScenoProt)

Project description:The aim of this study is to investigate the effects of dietary plant and animal proteins on gut metabolism and markers for colorectal cancer as well as blood protein metabolites and markers for type 2 diabetes in healthy adults. The study participants will be stratified into three groups with different protein composition in diets: 1) animal 70%/plant 30%; 2) animal 50%/plant 50% and 3) animal 30%/plant 70%. The participants will get part of their diet as ready foods or raw material to promote their compliance. The participants will also get personal advice for their diets. Blood, stool and urine samples will be collected in the beginning and in the end of the 12 week intervention, as well as phenotype measures like BMI, blood pressure and body composition. The participants will also fill food diary before and in the end of the intervention.

Project description:In this study, we designed a space simulation study, named “4 Subjects 180-Day Controlled Ecological Life Support System (CELSS) Integration Experiment”, which took place in Shenzhen, China, from June to December 2016. In this experiment, four subjects (3 males and 1 female) lived for 180 days in an enclosed simulated cabin, and multiple-sampling-point DNA methylation data was collected to conduct the epigenic analysis. Peripheral whole blood cells were extracted from all 4 subjects on the 12 sampling points (Pre45, Pre15, R2, R30, R60, R75, R90, R105, R120, R150, R175 and Post30 mission day during the experiment).

A chemical and synthetic biology approach to control plant membrane receptor kinase signaling

Project description:Plants have a large family of membrane receptor kinases (RKs) which sense extracellular signals to control plant growth, development, immunity, and stress response. The largest group of RKs contains an extracellular leucine-rich repeat (LRR) domain with over 200 members in Arabidopsis. However, the functional understanding of most of the LRR-RKs has been hampered by their genetic redundancy and the subtle phenotypes of RK overexpression. Here we show that the rapamycin-mediated heterodimerization of chimeric cytosolic kinase domains from receptor/co-receptor pairs in the plasma membrane can activate their downstream cellular signaling pathway, inducing the specific biological responses, including brassinosteroid, plant immunity, stomatal development, and lateral root development. This chemically controlled synthetic biology approach will be useful to investigate biological functions of LRR-RKs and their signaling pathways.

Interplay between m6A modification and transcripts quantity: Impacts on mRNA composition in plant stress granules

Project description:Stress granules (SGs) are cytoplasmic structures that emerge in response to unfavorable environmental conditions. The mechanisms governing the accumulation of transcripts in SGs are only partially understood. Despite the recognized role of m6A in plant transcriptome regulation, its impact on SGs' composition and assembly remains elusive. In L. angustifolius, SGs display a distinctive bi-zonal structure comprising of a ring and a central area with differences in ultrastructure and composition. Subsequent to the transcriptome analysis, specific mRNA were chosen for investigating their localization within SGs and assessing m6A levels. Transcripts of hypoxia-responsive genes (ADH1 and HUP7) showed significantly lower levels of m6A compared to housekeeping genes, but only ADH1 was absent in SGs. HUP7 mRNA, characterized by a low quantity of m6A, is present both in the SGs and cytoplasm, probably due to extremely high expression level. The high amount of m6A was observed only during the assembly of SGs. In mutants of A. thaliana with reduced levels of m6A, ECT2 was not observed in SGs, and poly(A) RNA levels and the number of SGs were slightly reduced. In summary, our findings demonstrate a limited impact of m6A modification on SGs assembly. However, the interplay between m6A modification and the overall transcript quantity in the cytoplasm appears to play a regulatory role in mRNA partitioning and assembly of SGs

The germ free mice colonized with human gut bacteria and four-plants derived nanoparticles uptaking bacteria were sorted with flow cytometry and identified with 16s rRNA next-generation sequencing.

Deciphering the biological processes underlying tomato biomass production and composition.

Project description:After tomato fruits harvesting huge amounts of biomass residues, including plant and immature fruit, remaining in the field can be utilized to produce bioenergy. Little is known about the molecular aspects underlying tomato plant biomass production and hydrolysis. To identify genes involved in the regulation of plant biomass accumulation and composition, two Solanum pennellii introgression lines (ILs) with contrasting phenotypes for plant architecture and biomass characteristics, were analyzed. A multiple approach aimed to characterize such near-isogenic lines was carried out for studying gene expression dynamics, microscopy cell traits and qualitative and quantitative cell wall chemical compounds variation. Transcriptomic analysis showed that the enhanced biomass production observed in IL2-6 line is due to a more effective coordination of chloroplast and mitochondria energy fluxes (photosynthesis, cell division, cell wall and hormone metabolism activation). In parallel, microscopy analysis revealed a higher number of cells and chloroplasts in leaf epidermis in the high biomass line whilst chemical measurements on the two lines pointed out striking differences in the cell wall composition and organization. Taken together, our findings shed light on the mechanisms underlying the tomato biomass production and open new routes for improving the tomato lignocellulosic processability.
