Project description:The algal holobiont, which consists of algae and the bacteria living on it, has developed strategies to handle environmental stresses, such as changes in temperature. The green alga Ulva (Chlorophyta) is home to bacteria that help the algae grow and develop, from germlings to mature plants. These bacteria release chemicals like thallusin, N-acyl homoserine lactone, and ectoine, which support the health and growth of the algae. However, there is limited research on the metabolome of cold-adapted Ulva holobionts, especially those from hard-to-reach regions like Antarctica, and how they respond to environmental changes. In this study, we examine the metabolites found in Ulva and its associated bacteria from Antarctica (Potter Cove) using high-resolution mass spectrometry. We identified small polar and zwitterionic metabolites, such as cysteinolic acid, ectoine, glutamine, glycerol, and proline, in the surface tissues of the algae with MALDI-MS. By profiling these metabolites using UHPLC-HRMS and ZIC-HILIC chromatography, we found that their levels change significantly in response to heat-stressed Antarctic Ulva holobionts. Our results suggest that environmental change can affect the interaction between seaweeds and their microbes by altering the metabolites involved in their chemical exchanges.
The Data contains the Ulva metabolome profiles (.mzXML) analyzed using ZIC-HILIC chromatography coupled with HRMS (QExactive). Acquisition was conducted with the polarity switch mode and ddMS acquisition was done on the QC pool samples. The Metadata, normalization factor and results files from CD 3.3 can be found in the supplementary files.

Project description:The transcript profiles of Nesterenkonia sp. AN1 grown at 5 ºC (Cold) and 21 ºC (Topt) were acccessed to evaluate the cold reposnse of this Antarctic Nesterenkonia strain. The strain was grown in triplicates at the optimum growth temperature of 21 ºC and a test temperature of 5 ºC. Total RNA was extracted from two replicate samples for each treatment condition and the total RNA was enriched for mRNA. RNA-seq was done using Illumina Miseq platform at Inqaba Biotech, South Africa. The reads were mapped against the genome sequence of Nesterenkonia sp. AN1 (obtained from NCBI database) and assesed for differeential gene expression using CLC Genomics Workbench 7.5.

Project description:The Western Antarctic Peninsula (WAP) is among the areas of the planet showing some of the most significant increases in air and water temperature. It is projected that increasing temperature will modulate communities of coastal ecosystems at species ecological performance and molecular composition. The main way that the organisms can cope with large thermal variation is by having a reversible phenotypic plasticity, which provides the organisms with a compensatory physiological response when facing challenging conditions. However, since Antarctic organisms have evolved in a very cold and stable environment. The giant Antarctic isopod Glyptonotus antarcticus is one of the most abundant in Antarctic waters. This species has a larval development inside of maternal marsupium, where juveniles have a short period to acclimate to environmental conditions after birth. In this sense, we hypothesize that juveniles exposed to unusual temperature increases even for short periods, would not respond adequately showing a narrow phenotypic plasticity. We assessed if early juveniles of G. antarcticus have the molecular plasticity when exposed to increased temperature at 5¡C during 1, 6, 12, and 24 hours in comparison to control 0¡C. Sequenced HIseq2000 libraries were compared between control and each treatment to detect differentially expressed transcripts. The main molecular pathways affected by thermal stress were antioxidants, proteases, endopeptidases, and ubiquitination transcripts which were up-regulated, and mitochondrial respiratory chain, cuticle, cytoskeleton, and a molt transcript which were down-regulated. Considering HSP transcript, only 3 were up-regulated at least in two points of the stress kinetic, without classical HSP70 and HSP90 transcripts. This study shows that juveniles of G. antarcticus do not show molecular phenotypic plasticity to cope with acute short-term heat stress, even for one or few hours of exposure without an eco-physiological capacity to respond. This may have consequences at the ecological population level, showing a reduced individual ability to survive decreasing population recruitment.

Project description:Cranberry (Vaccinium macrocarpon) is an important North American fruit crop with vulnerability to temperature extremes and a relatively recent domestication history. Hybridization with a cold-adapted crop wild relative (CWR), V. oxycoccos, offers a strategy for improving temperature stress tolerance. We conducted RNA sequencing (RNA-seq) on V. macrocarpon and F1 hybrids between V. macrocarpon and V. oxycoccos subjected to acute heat and cold stress, capturing early transcriptional responses up to 30 minutes (heat) and 95 minutes (cold) after treatment onset. We then evaluated differences in responses across genotypes and stress conditions. Differential expression analysis and functional profiling revealed cold-induced differences in pathways related to photosynthesis, ribosomes, defense, and hormone signaling. No subgenome-specific functional specialization was observed. Two F1 hybrids exhibited suggestive cold resilience, with expression changes elevated at 60 minutes but declining by 95 minutes. Hybrids also displayed substantial regulatory variation under stress and transgressive downregulation of photosynthesis genes under ambient conditions. These findings suggest that V. oxycoccos introgression could be utilized in breeding cold-tolerant cranberry cultivars. Variation observed between F1 hybrids reflects the diversity introduced through CWR germplasm and provides opportunities for selection. Conservation of V. oxycoccos and other CWRs remains critical for future crop improvement.
