Project description:Data used for the comparison of four different combinations of autosampler vials and small volume inserts for the analysis of per- and polyfluoroalkyl substances (PFAS) before and after extended storage times. Data is associated with analyses performed on the same day as sample preparation of isotopically labeled PFAS in neat solvent and analyses performed after one month in storage. These data were used to compare the repeatability of each combination, reproducibility between combinations, and changes in peak areas over time. All data were collected on an LC-IMS-MS platform with non-targeted data acquisition.

Project description:Per- and polyfluoroalkyl substances (PFAS) are a large group of persistent synthetic chemicals that are ubiquitous environmental contaminants. Mounting evidence demonstrates that PFAS can bioaccumulate and induce a suite of adverse health outcomes, including compromise of male reproduction. Despite this knowledge, there remains no clear consensus as to how PFAS elicit these responses. Accordingly, here we aimed to determine how an environmentally relevant cocktail of PFAS impact the fertility of male mice. After twelve weeks of continuous PFAS exposure, blood plasma was isolated for hormone and PFAS profiling and reproductive tissues and spermatozoa were subjected to a comprehensive battery of histological and functional analyses. These analyses revealed that PFAS exposure significantly reduced the rate of daily sperm production linked with reduced levels of circulating testosterone and dihydrotestosterone. Functional profiling of mature spermatozoa from PFAS exposed males failed to identify any overt changes in sperm viability, motility, DNA integrity, or ability to undergo capacitation and support in vitro fertilization and early embryonic development. PFAS exposed spermatozoa did, however, present with pronounced changes in their small non-coding RNA profile and were linked with significant dysregulation of early embryonic gene expression. These observations afford new mechanistic insight into how PFAS exposure impacts male reproductive health.

Project description:Per- and polyfluoroalkyl substances (PFAS) are a diverse family of industrially significant synthetic chemicals infamous for extreme environmental persistence and global environmental distribution. Many PFAS are bioaccumulative and biologically active mainly due to their tendency to bind with various proteins. These protein interactions may be the most important element in determining the accumulation potential and tissue distribution of individual PFAS. Trophodynamics studies including aquatic food webs present inconsistent evidence for PFAS biomagnification. This study strives to identify whether the observed variability in PFAS bioaccumulation potential among species could correspond with interspecies protein composition differences. Specifically, this work compares the perfluorooctane sulfonate (PFOS) serum protein binding potential and the tissue distribution of ten perfluoroalkyl acids (PFAAs) detected in alewife (Alosa pseudoharengus), deepwater sculpin (Myoxocephalus thompsonii), and lake trout (Salvelinus namaycush) of the Lake Ontario aquatic piscivorous food web. To identify interspecies differences in PFAS-binding serum proteins, fish sera were pre-equilibrated with PFOS, fractionated by serial molecular weight cut-off filter fractionation, followed by liquid chromatography–tandem mass spectrometry analysis of the tryptic protein digests and the PFOS extracts of each fraction. This workflow identified similar serum proteins for all fish species. However, serum albumin was only identified in lake trout, suggesting apolipoproteins are likely the primary PFAA transporters in alewife and deepwater sculpin sera. PFAA tissue distribution analysis provided supporting evidence for interspecies variations in lipid transport and storage, which may also contribute to the varied PFAA accumulation in these species.

Project description:Per- and polyfluoroalkyl substances (PFAS) are some of the most prominent organic contaminants in human blood that have the potential to disrupt biological processes and pathways in the liver. Although the toxicological implications from human exposure to perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) are well established, data on other, lesser-understood PFAS are limited. A challenge for regulatory authorities is to determine acceptable levels of human exposure to large, diverse classes of environmental contaminants. New approach methodologies (NAMs) that apply bioinformatics tools to interpret biological data are being increasingly considered to inform risk assessment when traditional toxicology methodologies are not amenable. The aim of the current investigation was to identify the biological responses relevant to PFAS mode of action as the concentration (benchmark dose) that these effects take place in order to utililze this information to inform/facilitate read-across for risk assessment of data-poor PFAS. A TempO-Seq platform (BioSpyder) measured gene expression changes in human liver microtissues (i.e., spheroids) after 1-day and 10-day exposures to increasing concentrations of PFAS. A bioinformatics framework was applied to 23 PFAS sub-classed into carboxylates (PFCAs), sulfonates (PFSAs), or PFAS precursors that were analyzed for the total altered transcripts, the concentration where effects take place, and identifying target genes of interest. Both PFCAs and PFSAs exhibited a trend toward increased transcriptional changes with carbon chain-length. Specifically, longer-chain compounds (7 to 10 carbons) were more likely to surpass liver-toxic transcriptomic thresholds established from previous studies than shorter chain PFAS. Longer-chain PFAS were also more potent, inducing transcriptional effects at lower concentrations. However, PFOS was the most potent PFAS and of all precursors, only PFOSA (a PFOS precursor) induced a response. The combined high-throughput transcriptomic and bioinformatics analyses revealed the capability of NAMs to assess the effects of PFAS in liver microtissues; such data improves our understanding of PFAS-related effects in humans and facilitates the use of read-across in human health risk assessment for other data-poor chemicals.

Project description:Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals utilized in various industrial settings, and include products such as flame retardants, artificial film-forming foams, cosmetics, non-stick cookware among others. Epidemiological studies suggest a link between increased blood PFAS levels and prostate cancer incidence, but the mechanism through which PFAS impact cancer development is unclear. To investigate the link between PFAS and prostate cancer, we evaluated the impact of metabolic alterations resulting from a high-fat diet combined with PFAS exposure on prostate tumor progression. Our studies are the first in the field to provide new and clinically relevant insights regarding novel metabolic and epigenetic states and support future development of effective preventative and therapeutic strategies for PFAS-induced prostate cancers. Our findings enhance understanding of how PFAS synergize with high-fat diets to contribute to prostate cancer development and establish an important basis to mitigate PFAS exposure.

Project description:Due to their environmental prevalence, persistence, and potential toxicity, per- and polyfluoroalkyl substances (PFAS) are contaminants of concern. However, except for a few well studied PFAS, hazard data required to evaluate their risk to aquatic ecosystems are limited. The present study used a 24-hour, high throughput assay to screen 22 PFAS with varying chain lengths and functional groups for effects on the transcriptome of a green algal species, Raphidocelis subcapitata. The tested PFAS included perfluoroalkyl carboxylic acids, perfluoroalkyl carboxylic acid ethers, alcohols, sulfonic acids, and sulfonamides. Algae were exposed in a 96-well microplate format to 8 concentrations (nominally, 100 to 0.03 uM) of each PFAS, in a 1/2 log dilution series. Analytical measurements of PFAS concentration were made at the initiation of exposure and after 24 hours. In addition to traditional endpoints (i.e., biomass, viability, and chlorophyll), RNA was extracted for sequencing of the transcriptome. Concentration response curves were fit to the resulting transcriptomic data to calculate gene-specific benchmark concentrations. The distribution of benchmark concentrations was subsequently used to calculate a transcriptomic point of departure (tPOD) which represents a concentration below which no concerted molecular change is detected in response to PFAS exposure. The resulting tPODs were compared with traditional endpoints and environmentally detected concentrations of specific PFAS.

Project description:This study used high-throughput transcriptomics (HTTr) to evaluate the gene expression effects of 133 per- and polyfluoroalkyl substances (PFAS) in HepaRG cells. The PFAS chemicals were selected using a category-based prioritization approach developed by the U.S. Environmental Protection Agency (EPA) to ensure representation of structurally diverse compounds with relevance to human exposure and regulatory interest (Patlewicz et al., 2019; https://doi.org/10.1289/EHP4555). Differentiated HepaRG cells were exposed to each PFAS in concentration-response format (0.1 to 100 µM) for 24 hours. Transcriptomic changes were measured using the TempO-Seq platform (BioSpyder Technologies), enabling expression profiling of approximately 20,000 human genes. The resulting gene expression data were used to characterize the concentration-dependent molecular responses to PFAS exposure using connectivity mapping and to derive transcriptomic points of departure (tPODs). This dataset contributes to a broader effort to screen the bioactivity of environmental chemicals using HTTr in human liver-derived cells. It supports the use of transcriptomic profiling to identify bioactive PFAS, elucidate affected molecular pathways, and inform chemical hazard assessment.

Project description:Per- and polyfluoroalkyl substances (PFAS) are a very large (thousands of chemicals) category; these substances have important industrial and consumer product applications. PFAS are highly persistent in the environment, and some are known to pose human health hazard. Regulatory agencies worldwide are considering restrictions and outright bans of PFAS; however, little data exists to make informed decisions. Therefore, a prioritization strategy is urgently needed for evaluation of potential hazards of PFAS. Structure-based grouping could expedite selection of PFAS for testing; still, the hypothesis that structure-effect relationships exist requires confirmation. We tested 26 structurally diverse PFAS from 8 groups in two human cell types from organs that are thought to be targets for PFAS. We used human induced pluripotent stem cell-derived hepatocytes and cardiomyocytes and tested concentration-response effects on both cell function and gene expression. Few phenotypic effects were observed in hepatocytes, but negative chronotropy was observed for 8 of 26 PFAS. Substance- and cell-dependent transcriptomic changes were more pronounced; however, little evidence of group-specific effects was observed. In hepatocytes, we found up-regulation of stress-related and extracellular matrix organization pathways, and down-regulation of fat metabolism. In cardiomyocytes, contractility-related pathways were most affected. Using these data, we derived phenotypic and transcriptomic point of departure estimates and compared them to predicted PFAS exposures. The conservative estimates for bioactivity and exposure were used to derive margin-of-exposure (MOE) for each PFAS. We found that most (23 of 26) PFAS had MOE>1. Overall, our data suggests that chemical structure-based grouping of PFAS may not be an appropriate strategy to predict their biological effects. This means that testing of the individual PFAS would be needed for confident decision-making. Our proposed strategy of using two human cell types and considering both phenotypic and transcriptomic effects, combined with dose-response analysis, may be used for prioritization of PFAS.
