Salt stress responses of Arabidopsis thaliana and Eutrema salsugineum (Thellungiella salsuginea)

Project description:Arabidopsis thaliana is a glycophyte with a low salt tolerance, while Eutrema is a halophyte with a very high salt tolerance. To elucidate the transcriptional basis of this difference, we performed hydroponis culture experiments where we grew plants under control conditions (25 mM NaCl) or under salt stress (200 mM NaCl for both species, 500 mM for Eutrema). Salt concentration was increased for the stress treatments by increments of 50 mM per day (25 mM on the first day). Plants were grown at the final NaCl concentration for an additional week, when rosettes were harvested for RNA isolation.Expression patterns were compared between treatments and between species.

Transcript profile analysis suggests marker genes to distinguish between Phase 0 and Phase 1 responses of Arabidopsis to salt stress

Project description:Most physiological and molecular mechanisms of salinity stress are researched based on salt shock conditions. However, salt shock doesn’t occur in agricultural practice or natural ecosystem. In the fields, salts accumulate gradually through high salt or sodium irrigation water and poor managements that allow ground water to rise to soil surface. Therefore, it is more reasonable to research salinity stress in a mild way (stepwise salt addition). The objective of this study is to select marker genes to differentiate between salt shock (Phase 0) and salt stress (Phase 1). Three replicates were used for all RNA-Seq experiments conducted on control, Phase 0 and Phase 1 samples in Arabidopsis thaliana. Phase 0 samples (rosette leaves) were harvested 1 hour later from 105 mM NaCl salt shock treatment; Phase 1 samples (rosette leaves) were harvested 2 days later after reaching 90 mM NaCl by a stepwise addition (15 mM NaCl per day).

Wt, ga1-3, penta seeds were sterilized, sown on MS agar plates then put for stratification for 3 days at 4degreeC. Plates were placed in growth cabinet for 9 days. Seedlings were transferred to 24 well-plates, with 2 seedlings per well (0.5 ml MS liquid per well). Plates were placed in the same condition for 3 days. Finally, NaCl were added (final concentration 200 mM), except for the control. The salt treatment was applied for 30 min and 1h. Treatment was stopped by freezing in liquid nitrogen. HAL2 overexpression induces iron acquisition and enhanced salt resistance in bdf1Δ

Project description:The strain bdf1Δ+HAL2 improved salt resistance of bdf1∆. To gain further insight into the mechanism of bdf1∆ salt sensitivity, DNA microarray analysis was performed to determine the reason for the salt sensitivity of bdf1∆ cells and the process of how HAL2 overexpression and HDA1 deletion improves salt resistance. Transcriptomic analysis under salt treatment (0.5 mol.L-1 NaCl for 45 min) was performed using four different strains: bdf1∆, W303, bdf1Δ+HAL2 and bdf1∆hda1∆. The transcription of 1,393 genes were significantly changed( > 2-fold) in bdf1∆ compared with W303 upon NaCl stress (0.5 mol.L-1 NaCl for 45 min). Only 108 genes were significantly changed ( > 2-fold) in bdf1Δ+HAL2 compared with bdf1∆ upon NaCl stress (0.5 mol.L-1 NaCl for 45 min). Only 88 genes were significantly changed ( > 2-fold) in bdf1Δ+HAL2 compared with bdf1∆ upon NaCl stress (0.5 mol.L-1 NaCl for 45 min).

Quantitative proteome, phosphoproteome and acetylome analysis of Arabidopsis thaliana root responses to persistent osmotic and salinity stress.

Project description:Environmental conditions contributing to abiotic stresses such as drought and salinity result in large annual economic losses around the world. As sessile organisms, plants cannot escape the environmental stresses they encounter, but instead must adapt to survive. Previous studies investigating osmotic and/or salt responses have largely focused on understanding short-term responses (0-1h) at the transcriptomic, proteomic and phosphoproteomic level; however, our understanding of intermediate to longer-term adaptation (24h - days) is relatively limited. In addition to protein abundance and phosphorylation changes, recent evidence suggests reversible protein acetylation may be important for abiotic stress responses. Therefore, to characterize the effects of osmotic and salt stress, we undertook a label-free proteomic and PTMomic analysis of Arabidopsis roots exposed to 300mM Mannitol and 150mM NaCl for 24 h. We quantitatively assessed protein abundance, phosphorylation and acetylation.
