GNPS - Differential Metabolomic Signatures in Plasma and Urine Under Mild and Moderate Hypothermia During Cardiopulmonary Bypass

ABSTRACT: Untargeted metabolomic profiling of plasma and urine samples from patients undergoing cardiopulmonary bypass (CPB) under mild or moderate hypothermia. Samples were collected at three time points: preoperative (T0), intraoperative (T1), and postoperative (T2). Both plasma and urine were analyzed using GC_MS and LC_qTOF_MS platforms. Plasma and Urine samples were extracted using cold methanol (9:1, v/v), and urine samples were pretreated with urease prior to extraction.

Comparison of gene expression signatures in human melanocytes after indirect cold physical plasma treatment and direct UV-irradiation

Project description:Cold atmospheric plasma is assembled out of various components such as UV-radiation, electrons, ions and an electromagnetic field at low temperature. In recent years cold atmospheric plasma gains interest as a medical tool used for improved healing of chronic wounds. In order to examine the impact of cold atmospheric plasma on the melanogenesis in human skin, the gene expression signature of differently treated lightly pigmented primary human melanocytes was analysed. Melanocytes were either treated in vitro indirectly by cold atmospheric plasma or irradiated by UV-radiation. After 2 h incubation the microarray analysis was performed.

Single cell RNAseq reveal covid19 patient immune response

Project description:Eighteen patients admitted to CSMC and diagnosed with COVID19 by RT-PCR were stratified into COVID19 mild/moderate, severe, and recovery groups (n=5-6/group). Venous blood was collected into EDTA coated tubes and centrifuged to separate plasma and buffy coat. Plasma was collected and frozen at -80C, and the buffy coat was collected into cryo-preservation media and frozen at -80C. Recovered cells are sorted for live-dead staining, then fixed with methanol. Fixed single cells were further captured using 10x chromium Next GEM 3 prime v3.1 kit. Two patients samples from the same group were mixed, captured and sequenced together.

RNA-seq Validation of MicroRNA Expression Signatures for Precision Melanoma Diagnosis and Prognostication

Project description:This study aimed to transition genomic signatures of invasive cutaneous melanoma (MEL38 for diagnosis and MEL12 for prognosis) from Nanostring to a high-throughput RNAseq platform using the gel-free NEXTflex Small RNA Library Prep Kit. We validated against 59 FFPE and 61 plasma samples from patients with melanoma and normal tissue . MEL38 accurately diagnosed invasive melanoma across different sample types and platforms, unaffected by disease stage or data origin. The MEL12 signature significantly stratified patients by survival rates in solid tissues, surpassing previous prognostic models based on DNA, mRNA, or protein analysis, with hazard ratios indicating high risk. In plasma, MEL12 also differentiated survival outcomes. Overall, the study confirms the reliability of MEL38 and MEL12 in predicting melanoma presence and prognosis, demonstrating their superiority and adaptability across specimen types and platforms, marking a substantial advancement in personalized melanoma management.
