ABSTRACT: This research aims to identify and describe molecular indicators of domoic acid events specific to the Santa Barbara Channel. Cathepsin X is a conserved cell death protein involved in algal response to environmental stress

Project description:Phytoplankton play a crucial role in global primary production and can form vast blooms in aquatic ecosystems. Bloom demise and the rapid turnover of phytoplankton are suggested to involve programmed cell death (PCD) induced by diverse environmental stressors. However, fundamental knowledge of the PCD molecular components in algae and protists in general remains elusive. Previously, we revealed that early oxidation in the chloroplast predicted subsequent cell death or survival in isogenic subpopulations that emerged in response to H2O2 in the diatom Phaeodactylum tricornutum. Here, we performed transcriptome analysis of sorted sensitive oxidized cells and resilient reduced cells, to discover genes linked to their contrasting fates. By cross-comparison with a large-scale mutant screen in the green alga Chlamydomonas reinhardtii, we identified functionally relevant conserved PCD gene candidates, including the cysteine protease cathepsin X/Z (CPX). CPX mutants in P. tricornutum CPX1 and C. reinhardtii CEP12 both exhibited profound resilience to oxidative stress, supporting a conserved function in algal PCD. P. tricornutum cpx1 mutants, generated using CRISPR-Cas9, also exhibited resilience to the toxic diatom-derived infochemical cyanogen bromide. Phylogenetic and predictive structural analyses show that CPX is highly conserved in eukaryotes, and algae of the green and red lineages exhibit strong structural similarity to human cathepsin CTSZ. CPX is expressed by diverse algae across the oceans and during toxic Pseudo-nitzschia blooms, supporting its ecological importance. Elucidating PCD components in algae sheds light on the evolutionary origin of PCD in unicellular organisms, and on the cellular strategies employed by the population to cope with stressful conditions.

Environmental palaeogenomic reconstruction of an Ice Age algal population.

Project description:Palaeogenomics has greatly increased our knowledge of past evolutionary and ecological change, but has been restricted to the study of species that preserve either as or within fossils. Here we show the potential of shotgun metagenomics to reveal population genomic information for a taxon that does not preserve in the body fossil record, the algae Nannochloropsis. We shotgun sequenced two lake sediment samples dated to the Last Glacial Maximum and reconstructed full chloroplast and mitochondrial genomes to explore within-lake population genomic variation. This revealed two major haplogroups for each organellar genome, which could be assigned to known varieties of N. limnetica, although we show that at least three haplotypes were present using our minimum haplotype diversity estimation method. These approaches demonstrate the utility of lake sedimentary ancient DNA (sedaDNA) for population genomic analysis, thereby opening the door to environmental palaeogenomics, which will unlock the full potential of sedaDNA.

AURATUS goldfish environmental genomics projects: dopamine depletion in hypothalamus and telencephalon

Project description:Sexually mature (mid-May) adult female goldfish received a single injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP; 50 ug/g body weight at 5 uL/g body weight) or 0.6% saline on day 0 and a single injection of alpha-methyl-p-tyrosine (aMPT; 240 ug/g body weight at 5 uL/g body weight) or 0.6% saline on Day 5 with the goal of severely deplete catecholamine (dopamine and norepinephrine) levels. Hypothalamus tissue was extracted on Day 6. Telencephalon tissue was extracted on Day 6. The objective of this study was to examine gene expression changes in the neuroendocrine brain in response to catecholamine depletion. A common reference design was used for both the hypothalamus and telencephalon groups: Hypothalamus: 3 independent groups of hypothalami from sexually mature female goldfish treated with 50 ug/g MPTP + 240 ug/g aMPT hybridized against a common vehicle control. An additional group was used for a dye-swap. Slides 4, 17, 21, 52.
