Amino acid conjugation detoxifies fusaric acid in Streptomyces sp. ATMOS43

Project description:Plant-associated streptomycetes are persistent in plant microbiomes, yet how they cope with pathogen-derived toxins remains poorly understood. Fusarium species produce fusaric acid (FA), a mycotoxin that contributes to virulence and perturbs plant-associated microbial communities. Here, we show that the Arabidopsis endophyte Streptomyces sp. ATMOS43 exhibits characteristic tolerance to FA. Metabolomics analysis revealed the formation of single amino acid and dipeptidyl conjugates of FA, including FA-Ser, a major conjugate that lacked detectable toxicity in bacterial and plant assays. Co-cultivation experiments showed that FA detoxification by ATMOS43 restores growth of sensitive streptomycetes in the presence of FA. This activity is enriched among plant-associated isolates, suggesting adaptation to toxin-rich environments. Proteomics and physiological analyses further indicate FA toxicity involves zinc chelation, which is abolished upon conjugation to FA-Ser, although additional cellular targets may also contribute. Together, our findings show that streptomycetes can protect plants not only by inhibiting pathogens themselves, but also by neutralizing pathogen-derived toxins.

Alanine scan of 11 amino acid long novel linear epitope from C. jejuni cj0669 protein

Project description:A short sequence of 11 amino acids belonging to the cj0669 protein from Campylobacter jejuni NCTC 11168, which was previously identified as potentially immunogenic, was analyzed via alanine scanning to narrow down the significant amino acid residues within the sequence. Twelve peptides, one representing the original sequence and eleven peptides with each residue replaced by alanine in turn, were synthesized on microarrays by JPTs Pepstar Technology. For each microarray, nine replicates for each peptide were spotted. The microarray was separated into three incubation chambers by the ProPlate 3-well module (Grace Biolabs) to allow for incubation with different antibodies in parallel. For specific interactions, rabbit polyclonal IgG to C. jejuni was used, while unspecific binding to the epitope sequence was checked using rabbit polyclonal IgG to Salmonella enterica.

A regulator of amino acid catabolism controls Acinetobacter baumannii gut colonization

Project description:Asymptomatic gut colonization increases the risk of clinical infection and transmission by the multidrug-resistant pathogen Acinetobacter baumannii. Ornithine utilization was shown to be critical for A. baumannii competition with the resident microbiota to persist in gut colonization, but the regulatory mechanisms and cues are unknown. Here, we identify a transcriptional regulator, AstR, that specifically activates the expression of the A. baumannii ornithine utilization operon astNOP. Phylogenetic analysis suggests that AstR was co-opted from the Acinetobacter arginine utilization ast(G)CADBE locus and is specialized to regulate ornithine utilization in A. baumannii. Reporter assays showed that astN promoter expression was activated by ornithine but inhibited by glutamate and other preferred amino acids. astN promoter expression was similarly activated by incubation with fecal samples from conventional mice but not germ-free mice, suggesting AstR-dependent activation of the astN promoter responds to intermicrobial competition for amino acids. Finally, AstR was required for A. baumannii to colonize the gut in a mouse model. Together, these results suggest that pathogenic Acinetobacter species evolved AstR to regulate ornithine catabolism, which is required to compete with the microbiota during gut colonization.
