Compararitive iTRAQ based proteomic changes in salt tolerant and salt sensitive cultivars of kenaf

ABSTRACT: In the present project we wished to see proteomic changes in salt sensitive (P3B）and salt tolerant （P3B）kenaf cultivars by using iTRAQ technology .The three weeks old kenaf seedlings of both the cultivars were exposed to 0 mM NaCl solutiom for control samples and 250 mM NaCl solution for salt stress treatment and incubation period was set for four days and then leaves were sampled after four days of treatment from controls and salt stressed kenaf seedlings for proteomic analysis by using iTRAQ technology.

High-throughput sequencing of small RNA transcriptome reveals salt stressed regulated microRNAs in sugarcane

Project description:Salt stress is a primary cause of crop losses worldwide, and it has been the subject of intense investigation to unravel the complex mechanisms responsible for salinity tolerance. MicroRNA is implicated in many developmental processes and in responses to various abiotic stresses, playing pivotal roles in plant adaptation. Deep sequencing technology was chosen to determine the small RNA transcriptome of Saccharum sp cultivars grown on saline conditions. We constructed four small RNAs libraries prepared from plants grown on hydroponic culture submitted to 170 mM NaCl and harvested after 1 h, 6 hs and 24 hs. Each library was sequenced individually and together generated more than 50 million short reads. Ninety-eight conserved miRNAs and 33 miRNAs* were identified by bioinformatics. Several of the microRNA showed considerable differences of expression in the four libraries. To confirm the results of the bioinformatics-based analysis, we studied the expression of the 10 most abundant miRNAs and 1 miRNA* in plants treated with 170 mM NaCl and in plants with a severe treatment of 340 mM NaCl. The results showed that 11 selected miRNAs had higher expression in samples treated with severe salt treatment compared to the mild one. We also investigated the regulation of the same miRNAs in shoots of four cultivars grown on soil treated with 170 mM NaCl. Cultivars could be grouped according to miRNAs expression in response to salt stress. Furthermore, the majority of the predicted target genes had an inverse regulation with their correspondent microRNAs. The targets encode a wide range of proteins, including transcription factors, metabolic enzymes and genes involved in hormone signaling, probably assisting the plants to develop tolerance to salinity. Our work provides insights into the regulatory functions of miRNAs, thereby expanding our knowledge on potential salt-stressed regulated genes.

De novo transcriptome characterization of Nitraria sibirica Pall. under salt stress

Project description:We will use high-throughput sequencing technology to study gene expression under NaCl treatment, screen for differential expressed genes, and then analyze the gene regulation features. Nitraria sibirica Pall. seedlings were treated by 0, 100 and 400 mM NaCl with 3 replicates，and the leaves were harvested after treated 3 days. Extracting total RNA of leaves and then sequencing by Illumina HiSeq 2000 sequencing platform. Our study provides a basis for the study of salt-tolerant gene resources in Nitraria sibirica Pall.

Microarray analysis of transcriptional responses to abscisic acid and salt stress in Arabidopsis thaliana

Project description:To investigate differences in plant responses to salt and ABA stimulus, differences in gene expression in Arabidopsis in response to salt and ABA were compared using an Agilent oligo microarray. Four-week-old Arabidopsis thaliana ecotype Columbia (Col-0) seedlings were treated with either 150 mM NaCl or 10 μM ABA for 6 hours; unstressed seedlings (control sample) were collected in parallel to avoid the possible effects of circadian rhythms. The results revealed that 31 genes were up regulated by both NaCl and ABA stress, and 23 genes were down-regulated by these stressors. To provide further validation of our microarray experiment data, ten genes from this signature were quantified in the same RNA samples by quantitative real-time PCR.

Characterization of differential gene expression profiles between wild-type Ler and proatcape1 mutant seedlings upon salt stress.

Project description:To gain insight into how AtCAPE1 regulates salt response, we investigated the gene expression profiles of wild-type (Ler) and proatcape1 mutant seedlings in the presence and absence of 125 mM NaCl by microarray analysis. Ten seedlings were grown vertically on the mesh attached on 1/2 MS medium for 10 days. Seedlings with the mesh were transferred to a new petri dish and then covered by buffer-saturated filter papers with 1/2 MS liquid medium (Control) or with 125 mM NaCl (Salt). Salt-treated seedlings (n=10 for each treatment) were sampled after 12 h. Three independent experiments were performed for the microarray analysis.

Project description:A comparative transcriptomic study of the impact of salt toxicity on rice plant (Oryza sativa L.; cv 'I Kong Pao') after short term (48 hours) exposure to NaCl (200 mM) or Na2SO4 (100 mM). Twenty five days old rice seedlings were exposed to 0, 200 mM NaCl or 100 mM Na2SO4 for 48 hours in hydroponic culture. Comparison between control and salt-stressed plants were done at the shoot and the root levels. The essays were replicated twice on two independent plant cultures.

Transgenic and conventional cultivar comparison in response to salt stress

Project description:Soybean is one of the main sources of oil worldwide. Salinity severely affect its yield. GmSIN1 is a NAC transcription factor coding gene. Its overexpression (OE) transgenic lines greatly improved the yield in both common and saline fields. This study focuses on founding changes genes between GmSIN1 OE transgenic seedlings and control seedlings under salt stress or non-salt stress conditions. Illumina Solexa sequencing platform was used for the comparative analysis of transcriptome profiles in the roots and leaves of GmSIN1 OE transgenic seedlings and WEI6823 (control) seedlings under mock or 150 mM NaCl treatment for 6 hrs.
