Data independent acquisition of Pyropia haitanensis under salt stress

ABSTRACT: The worsening of soil salinization has become an important factor limiting crop yield and quality. Pyropia haitanensis grows in the intertidal zone, and has extremely high salt tolerance. The response mechanism of organisms exposed to stress is linked to energy metabolism and protein processing. The Sm Core Protein SmEb Regulates Salt Stress Responses Through Maintaining Proper Splicing of RCD1 Pre-mRNA in Arabidopsis

Project description:Salt stress caused by soil salination inhibits plant growth and development that result in reduction of crop yield and threaten the food security. Several spliceosome components are considered to modify salt stress responses in plants. However, the molecular basis of spliceosome proteins adjustment to salt stress is still unclear. Here we report that an Sm core protein SmEb is required for salt tolerance in Arabidopsis. In addition, SmEb controls alternative splicing of hundreds of pre-mRNA to participate in plant response to salt stress. Our results further reveal that SmEb takes effect on maintain proper ratio of two RCD1 splicing variants to adjust to H2O2 accumulation under salt stress. Together, our findings uncover that proper alternative splicing of pre-mRNAs governed by the spliceosome component SmEb is essential for plant salt stress responses. Integrated transcriptomic and proteomic analysis of Tritipyrum provides insights into the molecular basis of salt tolerance

Project description:Soil salinity is a major environmental stress that restricts crop growth and yield. Here, crucial proteins and biological pathways were investigated under salt-stress and recovery conditions in Tritipyrum “Y1805” to explore its salt-tolerance mechanism. In total, 44 and 102 differentially expressed proteins (DEPs) were identified in “Y1805” under salt-stress and recovery conditions, respectively. A proteome-transcriptome-associated analysis revealed that the expression patterns of 13 and 25 DEPs were the same under salt-stress and recovery conditions, respectively. “Response to stimulus”, “antioxidant activity”, “carbohydrate metabolism”, “amino acid metabolism”, “signal transduction”, “transport and catabolism” and “biosynthesis of other secondary metabolites” were present under both conditions in “Y1805”. In addition, “energy metabolism” and “lipid metabolism” were recovery-specific pathways, while “antioxidant activity”, and “molecular function regulator” under salt-stress conditions, and “virion” and “virion part” during recovery, were “Y1805”-specific compared with the salt-sensitive wheat “Chinese Spring”. “Y1805” contained 83 specific DEPs related to salt-stress responses. The strong salt tolerance of “Y1805” could be attributed to the strengthened cell walls, reactive oxygen species scavenging, osmoregulation, phytohormone regulation, transient growth arrest, enhanced respiration, ammonium detoxification, transcriptional regulation and error information processing. These data will facilitate an understanding of the molecular mechanisms of salt tolerance and aid in the breeding of salt-tolerant wheat.

RNA-seq Analysis of the Salt Stress Induced Transcripts in Fast Growing Bioenergy Tree, Paulownia elongata

Project description:Paulownia elongata is a fast-growing tree species native to China that is grown in different climates, types of soils, and can be easily re-grown. The versatility of the P. elongata species makes it an ideal candidate for biofuel production. High soil salinity is known to inhibit plant growth dramatically or lead to death. Salinity in soil is a detrimental abiotic stress affecting crop production worldwide and a hindrance for potential crop candidates used for biofuel production. The purpose of this study was to characterize the salt-induced transcriptome of P. elongata. Transcriptome differences in response to salt stress were determined by RNA sequencing (RNA-seq) using next generation sequencing and bioinformatics analysis. A total of 646 genes were found to have significant altered expression in response to salt stress, and expression levels of a selective subset of these genes were chosen and confirmed using quantitative real-time PCR. To the best of our knowledge, this is the first report of salt-induced transcriptome analysis in P. elongata. The current study indicates that the differential expression of certain genes may have an important role in the adaptation of P. elongata in response to salt stress. Functional characterization of these genes will assist in future development of salt tolerance in P. elongata, which could be used to enhance biofuel production.

Soil salinity and mechanical obstruction differentially affects embryonic root architecture in native rice varieties of West Bengal

Project description:Roots make the first contact with the soil environment and are the first responders of stress. These root behaviors are quantifiable and adaptive. The response of rice varieties in mechanical and salinity stress was measured in a novel experimental setup that mimics the soil environment. We analyzed the response of roots by means of SAC (Stress Adaptation Coefficient) in 28 rice varieties that include high-yield salt tolerant varieties as well as geographically isolated native rice varieties. cDNA microarray of IR64 root-tip shows about 6000 common transcripts to be differentially regulated among the two stresses and common pathways were identified. Overall, our study indicates that there is an important commonality in the molecular basis of salt and mechanical stress and presents an easy-to-perform early establishment stress screen for rice varieties.

Integrated physiological and comparative proteomics analysis of contrasting genotypes of pearl millet reveals underlying salt-responsive mechanisms

Project description:Salinity stress poses a significant risk to agricultural yield and productivity. Therefore, elucidation of plant salt-response mechanisms has become essential to identify stress-tolerance genes. In the present study, two pearl millet genotypes with contrasting salt-tolerance showed differential morpho-physiological and proteomic responses under 150 mM NaCl, and the genotype IC 325825 could withstand salt-stress better than IP 17224. The salt-tolerance potential of IC 325825 was associated with its ability to maintain ionic, osmotic balance and membrane integrity under stress. The IC 325825 exhibited better growth under salinity as compared to IP 17224 due to higher expression of C4 photosynthesis enzymes, efficient antioxidant system, and lower Na+/K+ ratio. Comparative proteomics analysis revealed greater metabolic perturbation in IP 17224 under salinity, in contrast to IC 325825 that harbored pro-active stress-responsive machinery, allowing its survival and better adaptability under salt-stress. The differentially expressed proteins were in-silico characterized for their functions, subcellular-localization, pathway/ interaction analysis, and relative transcript levels. This study has provided novel insights into salinity stress adaptive mechanisms in pearl millet, demonstrating the power of proteomics-based approaches. The critical proteins identified in the present study could be further explored as potential objects for increasing salt-tolerance in sensitive crop plants.
