Transcription profiling of human samples from patients with acute lung injury plus sepis vs sepsis controls to investigate interactions between genes involved in ALI/ARDS

Project description:The Acute Respiratory Distress Syndrome (ARDS)/Acute Lung Injury (ALI) was described 30 years ago, yet the interaction between specific sets of genes involved in this syndrome remains incompletely understood. Experiment Overall Design: 13 patients with ALI + sepsis and 21 patients with sepsis alone were recruited from the Medical Intensive Care Unit of the University of Pittsburgh Medical Center between February 2005 and June 2007. Whole blood was obtained from each patient within 48 hours of admission, and RNA was extracted for gene expression profiling, and comparison analysis.

Prostate cancer specific gene expression

Project description:To identify prostate cancer-specific gene expression, we have employed whole cDNA microarray expression profiling between benign prostate hypertrophy and prostate cancer. We performed needle biopsy of patients with prostatic hyperplasia who were suspected as prostate cancer for minor high level (4.0-10 ng/ml). As a result of needle biopsy, we used the tissue of patients with prostatic hyperplasia that prostate cancer was denied for cDNA microarry analysis. Furthermore, we also performed needle biopsy of patients that prostate cancer should have been strongly suspected because of high PSA. Needle biopsies were performed after we obtained informed consent from patients with the document approved from the Graduate School of Medical Science, Kanazawa University.

Comparison between matched microdissected and whole tissue breast cancer specimen

Project description:A total of 38 flash-frozen breast cancer tissues were collected from three medical centers in the Netherlands (i.e. National Cancer institute: n=19; Radboud University Medical Center: n=4; Erasmus University Medical Center: n=15). All tissues were analyzed as laser capture microdissected (LCM) and whole samples (whole tissue lysate: WTL). All gathered samples were primary estrogen receptor (ER) tumors, which were subdivided in two patient groups according to outcome to first line (i.e. recurrent disease setting) tamoxifen therapy based on time-to-progression (TTP): patients which manifested progression of disease before (i.e. TTP <=) 6 months were defined as "poor outcome", while patients which manifested progression after (i.e. TTP>) 6 months were defined as "good outcome". All patients did not receive adjuvant hormonal therapy (i.e. tamoxifen or aromatase inhibitors post-surgical resection of primary tumor). The LCM subset derives from PXD000484 (Erasmus Medical Center samples) and PXD000485 (National Cancer Institute and Radboud University Medical Center).

Project description:Interventions: Group 1: Colon tissue (tumor, control tissue and tissue from diverticulitis patients) which is optained within a surgical intervention at the Asklepios Paulinen Clinic in Wiesbaden is directly transferred to the Clinic of Pathology of the Helios Dr.-Horst-Schmidt-Clinic Wiesbaden, where it is categorized. Within 24 h the tissue is collected from a staff of the Insitute of clinical Pharamcology University of Frankfurt. At the Institute of clinical Pharmacology of the University hospital Frankfurt the tissue is prepared for immunohistochemistry, for detection of lipids by liquid-chromatography-tandem mass spectrometry, for single cell isolation, FACS analysis, and in vitro culture experiments. DCK (diquat)-induced lung injury

Project description:Diquat (DQ) poisoning is a severe medical condition with life-threatening implications and multi-organ dysfunction, yet its underlying mechanism remains incompletely understood. This study unveils a critical process in DQ-induced toxicity. DQ disrupts the mitochondrial genome stability in endothelial cells, leading to the accumulation of Z-form DNA. This triggers an interaction between Z-DNA binding protein 1 (ZBP1) and receptor interacting protein kinase 3 (RIPK3), setting off RIPK3-dependent necroptotic and ferroptotic signaling pathways. Depletion of ZBP1 or RIPK3 in endothelial cells effectively inhibits both necroptosis and ferroptosis, reducing organ damage and mortality. Importantly, we discover that RIPK3 plays a dual role. It phosphorylates MLKL to induce necroptosis and phosphorylates FSP1, inhibiting its enzymatic activity and promoting ferroptosis. The phosphorylation of T163 in FSP1 is crucial for suppressing its activity. Combining the deletion of MLKL with vitamin K treatment proves highly effective in mitigating multi-organ damage and lethality caused by DQ.
