ITRAQ-based Quantitative Proteomic Analysis of Thoracic Aortic from Adult Rats Born to Pre-eclamptic Mothers


ABSTRACT: Epidemiological studies suggest that both preeclampsia and gestational hypertension could lead to offspring with vascular dysfunction. Previously, we have analyzed the protein profile of umbilical artery tissue from preeclampsia patients. In order to gain insights into the mechanisms of vascular dysfunction in the adult offspring of preeclampsia, we present analysis thoracic aortic from adult rats born to pre-eclamptic dams. By using iTRAQ method, we analyzed 1825 proteins in which 106 proteins have shown significantly differential expression in thoracic aortic. Subsequent analysis of the data by ingenuity pathway analysis (IPA) shows that the majority of differentially expressed proteins are related with cardiovascular function. Further analysis indicates the glucose-6-phosphate dehydrogenase (G6PD), which is inhibited by miR-423-5p and activated by TP53, was most affect the cardiovascular function. The expression level of G6PD up-regulated in thoracic aortic tissues and the result has been further verified by western blotting. Another two vascular function related proteins Cysteine and glycine-rich protein 2 (CSRP2) and Tubulin alpha-4A (TUBA4A) were also proved to be increased by both mass spectrometry (MS) based on quantification results and western blotting. Although the results need further functional validation, these data provide the first evidence for new insights into the mechanisms of vascular dysfunction in the adult offspring of mothers with preeclampsia.
