Inherited thrombocytopenia 2 (THC2) is difficult to diagnose due to the lack of specific clinical characteristics and diagnostic methods. Project description:In the early stages of acute pancreatitis, diagnostic sensitivity is limited and prognosis is difficult to determine. We provide a new idea for the diagnosis and prognosis of AP by studying the expression of plasma miRNAs in patients. Plasma samples from 22 normal subjects and AP patients at different stages were screened for miRNAs sequencing. A total of 27 miRNAs were found to be differentially expressed. Then we performed downstream target gene prediction for differentially expressed miRNAs. Analysising of KEGG, GO, GSEA and network interaction, it was found that its therapeutic effect was closely related to apoptosis, inflammatory response and autophagy process.

Project description:Tuberculosis (TB) is difficult to diagnose under complex clinical conditions. Exosomal miRNAs have emerged as promising disease biomarkers. We aim to investigate the potential of exosomal miRNAs to assist with TB clinical diagnosis. In the present research, we used the Affymetrix Genechip miRNA 4.0 Array to investgate the profiles of differentially expressed miRNAs (DEMs) in the exosomes of peripheral blood plasma. As a result, exosomal miRNA profiling yielded a total of 102 DEMs (98 with up-expression and 4 with down-expression) between the TB (pulmonary tuberculosis and tuberculosis meningitis) patients and controls.

Project description:In this study, we examined if the composition of plasma miRNAs is altered in patients with traumatic brain injury (TBI), and if these changes can be used as diagnostic markers. A microarray containing 875 human miRNAs was used to compare the miRNA profile of plasma collected from severe TBI patients (GCS ≤ 8) to that of age-, gender-, and race-matched healthy volunteers. This screen identified 108 miRNAs in the plasma of healthy volunteers. Of these, 52 were found to be altered in plasma samples from persons with severe TBI, and an additional 8 miRNAs were detected only in the plasma of TBI patients.

Project description:When faced with clinical symptoms of scarring alopecia – the standard diagnostic pathway involves a scalp biopsy which is an invasive and expensive procedure. Furthermore, clinical activity of scarring alopecias is often difficult to assess as symptoms of permanent damage and signs of activity can overlap or be difficult to distinguish. Here we report that gene expression analysis of only a small number of hair follicles (HF) plucked from lesional areas of the scalp is sufficient to characterise chronic discoid lupus erythematosus (CDLE). Lesional and non-lesional HFs were extracted from the scalp of patients with CDLE, psoriasis and healthy controls. The expression profile from CDLE HFs coincides with published profiles of CDLE from skin biopsy and was consistent with histopathological diagnostic features of CDLE. We therefore propose that information obtained from this non-invasive approach are sufficient to diagnose scalp LE.

Project description:In order to explore the role of circRNA in diabetic kidney disease (DKD), we employed circRNAs microarray expression profile in the plasma of daibetes patients (n=6), diabetic kidney disease patients (n=6) and healthy controls (n=6). According to the microarray data, we found 2079 differentially expressed circRNA in the plasma of DKD patients compared to the healthy people, including 1182 up-regulated and 897 down-regulated.Among the differentially expressed circRNA, nine DKD-related circRNAs were chosen and further validated by qRT-PCR.The results revealed that circUBXN7 was significantly elevated in the plasma of DKD patients, and might be a diagnostic plasma marker for DKD.

An alternative approach that consists in measuring transcriptome responses in reporter cells exposed in vitro to patient plasma has been successfully employed in a limited number of studies. Here we devised such a “Transcriptomic Reporter Assay” system to assess the immunogenicity of plasma from septic patients and evaluate its potential for biomarker discovery. Sepsis is a common, severe systemic infectious process for which physicians still lack efficient diagnostic or prognostic tools. Of the three different cell reporter systems tested, neutrophils were identified as the most capable “plasma sensor”. Compared to peripheral blood mononuclear cells and dendritic cell preparations neutrophils were best able to discriminate between plasma from septic and control subjects and responded by upregulating a robust immune transcriptional program. Additionally, the amplitude of the neutrophil transcriptomic response was shown to be associated with disease severity in two additional sets of patients. Overall, our results demonstrate both the suitability and potential clinical relevance of a neutrophil reporter assay for assessing immunopathogenic processes in a complex and severe condition such as sepsis. Plasma samples from 10 patients from either severe TBI (experimental group) or healthy volunteers (reference group; age-, gender-, and race-matched ) were pooled, the total RNA extracted in parallel, eluted in 100ul, and dried to 30 ul. Equal volumes of extracted plasma RNAs were assayed for global miRNA content using a service provider (LC Sciences, Houston, TX). There were no replicates performed for this screen. Healthy volunteer group served as the reference.
