Effect of Epicatechin on LPS-induced MAC-T cells by proteomics

ABSTRACT: Mastitis, a most common disease affecting dairy cows worldwide, causes great economic losses to the dairy industry. And, Epicatechin belongs to natural polyphenols, which are abundant in cocoa, grapes, tea and many other fruits and vegetables. We sought to identify Epicatechin-regulated on MAC-T cells by proteomics.

RNA-Seq data from dairy goat milk somatic cells in response to fish oil diet induced milk fat depression

Project description:Dietary supplementation with fish-oil modulates ruminant milk composition towards a healthier fatty acid profile for consumers, but it also causes milk fat depression (MFD). Because the dairy goat industry is mainly oriented towards cheese manufacturing, MFD can elicit economic losses. There is large individual variation in animal susceptibility with goats more (RESPO+) or less (RESPO−) responsive to diet-induced MFD. Thus, we used RNA-Seq to examine gene expression profiles in mammary cells to elucidate mechanisms underlying MFD in goats and individual variation in the extent of diet-induced MFD.

MiRNA-seq analysis of 15 year old bovine serum miRNAs in the presence of Mycobacterium avium paratuberculosis infection.

Project description:Mycobacterium avium subspecies paratuberculosis (MAP) is capable of causing a chronic enteritis known as Johne's disease (JD) in ruminants, notably resulting in substantial economic losses within the dairy industry. Prognostic and diagnostic assays that are both sensitive and specific are required to limit the spread of infection. Circulating microRNAs (miRNAs) have shown potential as biomarkers for various pathologies. We had access to a MAP infection model that consisted of serum samples from unchallenged, silent and subclinical phases in cattle collected across a four year period. Hence, the first aim of this study was to determine whether small RNAs could be sequenced from one ml volumes of serum using this model. We sought to identify potential prognostic miRNA biomarkers of MAP infection. We also had the opportunity to confirm the stability and longevity of circulating miRNAs as the serum samples that had been stored at -20degree C for approximately 10-15 years.

Project description:Up to 50% of ovulations go undetected in modern dairy herds due to attenuated oestrus behavior and a lack of high-accuracy methods for detection of fertile oestrus. This significantly reduces overall herd productivity and constitutes a high economic burden to the dairy industry. MicroRNAs (miRNAs) are ubiquitous regulators of gene expression during both health and disease and they have been shown to regulate different reproductive processes. Extracellular miRNAs are stable and can provide useful biomarkers of tissue function; changes in circulating miRNA profiles have been reported during menstrual cycles. This study sought to establish the potential of circulating miRNAs as biomarkers of oestrus in cattle using Illumina small-RNA sequencing. In this experiment a total of 24 bovine plasma samples were analysed using Illumina miRNA sequencing. These samples were collected at three time-points during a normal oestrous cycle from each of 8 animals (Day 0, Day 8 and Day 16 after oestrus) in a repeated-measures design. Therefore this study included 8 biological replicates for each of the three time-points analysed.

Integrated host and viral transcriptome analyses reveal pathology and inflammatory response mechanisms to ALV-J injection in SPF chicken

Project description:Avian leukosis virus (ALV) causes substantial economic losses from mortality and decreased performance in poultry industry. To characterize the response to ALV challenge, we developed a novel methodology that combines four datasets: mRNA expression and their associated regulatory factors of miRNA and lncRNA, and ALV gene expression. Specific Pathogen-Free (SPF) layer chickens were assigned to the ALV-infected or control group. Spleen samples (n=6) were collected at 40 days post injection (dpi), and sequenced. Comparing the infected and non-infected groups, 864 genes, 7 miRNAs and 17 lncRNAs were differentially expressed.

The Small RNA sequencing of 12 mammary gland for Chinese Holstein Cattle

Project description:Milk protein is one of the most important economic traits in the dairy industry. Yet, the miRNA gene regulatory network for the synthesis of milk protein in mammary is poorly understood. In this study, the hypothesis was that miRNAs have potential roles in bovine milk protein production. Using miRNA-seq and RNA-seq, we investigated the miRNAs profiles of mammary glands from 12 Chinese Holstein cows with six cows at peak of lactation and six in non-lactating period, from which three cows were in high and three in low milk protein percentage.
