Microarray analysis of expressed genes from R. microplus larvae, adult ovary, adult female gut associated with B bovis.

Project description:Babesia bovis exposed tissues: Three tissues were looked at. 1. Adult Female Gut 2. Adult Ovary 3. Larvae. In all there are 24 measurements for feature (EST), and 4 measurements per treatment for each of the 6 treatment groups. The 6 treatment groups are: Gut infected (GI), Gut control (GC), and similarly for Ovary and Larval: OI, OC, LI, LC. There are 12 chips, each with a spot replicate. **Note: contact person: Felix D. Guerrero email: felix.guerrero@ars.usda.gov Keywords: Rhipicephalus (Boophilus) microplus, Babesia, microarrays. In the study presented here, 12 hybridizations were performed. Expression profiles for 13601 unique ESTs were analyzed. Uninfected ticks were collected in one batch, as were the infected ticks. Replicates performed are technical.

RNAseq analysis of antibacterial mechanism of Cinnamomum camphora essential oil against Escherichia coli

Project description:Transcriptome analysis plays a central role in elucidating the complexity of gene expression regulation in Escherichia coli. By analyzing the transcriptomics of E. coli treated with water, acetone, and Cinnamomum camphora essential oil, the inhibitory mechanism of the essential oil on the human intestinal microbe was studied. The results showed that the inhibitory effect of the essential oil on E. coli increased with an increase in concentration; 1/4 minimum inhibitory concentration was the reaction equilibrium point. RNAseq transcriptomic comparison indicated that the essential oil inhibited the growth of E. coli by inhibiting the metabolism, chemotaxis, and some resistance reactions of them, while E. coli maintained its life activities by enhancing its resistance reactions. These results are of great importance to the study of the medical use of C. camphor essential oil and gene regulation in E. coli under stress conditions.

Haemaphysalis longicornis salivary gland LC-MSMS

Project description:Ticks are notorious carriers of pathogens; these blood-sucking arthropods can spread a variety of deadly diseases. The salivary gland is the main organ in ticks, and this organ begins to develop rapidly when Ixodidae ticks suck blood. When these ticks reach a critical weight, the salivary glands stop developing and begin to degenerate. Specific developmental features of the salivary glands are regulated by multiple factors, such as hormones, proteins and other small molecular substances. In this study, we used iTRAQ quantitative proteomics to study dynamic changes in salivary gland proteins in female Haemaphysalis longicornis at four feeding stages: unfed, partially fed, semi-engorged, and engorged. Through bioinformatics analysis of a large number of proteins, we found that molecular motor- and TCA cycle-related proteins play an important role during the development of the salivary glands. The results of RNAi experiments showed that when dynein, kinesin, isocitrate dehydrogenase, and citrate synthase were knocked down, ticks were unable to suck blood normally. The structure and function of the salivary glands were also significantly affected. In addition, four proteins from H. longicornis were found to have very low homology with those from mammals, including humans. Therefore, it is expected that drugs or antibodies targeting these unique sequences can be designed to kill ticks.

Rational identification of novel drug targets and vaccine candidates against the cattle tick, Rhipicephalus microplus

Project description:Control of complex parasites via vaccination remains challenging, with the current combination of vaccines and small drugs remaining the choice of control strategy. The development of new vaccines against complex endo- and ectoparasites remains challenging and more studies are providing evidence that multicomponent vaccines will be needed for the development of protective vaccines against parasites. The development of multicomponent vaccines, require an in-depth understanding of parasite biology which remains insufficient for ticks. With the rapid development and spread of acaricide resistance in ticks, new targets for acaricide development also remains to be identified, along with novel targets that can be exploited for the design of lead compounds. In this study, we analysed the differential gene expression of Rhipicephalus microplus ticks that were fed on cattle vaccinated with a multi-component vaccine (Bm86 and 3 putative Bm86-binding proteins). The data was scrutinised for the identification of vaccine targets, small drug targets and novel pathways that can be evaluated in future studies. Limitations associated with targeting novel proteins for vaccine and/or drug design is also discussed and placed into the context of challenges arising when targeting large protein families and intracellular localised proteins. Lastly, this study provide insight into how Bm86-based vaccines may reduce successful uptake and digestion of the bloodmeal and overall tick fecundity.
