Project description:Knockout mutants of polyketide synthase genes were generated (pks1, pks2, pks3, pks14, pks18, pks37) and compared to wild-type AX2 strain of Dictyostelium discoideum using Agilent custom microarrays. The Dictyostelium cells were harvested at eight developmental stages (Amoeboid, Intiation of starvation, loose aggregate, Streaming, Mound, Slug, early culminant, Fruiting body) and subjected to microarray based expression profiling.

Transcriptomic comparison of Botrytis cinerea wild type and VELVET mutants

Project description:An RNAseq-based transcriptomic comparison of Botrytis cinerea wild type and the three VELVET mutants was performed, to correlate common virulence defects of the mutants with changes in gene expression. A large number of genes encoding infection-related and secreted proteins was strongly underexpressed in the mutants, in particular genes encoding proteases and carbohydrate active enzymes. Many VELVET-regulated genes were found to be upregulated in the wild type during host plant infection (Müller et al.: Investigations on VELVET regulatory mutants confirm the role of host tissue acidification and secretion of proteins in pathogenesis of Botrytis cinerea; New Phytologist, 2018)

RNA-seq of Xenopus tropicalis rax mutants and wild type siblings

Project description:RNA was isolated from Xenopus tropicalis rax mutants and wild type siblings. This was used to generate mutant and wild type RNA libraries for solexa sequencing. The sequencing data will be compared to isolate changes that may be caused by loss of rax activity. The RNA samples were extracted with Trizol, then were DNAse treated following the Invitrogen DNAse I protocol and re-precipitated with ethanol. This data is part of a pre-publication release. Liver and Plasma metaboites for 13C-glucose load in wild type, LIRKO GTT 1 mice

Project description:Mice were fasted for 18 hr overnight then sacrificed or treated with 13C-U-glucose (2 g/kg ip) and sacrificed 1 hr later by decapitation and liver was immediately freeze-clamped and stored in liquid N2 and then at -80 C. Wild type (IR and IR/FoxO1 floxed) mice were sacrificed after fasting and 1 hr post-glucose treatment. Comparing gene expression between wild type and trichomeless mutants

Project description:Trichomes are found on the leaf surface of many plant species. They can be classified into either glandular or non-glandular types. Glandular trichomes are usually multicelluar structures and have long been known to participate in defense against pathogens through the production of specialized chemical products. Non-glandular trichomes, such as those in Arabidopsis thaliana, have been hypothesized to have roles in physical defense against insects, UV protection, detoxification, and heat dispersal. However, it has been difficult to separate a physical role from other possible functions for non-glandular trichomes. We have taken advantage of several mutants in the trichome developmental pathway and gene expression analyses to identify a set of genes expressed predominantly in Arabidopsis trichomes.
