Proteome of Tcte1 deficient mouse sperm

Project description:Sperm motility is one of the most important determinants of male fertility. Elucidating the different expression proteins of nomal and asthenospermia spermatozoa should advance our understanding of the underlying molecular mechanisms. Tcte1 null mouse sperm reduced motility and failed to display the forward progressive motile. With tandem mass tag (TMT) labeling and mass spectrometry analysis， 3697 proteins were quantified from Widetype and Tcte1 deficient mouse spermatozoa. Of these proteins, 447 were found to be different expression proteins (DEPs), including 398 proteins down regulated (DDEPs, 89%) and only 49 proteins up regulated.

Proteomic analysis of Tmem232-/- and wild type mice sperm

Project description:The sperm were harvested from caudal epididymides of 8-week-old Tmem232-/- and wild-type adult male mice, and subjected to LC-MS/MS analysis.The total proteins of 5,199 were identified and 4,380 of them were quantified. Results showed that 385 differentially expressed proteins were identified including 88 up-regulated and 255 down-regulated genes (Fold Change≥1.5, P<0.05), and group comparisons were presented in a volcano plot.

Inhibition of Apoptosis Reduces Diploidization of Haploid Mouse Embryonic Stem Cells during the Differentiation

Project description:Phenotypes of haploid embryonic stem cells (haESCs) are dominant for recessive traits in mice. However, one major obstacle to their use is self-diploidization in daily culture. Although haESCs maintain haploidy well by deleting p53, whether they can sustain haploidy in differentiated status and the mechanism behind remain unknown. To address that, we induced p53-deficient haESCs into multiple differentiated lineages keeping a haploid status in vitro. Besides, haploid cells also remained in chimeric embryos and teratomas arising from p53-null haESCs. Transcriptome analysis revealed that apoptosis genes were down-regulated in p53-null haESCs, comparing to that in wild-type haESCs. Finally, we knocked-out p73, another apoptosis gene, and observed stabilization of haploidy in haESCs, either. These results indicated that the main mechanism of diploidization was apoptosis-related genes triggered cell death in haploid cell cultures. Thus, we can derive haploid somatic cells by manipulating apoptosis gene, facilitating genetic screens of lineage-specific development.

SIRT5 Regulates Lysine Malonylation

Project description:Protein acylation links energetic substrate flux with cellular adaptive responses. SIRT5 is a NAD+-dependent lysine deacylase and removes both succinyl and malonyl group. Using affinity enrichment and label free quantitative proteomics, we characterized the SIRT5-regulated lysine malonylome in wild type (WT) and Sirt5-/- mice. 1137 malonyllysine sites were identified across 430 proteins, with 183 sites (from 120 proteins) significantly increased in the Sirt5-/- animals. Pathway analysis identified glycolysis as the top SIRT5-regulated pathway. Importantly, glycolytic flux was diminished in primary hepatocytes from Sirt5-/- compared to WT mice. Substitution of malonylated lysine residue 184 in glyceraldehyde 3-phosphate dehydrogenase with glutamic acid, a malonyllysine mimic, suppressed its enzymatic activity. Comparison of our previous reports on acylation reveals that malonylation targets a different set of proteins than acetylation and succinylation. These data demonstrate that SIRT5 is a global regulator of lysine malonylation and provide a mechanism for regulation of energetic flux through glycolysis.

Regulation of ILC2s by Brg1 (ATAC-seq on WT and KO ILC2mem)

Project description:The alteration of chromatin status critically dictates the expansion and function of group 2 innate lymphoid cells (ILC2s), which have been shown to play a pathogenic role in allergic lung inflammation. In this study, we use multi-omics approaches to dissect the molecular mechanisms of how Brg1 regulates ILC2s in allergic lung diseases through a cell intrinsic manner. Our data indicate that Brg1 regulates ILC2 properties through orchestrating ILC2 energetic metabolism. These findings are enlightening for the understanding of ILC2-mediated pathogenesis in allergic lung inflammation and highlight Brg1 as a potential therapeutic target.

Development of gene expression signatures for CXXC5 in pDCs

Project description:We have employed whole genome microarray expression profiling as a discovery platform to identify genes regulated by CXXC5 in pDCs. pDCs (CD11c+B220+CD11b-) were differentitated from wild-type and CXXC5-deficient bone marrows by FLT3L for 6 days and sorted by FACS. Expression of five genes (Irf7,Ccl1,Smtn,Dppa2,Ifnab) from this signature was quantified in the same RNA samples by real-time PCR.

Transcriptional profiling of IRF8 KO B cell differentiation

Project description:To understand the gene expression programs regulated by IRF8 in B cells responding to LPS stimulation we generated IRF8 deficient B cells by CRISPR/Cas9. IRF8 knockout and wild-type CD43- splenic B cells were isolated and directly used for RNA-seq (naive B cells) or simulated to differentiate with LPS, IL2, and IL5 for 3 days. At this time activated Bcells and plasmablasts were sorted for RNA-seq.
