Project description:Stroke is one of the major causes of death and long-term disability, of which acute ischemic stroke (AIS) is the most common type. Although circRNA expression profiles of AIS patients have been reported to be significantly altered in blood and peripheral blood mononuclear cells, the role of exosome-contained circRNAs after AIS is still unknown. Plasma exosomes from ten AIS patients and ten controls were isolated, and through microarray and bioinformatics analysis, profile and putative function of circRNAs in the plasma exosomes were studied. A total of 198 circRNAs were differentially quantified (|log2 FoldChange|≥1.00, P<0.05) between AIS patients and controls.The functions of host genes of differentially quantified circRNAs, including RNA and protein process, focal adhesion and leukocyte transendothelial migration, were associated with the development of AIS. As miRNA sponge, differentially quantified circRNAs had the potential to regulate pathways related to AIS, like PI3K-Akt, AMPK and chemokine pathways. Of 198 differentially quantified circRNAs, 96 circRNAs possessing strong translational ability could affect cellular structure and activity, like focal adhesion, tight junction and endocytosis. Most differentially quantified circRNAs were predicted to bind to EIF4A3 and AGO2- two RNA binding proteins (RBPs)- and play a role in AIS. In conclusion, plasma exosome-derived circRNAs were significantly differentially quantified between AIS patients and controls, and participated in the occurrence and progression of AIS by sponging miRNA/RBPs or translating into proteins, indicating that circRNAs from plasma exosomes could be crucial molecules in the pathogenesis of AIS and promising candidates as diagnostic biomarkers and therapeutic targets for the condition.

Project description:To understand the role of exosome miRNAs in AIS disease and to explore the miRNAs and the potential mechanisms related to the occurrence and development of the disease. This research collected serum samples from AIS patients and healthy volunteers and extracted exosome miRNA. The exosome miRNA associated with AIS was screened by illumina high-throughput sequencing . The differentially expressed target genes related to miRNAs were analyzed using bioinformatics analysis including GO functional enrichment analysis, KEGG pathway enrichment analysis, and the screening of key genes via PPI diagram. These differentially expressed mRNAs were indicated an important role in a variety of biological processes and multiple pathways of AIS disease.

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are to compare NGS-derived retinal transcriptome profiling (RNA-seq) to microarray and quantitative reverse transcription polymerase chain reaction (qRT–PCR) methods and to evaluate protocols for optimal high-throughput data analysis Methods: The serum microvesicles of five acute ischemic stroke (AIS) and healthy controls was purified using Ribo™ Exosome Isolation Reagent (C10110-2, RIBOBIO, Guangzhou, China) and analyzed by flow cytometry and nanoparticle tracking analysis (NTA).The miRNA expression profiles of serum microvesicles of five acute ischemic stroke (AIS) and healthy controls were detected by RNA-seq using llumina HiSeqTM 2500. Results: Using an optimized data analysis workflow, 732 miRNA species were detected in total. The levels of 51 individual miRNA species were significantly different between AIS patients and healthy controls. Conclusions: Our study represents the first detailed analysis of miRNA expression profiles of serum microvesicles in AIS and healthy controls, generated by RNA-seq technology. The optimized data analysis workflows reported here should provide a framework for comparative investigations of expression profiles. Our results show that NGS offers a comprehensive and more accurate quantitative and qualitative evaluation of miRNA content in serum microvesicles. We conclude that RNA-seq based non-coding RNA characterization would expedite genetic network analyses and permit the dissection of complex biologic functions.

Project description:Adolescent Idiopathic Scoliosis (AIS) is the most common orthopedic condition requiring surgery, affecting 4% of adolescents. There is currently no proven method or prognostic test to identify symptomatic patients at risk of developing severe scoliosis who could benefit from growth-guided devices or minimally invasive non-fusion instrumentation surgeries. These innovative treatments must be performed at an early disease stage in younger patients to benefit from their growth potential. In this prospective cross-sectional study, we investigated the clinical utility of circulating microRNAs (miRNAs), an important class of small non-coding RNA, as biomarkers to predict the risk of developing severe scoliosis in AIS. Blood samples and clinical data were collected from 116 AIS patients who were followed until skeletal maturity and stratified according to their clinical outcome. Genome-wide expression profiling of miRNAs was performed with plasma obtained at the time of diagnosis of AIS (mean age of 13.3 ± 1.7 years with a mean Cobb angle of 24.4° ± 12.4°). This approach led to the identification of 15 circulating miRNAs that are upregulated in AIS patients who developed a severe scoliosis (Cobb angle ≥45°) at skeletal maturity compared to moderate and mild scoliosis groups (Cobb angle between 25°-44° and <25° respectively). After optimization and the application of Random Forest Models a panel of six miRNAs (miR-1-3p, miR-19a-3p, miR-19b-3p, miR-133b, miR-143-3p, and miR-148b-3p) out of 15 led us to develop an algorithm predicting the risk of developing a severe scoliosis with great accuracy (100%), sensitivity (100%) and specificity (100%). Having a scoliosis predictive bioassay and decision-making tools to predict curve progression in order to find the best treatment plan will undoubtedly transform the orthopedic care system in the field of pediatric scoliosis by integrating innovative precision medicine approaches. In addition, investigation of genes targeted by these miRNAs could fill our gaps in our understanding of AIS pathogenesis and reveal new actionable targets.

Project description:ST-segment elevated myocardial infarction (STEMI) is one of the most severe forms of cardiovascular heart diseases. In the present study, we aim to describe differential expressed plasma exosome-miRNAs in patients with post-STEMI 3-6 months. Methods: Consecutive patients with ages from 40 to 80 years and 25 males among them 3-6 months after STEMI (n=11) were compared to sex-matched healthy control (n=10). The mean age of the patient was 64.71±10.40 years and 89.2% were male. Compared to the healthy control group, the plasma exosome-miRNAs were assessed by using microarray assay (IIIumnia Hiseq 2500). Profile of plasma exosome-miRNAs related to heart diseases was established both in the patient and control groups.
