Project description:ETX treatment promoted cardiomyocyte proliferation. To investigate the molecular mechanism, we performed microarray analysis to explore the molecular mechanism underlying the effect of ETX on cardiomyocyte proliferation. In this study, we sought to determine if and how metabolic reprogramming regulates cardiomyocyte proliferation. In neonatal mice, blockade of glycolysis by inhibiting glucokinase reduced cardiomyocyte proliferation, while blockade of fatty acid oxidation by inhibiting carnitine palmitoyltransferase 1 (CPT1) delayed the cell cycle arrest in cardiomyocytes. ETX (Etomoxir) is an effective inhibitor of CPT1 which is the key enzyme of fatty acid oxidation,ETX induced cardiomyocyte proliferation and improved cardiac function post-MI in adult mice.

Project description:Compensatory angiogenesis is crucial for recovery in patients with diabetic myocardial infarction (DMI), yet effective interventions are still unclear. This study employed liquid chromatography-tandem mass spectrometry to identify lactylated proteins in the infarct border zone of DMI mouse hearts. Our findings revealed that IDH2 is lactylated at lysine 272, enhancing its binding to Cav1 while inhibiting the Cav1-eNOS interaction. This modification promotes eNOS activity and facilitates the proliferation, migration, and angiogenesis of cardiac microvascular endothelial cells under high glucose and hypoxic conditions. In endothelial cell-specific IDH2-K272R knock-in mice, the loss of K272 lactylation impairs cardiac function and exacerbates pathological remodeling due to disrupted angiogenesis.

Project description:Objective: This study aimed to investigate the relationship between sleep disorders and cardiac microvascular injury in myocardial ischemia-reperfusion injury (MI/RI) mice. Design: Mice were placed in a sleep deprivation chamber. The sweep bar moved along the bottom of the cage every 2 min during the light cycle (ZT0-12) and was stationary during the dark cycle (ZT12-24) for 16 weeks. Then, a MI/RI model was prepared to evaluate cardiac microvascular injury and investigate its specific mechanism via proteomics analysis. Results: Here, we showed that 16 weeks of sleep fragmentation (SF) exacerbated cardiac microvascular injury in MI/RI mice. Mechanistically, we found that SF promotes sympathetic overactivity, increases the level of norepinephrine (EPI) in plasma, thereby promotes the chemotaxis of neutrophils (NEs) and formation of Neutrophil extracellular traps (NETs). Furthermore, our data indicated that NETs inhibit Atox1 expression and leads to impaired ATP7A related copper transport and copper overload in cardiac microvascular endothelial cells (CMECs). Copper overload further leads to exacerbated cuproptosis, and these effects can be rescued by excision of the sympathetic nerve, targeted administration of copper chelating agent or NETs inhibitor. Conclusions: This study demonstrated that SF exacerbated MI/RI by promoting copper overload in CMECs. Our study elucidated one of the molecular mechanisms by which sleep disorders aggravate cardiac microvascular injury and suggested possible tar gets for intervention.

Project description:Endothelial cells (EC) were obtained from commercial source (Lonza), expanded and culture until passage 3. HMVEC-LBI correspond to lung microvascular endothelial cells, HPAEC to lung macrovascular endothelial cells, HAEC to cardiac macrovascular endothelial cells and HMVEC-C to cardia microvascular endothelial cells. These cells presented different capacity to generate endothelial microvesicles. The objective of the study was to identify differentially genes between these cells to understand this difference. Total RNA was extracted using the mirVana miRNA Isolation Kit (Ambion), according to the manufacturer’s recommendations. Cy3-CTP labeled RNA was prepared according to standard Agilent protocol from 400ng total RNA. The hybridization was performed for 17 hrs at 65°C. cRNA labeling and hybridization performance were performed and all parameters checked were found within the manufacturers specifications. Arrays were scanned as described in the manufacturers’ protocol. Signal intensities on 20 bit tiff images were calculated by Feature Extraction software (FE, Version 8.5; Agilent Technologies). Data analyses were conducted with GeneSpring GX software (Vers.12.6; Agilent Technologies)

Project description:Glucagon supports glucose homeostasis by stimulating hepatic gluconeogenesis, in part by promoting the uptake and conversion of amino acids into gluconeogenic precursors. Genetic disruption or pharmacologic inhibition of glucagon signaling results in elevated plasma amino acids and compensatory glucagon hypersecretion involving expansion of pancreatic a cell mass. Recent findings indicate that hyperaminoacidemia triggers pancreatic a cell proliferation via an mTOR-dependent pathway. We confirm and extend these findings by demonstrating that glucagon pathway blockade selectively increases expression of the sodium-coupled neutral amino acid transporter Slc38a5 in a subset of highly proliferative a cells and that Slc38a5 controls the pancreatic response to glucagon pathway blockade; most notably, mice deficient in Slc38a5 exhibit markedly decreased a cell hyperplasia to glucagon pathway blockade-induced hyperaminoacidemia. These results show that Slc38a5 is a key component of the feedback circuit between glucagon receptor signaling in the liver and amino-acid-dependent regulation of pancreatic a cell mass in mice.

Project description:Experiments were conducted with mice carrying an inducible EC-specific Hif2α-knockout (ecHif2α–/–) mutation, with mouse cardiac microvascular endothelial cells (CMVECs) isolated from the hearts of ecHif2α–/– mice after the mutation was induced, and with human CMVECs and umbilical-vein endothelial cells transfected with ecHif2α siRNA. After MI induction, echocardiographic assessments of cardiac function were significantly lower, while measures of cardiac microvascular leakage (Evans blue assay), plasma IL6 levels, and cardiac neutrophil accumulation and fibrosis (histology) were significantly greater, in ecHif2α–/– mice than in control mice, and RNA-sequencing analysis of heart tissues from both groups indicated that the expression of genes involved in vascular permeability and collagen synthesis was enriched in ecHif2α–/– hearts. In cultured ECs, ecHif2α deficiency was associated with declines in endothelial barrier function (electrical cell impedance assay) and the reduced abundance of tight-junction proteins, as well as an increase in the expression of inflammatory markers, all of which were largely reversed by the overexpression of ARNT. We also found that ARNT, but not Hif2α, binds directly to the IL6 promoter and suppresses IL6 expression.
