Project description:ETX treatment promoted cardiomyocyte proliferation. To investigate the molecular mechanism, we performed microarray analysis to explore the molecular mechanism underlying the effect of ETX on cardiomyocyte proliferation. In this study, we sought to determine if and how metabolic reprogramming regulates cardiomyocyte proliferation. In neonatal mice, blockade of glycolysis by inhibiting glucokinase reduced cardiomyocyte proliferation, while blockade of fatty acid oxidation by inhibiting carnitine palmitoyltransferase 1 (CPT1) delayed the cell cycle arrest in cardiomyocytes. ETX (Etomoxir) is an effective inhibitor of CPT1 which is the key enzyme of fatty acid oxidation,ETX induced cardiomyocyte proliferation and improved cardiac function post-MI in adult mice.

Project description:Mitochondrial fatty acid oxidation (FAO) is an important energy provider for cardiac work and changes in cardiac substrate preference are associated with different heart diseases. Carnitine palmitoyltransferase 1B (CPT1B) is thought to perform the rate limiting enzyme step in FAO and is inhibited by malonyl-CoA. The role of CPT1B in cardiac metabolism has been addressed by inhibiting or decreasing CPT1B protein or after modulation of tissue malonyl-CoA metabolism. We assessed the role of CPT1B malonyl-CoA sensitivity in cardiac metabolism.

Project description:Microvascular dysfunction has been proposed to drive heart failure with preserved ejection fraction (HFpEF), but the initiating molecular and cellular events are largely unknown. Our objective was to determine when microvascular alterations in HFpEF begin, how they contribute to disease progression, and how pericyte dysfunction plays a role herein. We aimed to assess microvascular dysfunction with respect to the development of cardiac dysfunction, in the Zucker fatty and spontaneously hypertensive (ZSF1) obese rat model of HFpEF at three time points: 6, 14, and 21 weeks of age. We found that pericyte loss was the earliest and strongest microvascular change, occurring before prominent echocardiographic signs of diastolic dysfunction were present. Pericytes were shown to be less proliferative and had a disrupted morphology at 14 weeks in the obese ZSF1 animals, who also exhibited an increased capillary luminal diameter and disrupted endothelial junctions. Pericytes exposed to oxidative stress in vitro showed downregulation of cell cycle associated pathways, and induced a pro-inflammatory state in endothelial cells upon co-culture. We propose pericytes are important for maintaining endothelial cell function, where loss of pericytes enhances the reactivity of endothelial cells to inflammatory signals and promotes microvascular dysfunction, thereby accelerating the development of HFpEF.

Project description:Endothelial cells (EC) were obtained from commercial source (Lonza), expanded and culture until passage 3. HMVEC-LBI correspond to lung microvascular endothelial cells, HPAEC to lung macrovascular endothelial cells, HAEC to cardiac macrovascular endothelial cells and HMVEC-C to cardia microvascular endothelial cells. These cells presented different capacity to generate endothelial microvesicles. The objective of the study was to identify differentially genes between these cells to understand this difference. Total RNA was extracted using the mirVana miRNA Isolation Kit (Ambion), according to the manufacturer’s recommendations. Cy3-CTP labeled RNA was prepared according to standard Agilent protocol from 400ng total RNA. The hybridization was performed for 17 hrs at 65°C. cRNA labeling and hybridization performance were performed and all parameters checked were found within the manufacturers specifications. Arrays were scanned as described in the manufacturers’ protocol. Signal intensities on 20 bit tiff images were calculated by Feature Extraction software (FE, Version 8.5; Agilent Technologies). Data analyses were conducted with GeneSpring GX software (Vers.12.6; Agilent Technologies)

Project description:Cardiac function is dynamically regulated by intercellular signalling. As an important intercellular signalling mediator, this study focused on isolating and defining membrane bound signalling packages termed extracellular vesicles (EVs) directly from heart. Here, we utilised a gentle, enzymatic perfusion to extract EVs from whole cardiac tissues (cEVs) for biophysical and proteomic characterisation; cEVs (100-300 nm) are Cd63+, Tsg101+, Pdcd6ip/Alix+ (EV markers) and contain proteins associated with cardiomyocyte (Actn1, Myh6), cardiac progenitor (Cd34, Abcg2), smooth muscle (Cald1, Tagln), endothelial (Vwf), and fibroblast (Ckap4) origins. Proteomic profiling of cEVs revealed 1721 proteins, which bioinformatics revealed hold an enrichment in metabolic functions of glycolysis, fatty acid oxidation, and antioxidant action, presumably to meet the high energetic demands and balance resulting oxidative stress in cardiac tissue.

Project description:Glucagon supports glucose homeostasis by stimulating hepatic gluconeogenesis, in part by promoting the uptake and conversion of amino acids into gluconeogenic precursors. Genetic disruption or pharmacologic inhibition of glucagon signaling results in elevated plasma amino acids and compensatory glucagon hypersecretion involving expansion of pancreatic a cell mass. Recent findings indicate that hyperaminoacidemia triggers pancreatic a cell proliferation via an mTOR-dependent pathway. We confirm and extend these findings by demonstrating that glucagon pathway blockade selectively increases expression of the sodium-coupled neutral amino acid transporter Slc38a5 in a subset of highly proliferative a cells and that Slc38a5 controls the pancreatic response to glucagon pathway blockade; most notably, mice deficient in Slc38a5 exhibit markedly decreased a cell hyperplasia to glucagon pathway blockade-induced hyperaminoacidemia. These results show that Slc38a5 is a key component of the feedback circuit between glucagon receptor signaling in the liver and amino-acid-dependent regulation of pancreatic a cell mass in mice.

Project description:Experiments were conducted with mice carrying an inducible EC-specific Hif2α-knockout (ecHif2α–/–) mutation, with mouse cardiac microvascular endothelial cells (CMVECs) isolated from the hearts of ecHif2α–/– mice after the mutation was induced, and with human CMVECs and umbilical-vein endothelial cells transfected with ecHif2α siRNA. After MI induction, echocardiographic assessments of cardiac function were significantly lower, while measures of cardiac microvascular leakage (Evans blue assay), plasma IL6 levels, and cardiac neutrophil accumulation and fibrosis (histology) were significantly greater, in ecHif2α–/– mice than in control mice, and RNA-sequencing analysis of heart tissues from both groups indicated that the expression of genes involved in vascular permeability and collagen synthesis was enriched in ecHif2α–/– hearts. In cultured ECs, ecHif2α deficiency was associated with declines in endothelial barrier function (electrical cell impedance assay) and the reduced abundance of tight-junction proteins, as well as an increase in the expression of inflammatory markers, all of which were largely reversed by the overexpression of ARNT. We also found that ARNT, but not Hif2α, binds directly to the IL6 promoter and suppresses IL6 expression.

Project description:Blockade of the glucagon receptor (GCGR) has been shown to improve glycemic control. However, this therapeutic approach also brings side effects, such as α-cell hyperplasia and hyperglucagonemia, and the mechanisms underlying these side effects remain elusive. Here, we conduct single-cell transcriptomic sequencing of islets from male GCGR knockout (GCGR-KO) mice. Our analysis confirms the elevated expression of Gcg in GCGR-KO mice, along with enhanced glucagon secretion at single-cell level. Notably, Vgf (nerve growth factor inducible) is specifically upregulated in α cells of GCGR-KO mice. Inhibition of VGF impairs the formation of glucagon immature secretory granules and compromises glucagon maturation, lead to reduced α-cell hypersecretion of glucagon. We further demonstrate that activation of both mTOR-STAT3 and ERK-CREB pathways, induced by elevated circulation amino acids, is responsible for upregulation of Vgf and Gcg expression following glucagon receptor blockade. Thus, our findings elucidate a previously unappreciated molecular mechanism underlying hyperglucagonemia in GCGR blockade.
