Project description:Iron restriction is a growth limiting condition encountered by M. tuberculosis in the host. In this study we examined the survival and physiology of iron depleted M. tuberculosis. We found that iron depleted M. tuberculosis survive for long time with little or no replication and is able to resume normal growth when iron becomes available. To understand the mechanism used by M. tuberculosis to survive under iron starvation we examine the transcriptional profile of iron-starving in comparison to that of iron-sufficient M. tuberculosis.

Project description:Iron is an essential cofactor for a wide range of cellular processes. Previous studies have shown that siderophore-mediated uptake and intracellular handling of iron are crucial for virulence of Aspergillus fumigatus. Here we show that the bzip-type transcription factor HapX plays a crucial role in the transcriptional remodeling required for adaption to iron starvation in this opportunistic fungal pathogen. HapX was found to be interconnected in a negative feed-back loop with the previously identified iron regulator SreA: SreA repressed expression of hapX during iron sufficiency and HapX repressed sreA during iron starvation. Genome-wide transcriptional profiling and analysis of selected metabolites (protophorphyrine IX, siderophores and amino acids) indicated extensive metabolic remodeling in response to iron starvation. HapX was found to participate in both, repression and activation of genes during iron starvation. HapX was in particular required for repression of iron-dependent and mitochondrial-localized activities including respiration, TCA cycle, amino acid metabolism, iron-sulfur-cluster biosynthesis and heme biosynthesis. Pathways positively affected by HapX included production of siderophores and the ribotoxin and major allergen AspF1. Analysis of the free amino acid pool revealed HapX-dependent coordination of the production of siderophores with the supply of its precursor ornithine. Consistent with the hapX expression pattern, HapX-deficiency was deleterious with respect to growth rate and conidiation during iron depleted but not iron-replete conditions. HapX-deficiency caused significant attenuation of virulence in a murine model aspergillosis underlining that A. fumigatus faces iron starvation in the host and that the HapX-dependent metabolic reprogramming is therefore crucial for virulence.

Project description:C. reinhardtii cells exposed to abiotic stresses (e.g. iron-, nitrogen-, zinc- or phosphorus-deficiency) accumulate TAGs which are stored in lipid droplets. Here, we report that iron starvation leads to formation of lipids bodies and accumulation of TAGs. This occurs between 12 and 24 h of iron-starvation. C. reinhardtii cells deprived for iron have more saturated FA, due to the loss of functional FA desaturases, which are diiron enzymes. The abundance of a plastid ACP-acyl desaturase (FAB2) is significantly decreased to the same degree as observed for ferredoxin, which is a substrate of the desaturases. The increase in saturated FA (C16:0 and C18:0) is concomitant with the decrease in saturated FA (C16:4, C18:3 or C18:4). This pattern was observed for MGDG, DGTS or DGDG. When we monitored the absolute levels of glycerolipids, MGDG content dropped significantly after only 2 h of iron-starvation. On the other hand, DGTS and DGDG contents gradually decrease until a minimum is reached after 24-48 h of iron-deprivation. RNA-Seq analysis of iron-starved C. reinhardtii cells revealed significant changes in many transcripts coding for enzymes involved in FA metabolism. The mRNA abundances of genes coding for components involved in TAG accumulation (DGAT or MLDP) are increased. A more dramatic increase at the transcript level has been observed for many lipases, suggesting that a major remodeling of lipid membranes occurs during iron-starvation in C. reinhardtii. Sampling of Chlamydomonas CC-4532 (2137) cells cultivated photoheterotrophically (TAP) under iron-starvation condition (0 uM Fe-EDTA). Samples were collected from biological duplicates after washing in TAP medium lacking Fe at 0, 0.5, 1, 2, 4, 8, 12, 24 and 48 hours.

Project description:Respiration deficient S. aureus small colony variants (SCVs) frequently cause persistent infections, which necessitates they acquire iron, yet how SCVs obtain iron remains unknown. To address this, we created a stable hemB mutant in S. aureus USA300 strain LAC. The hemB SCV utilized exogenously supplied heme but was attenuated for growth under conditions of iron starvation. RNA-seq showed that both WT S. aureus and the hemB mutant sense and respond to iron starvation.

Project description:The gene PA2384 in Pseudomonas aeruginosa PAO1, annotated originally as unknown function, has been previously shown to be dramatically responsive to iron limitation. In the present study, PA2384 is shown by bioinformatics analysis to have a weak similarity to the N-termini DNA binding domain of fur, a well-known ferric uptake regulator. To experimentally investigate the function of PA2384 P. aeruginosa PAO1 recombinant (pUCP20::PA2384) overexpressing PA2384 and PA2384 knock-out mutant PAO1-23844 are constructed and studied. Physiological characterization in a carefully controlled cultivation system shows that the knockout strain needed a longer lag phase to grow and exhibited a significantly reduced production speed of siderophore (pyoverdine and pyochelin) in the stationary phase. Genome-scale transcriptional profiles at different growth stages are compared between the wild type and ∆PA2384 mutant grown under iron-limiting conditions. The expression of more than 350 genes is affected. Among them, seventy-one genes involved in iron uptake are significantly induced by PA2384. 102 quorum sensing (QS) dependent genes exhibited differential transcription. They include genes related to the biosynthesis of some important virulence factors such as pyocyanin, rhamnolipids and hydrogen cyanide. Transcription of genes responsible for the synthesis of Pseudomonas Quinolone Signal (PQS) is greatly advanced by the knockout of PA2384. We postulate that PA2384 affects QS via PQS. Furthermore, the knockout of PA2384 also results in altered expression of genes involved in electron transfer, central metabolism, phosphorus starvation and translation. It implies that PA2384 may affect more basic physiology than only respond to iron uptake and is a versatile global regulator in P. aeruginosa under iron starvation. A. fumigatus 293 and hapX mutants were grown in the presence and absence of iron and in cultures shifted from no iron to iron-containing conditions after 1 h incubation. Hybridizations were performed with biological replicates for wt vs hapX +/- iron. For the iron-shift experiments, there were biological replicates for wt in both conditions and for hapX in -iron but there was only a single biological sample for hapX iron-shift sample. All hybs were performed with flip-dye pairs.
