As a primary goal, adaptation to cold climate could expand an invasion range of exotic plants. Here, we aimed to explore the regulation strategy of M. micrantha seed development under cold stress through molecular physiology and multi-omics analysis. Re-establishment of desiccation tolerance in protruded radicles of Medicago truncatula Wt and abi5 seeds

Project description:Desiccation tolerance (DT) is the capacity to withstand total loss of cellular water. This property is acquired during seed filling and lost just after germination. However, in many species, a germinated seed can regain DT under adverse conditions such as osmotic stress. We discovered that the germinated seeds of the abscisic acid insensitive 5 (Mtabi5) mutant of Medicago truncatula lost their ability to re-establish DT during osmotic stress. To characterize the molecular processes that are influenced by MtABI5 during the re-establishment of DT tolerance, a transcriptome analysis was performed on the protruded radicles of germinated Mtabi5 mutants and wilt type before and after an osmotic treatment.

Shotgun proteomic analysis of embryonic axis of soybean seeds during germination under salt stress

Project description:Salinity is one of the main environmental stresses worldwide limiting soybean growth and yield. Seed imbibition and radical emergence are generally less affected by salinity in soybean. Towards unraveling the mechanisms underlying salt tolerance in soybean at germination stage, a comprehensive quantitative proteomic analysis of proteins from soybean embryonic axis during germination sensu stricto (GSS) under saline conditions was performed. Application of 100 and 200 mmol L-1 NaCl at GSS was significantly accompanied by the change in abundance (>2-fold) of 97 and 75 proteins, respectively. Most of these proteins were involved in three major functions, namely stress response and defense, protein turnover and protection and primary metabolism. Our results pave the way towards the identification of suitable biomarkers useful for improving salt tolerance in soybean.

Deep sequencing-based characterization of transcriptome and transcriptional profiling of trifoliate orange (Poncirus trifoliata (L.) Raf.) in response to cold stress

Project description:A large number of high-quality transcripts were fistly obtained by sequencing the pooled RNAs from trifoliate orange under different stress treatments (cold, salt, drought). Four cold-treated libraries were further mapped to the assembled transcriptome, leading to identification of more 5,000 DEGs. Plant hormone signal transduction, plant-pathogen interaction, and secondary metabolism were most significantly enriched in the DEGs. A total of 60 transcription factors were revealed to be cold responsive.n addition, a number of genes involved in catabolism and signaling of hormones, such as abscisic acid, ethylene and gibberellin, were either up- or down-regulated under cold stress. These data provide valuable information for elucidating transcriptomic reprogramming and underlying mechanisms of trifoliate orange in response to cold and underpin exploitation of genes of interest in the future.

Transcriptome and physiological analysis of rapeseed tolerance to post-flowering temperature increase

Project description:This study investigates the transcriptome and physiological responses of rapeseed to post-flowering temperature increases, providing valuable insights into the molecular mechanisms underlying rapeseed tolerance to heat stress. Two rapeseed genotypes, Lumen and Solar, were assessed under control and heat stress conditions in field experiments conducted in Valdivia, Chile. Results showed that seed yield and seed number were negatively affected by heat stress, with genotype-specific responses. Lumen exhibited a 9.3% average seed yield reduction, while Solar showed a 28.7% reduction. RNA-seq analysis of siliques and seeds revealed tissue-specific responses to heat stress, with siliques being more sensitive to temperature stress. Hierarchical clustering analysis identified distinct gene clusters reflecting different aspects of heat stress adaptation in siliques, with a role for protein folding in maintaining silique development and seed quality under high temperature conditions. In seeds, three distinct patterns of heat-responsive gene expression were observed, with genes involved in protein folding and response to heat showing genotype-specific expression. Gene coexpression network analysis revealed major modules for rapeseed yield and quality, as well as the trade-off between seed number and seed weight. Overall, this study contributes to understanding the molecular mechanisms underlying rapeseed tolerance to heat stress and can inform crop improvement strategies targeting yield optimization under changing environmental conditions.

Proteomic analysis of Glycine max seeds during controlled deterioration treatment

Project description:Controlled deterioration treatment (CDT) negatively affects the seed quality and vigor during post-harvest storage. A label-free proteomic approach was utilized to understand the CDT responses in soybean seeds. Soybean seed are rich in seed-storage proteins (SSPs) constituting up to 70 to 80% of the total seed protein content. Due to the presence of these SSPs, it is very difficult to identify and/or characterize the low-abundance regulatory proteins. Availability of appropriate methods for extraction of low-abundance proteins (LAPs) are now providing a platform for the identification of novel proteins involved in the signal perception and transduction during environmental perturbations. To enrich LAPs, the extracted total seed proteins were subjected to protamine sulfate precipitation (PSP) method to deplete SSPs. Fractionated protein fractions thus prepared were analyzed for identification of differential proteins using Label-free quantitative proteomics approaches.
