Project description:Previous studies have demonstrated the requirement of autophagy for neuronal survival and synaptic function, mostly focusing on projection neurons and excitatory neurotransmission. Here, we examine the effects of conditional atg5 ablation in neurons expressing parvalbumin (PV-cKO), which in the forebrain mainly represent fast-spiking inhibitory interneurons. Contrary to the prevailing view, we show that autophagy-deficient PV neurons survive throughout the brain, with the exception of the projection Purkinje neurons in the cerebellum, which rapidly degenerate. While not affecting their maintenance, autophagy deficiency leads to aberrant proteostasis of key synaptic and other proteins, culminating to reduced inhibitory neurotransmission in hippocampal PV-interneurons. Consistently, PV-cKO animals exhibit associative and recognition memory deficits. Our findings demonstrate a neuronal type-specific vulnerability to autophagy deficiency, while also identifying PV-interneurons as the cellular substrates where autophagy is required for memory formation.

Project description:Apoptosis and autophagy are two fundamental pathophysiological processes of cell fate regulation under stress conditions, especially in anti-cancer therapy. However, the interplay between apoptosis and autophagy in the cancer chemoresistance is not fully elucidated. Herein, we identified apoptosis-related protein in TGF-β signaling pathway (ARTS) as a key link between apoptosis and autophagy, facilitating cell survival and chemoresistance. ARTS was highly expressed in chemoresistant breast cancer tissues and was associated with patient poor prognosis. Expression of ARTS conferred the resistance of doxorubicin (DOX) and docetaxel (DTX) by inducing autophagy in breast cancer cells in vivo and in vitro. As regards the mechanism of action, ARTS translocated from the mitochondrial intermembrane space to the cytoplasm after pro-apoptotic signals from chemotherapeutic drugs, subsequently inducing autophagy through the SIAH1-mediated Livin degradation and further activation of the MDM2/p53 pathway, thus resulting in the survival of breast cancer cells. The inhibition of apoptosis or autophagy effectively suppressed the chemoresistance mediated by ARTS. Generally, this study identified a new pathway of ARTS/Livin/MDM2/p53 conferred chemoresistance of breast cancer through inducing autophagy in an apoptosis-dependent manner.

Project description:RUBCN/Rubicon was originally identified as a negative regulator of canonical autophagy. However, its function remains controversial. Here we show that RUBCN is expressed as two isoforms of different sizes, RUBCN130 and RUBCN100, that are translated from alternative translation initiation sites. Selective RUBCN130 deficiency in B cells enhances autophagy and memory B-cell generation. Autophagy suppression by ATG7 deficiency reverses the enhancement of memory B-cell generation in B-cell–specific RUBCN130-deficient mice. While RUBCN130 is intracellularly localized in late endosomes and lysosomes, RUBCN100 is preferentially located in early endosomes and enhances the enzymatic activity of class III PI(3)K VPS34. Additionally, RUBCN100 plays a positive regulatory role in autophagy since its expression promotes autophagy and suppresses mTORC1 activation. Our findings reveal the critical role of functional interaction between RUBCN isoforms in autophagy regulation and also provide clues for developing a new vaccine strategy to induce efficient B-cell memory.

Project description:In nature, plants are constantly exposed to many transient, but recurring, stresses. Thus, to complete their life cycles they require a dynamic balance between capacities to recover following cessation of stress and maintenance of stress memory. Recently, we uncovered a new functional role of autophagy in regulating recovery from heat stress (HS) and resetting cellular memory of HS in Arabidopsis thaliana. Here, we demonstrate that NBR1 (Next-to-BRCA1) plays a crucial role as an adaptor receptor for selective autophagy during recovery from HS. Immunoblot analysis and confocal microscopy revealed that levels of NBR1 protein, NBR1-labeled puncta and NBR1 activities were all higher during the HS recovery phase than before and after this phase. Co-immunoprecipitation analysis of proteins interacting with NBR1 and comparative proteomic analysis of a nbr1 knockout mutant and wild-type plants identified 58 proteins as potential novel targets of NBR1. Cellular, biochemical and functional genetic studies confirmed that NBR1 targets Heat Shock Protein 90 (HSP90) and ROF1, a member of the FKBP family, and mediates their degradation by autophagy, which represses the expression of HSPs regulated by HsfA2 transcription factor and the response to HS. Accordingly, loss-of-function mutation of NBR1 resulted in a stronger HS memory phenotype. Together our results provide new insights into the mechanistic principles by which autophagy regulates plant response to recurrent HS.

Project description:Memory B cells are long-lived lymphocytes essential for humoral immunological memory. Autophagy is required for the long-term survival of memory B cells, however, the molecular mechanisms for the protection by autophagy have not been fully elucidated. Here we show that mitochondrial autophagy mediated by Nix and Bnip3 prevents necroptosis in memory B cells by maintaining mitochondrial homeostasis and normal lipid metabolism. In mice with B cell-specific deletion of Nix and Bnip3, memory B cells showed significant increases in viable mitochondria and de novo fatty acid synthesis, with accumulation of lipid droplets in the cytoplasm and accelerated cell death. Inhibiting the mitochondrial citrate transporter Slc25a1 or silencing the necroptosis gene Ripk3 rescued Nix–/–Bnip3–/– memory B cells in vivo. These data indicate that Nix- and Bnip3-dependent mitochondrial autophagy in memory B cells is important for restricting de novo fatty acid synthesis and preventing necroptosis. Our results suggest a mechanism for autophagy in the maintenance of mitochondrial homeostasis for the protection of immunological memory.

Project description:Endometriosis (EMs) is a common infertility-related disease in women of reproductive age. Impaired endometrial decidualization is one of the most important factors contributing to the embryo implantation failure EMs patients. However, the exact mechanism remains unclear. Previous studies have shown collagen I deposition in the eutopic endometrium of EMs patients, which may lead to impaired endometrial decidualization. The level of collagen I in eutopic endometrium of EMs was analyzed. We performed a proteomic analysis of ectopic endometrial stromal cell-derived extracellular vesicles (EMs-EVs) and extracellular vesicles derived from endometrial stromal cells in the endometrium of control patients (CTL-EVs). An endometrial transcriptional profiles of EMs patients and normal controls in the mid-secretory phase of menstrual cycle was compared. Endometrial stromal cells (ESCs) were stimulated with chloroquine or rapamycin to evaluate the association between autophagy and collagen I. The expression of PKM2 in EMs-EVs and serum extracellular vesicles from EMs patients were examined by western blotting. PKM2 was overexpressed or knockdown in ESCs to investigate the level of autophagy and collagen I. ESCs were treated with ectopic ESC-derived extracellular vesicles with highly expressed PKM2 protein, and the potential molecular mechanisms were further confirmed through western blotting and immunohistochemical analysis. We found that endometrial collagen I expression during the mid-secretory phase was increased in the EMs group. We demonstrated that autophagy is defective in eutopic endometrial stromal cells (ESCs) of EMs patients. In ESCs, pharmacological inhibition of autophagy by chloroquine (CQ) promoted collagen I deposition. Ectopic endometrial stromal cell-derived extracellular vesicles (EMs-EVs) inhibited autophagy of ESCs and promoted collagen I deposition in vivo and in vitro. Mechanistically, EMs-EVs encapsulating PKM2 impair eutopic endometrial autophagy via Akt/mTOR signaling pathway. Together, we demonstrated that EMs-EVs encapsulating PKM2 impaired endometrial autophagy inducing collagen I deposition in EMs, which provided a potential target for therapeutic implications.
