Masson pine pine wood nematode inoculation proteome LC-MSMS

ABSTRACT: Using pine wood nematode resistant Pinus massoniana clones as materials, after inoculation with pine wood nematode, needles from 5 locations of the same plant were collected and mixed as a biological replicate at 0d,3d and 10d, and a total of two biological replicates were performed for proteomics analysis based on TMT tags.

A Comprehensive Transcriptional Profiling of Pepper Responses to Root Knot Nematode

Project description:We sequenced the transcriptomes of resistant and susceptible cultivar roots uninoculated and with RKN M. incognita, over three time points; 1-, 4-, and 7-days post inoculation. Over 300 million high quality reads from 36 samples were mapped to the pepper reference genome. Comparing inoculation and resistance profiles revealed generic biotic stress responses and specific nematode resistance responses. Comparisons along the time course revealed resistance responses are often induced early in the infection process and a large portion of these are restricted to chromosome P9, encoding the known nematode resistance loci

Proteomic profiling of rice (Oryza sativa Nipponbare) roots during compatible interaction with the root-knot nematode Meloidogyne graminicola

Project description:This study employed label-free quantitative mass spectrometry to investigate the proteomic changes in roots of the susceptible rice cultivar Nipponbare during a compatible interaction with the root-knot nematode Meloidogyne graminicola. Root samples were collected at four time points: before inoculation (0 dpi), and at 1, 3, and 7 days post-inoculation (dpi). For time points where galls were visible (3 and 7 dpi), root sections containing galls were specifically collected. Three biological replicates were analyzed for each condition. In total, 6,072 proteins were identified. Comparative analysis between infected and uninfected roots identified 513 proteins uniquely regulated in response to nematode infection. Bioinformatic analyses (GO, KEGG) of these proteins highlighted significant enrichment in defense-related pathways, including phenylpropanoid biosynthesis, glutathione metabolism, and alpha-linolenic acid metabolism. Key downregulated defense-related proteins were further validated, and their corresponding gene expression was analyzed in a resistant rice accession.

Transcriptional profiling of root-knot nematode induced feeding sites in cowpea (Vigna unguiculata L. Walp)

Project description:In this study we have looked at the transcriptome profile of both incompatible and compatible cowpea-RKN interaction for two different time points using the Affymetrix soybean GeneChip. This is the first study of this kind in cowpea-RKN interaction. This study provides a broad insight into the Rk-mediated resistance in cowpea and creates an excellent dataset of potential candidate genes involved in both nematode resistance and parasitism, which can be further tested for their role in this biological process using functional genomics approaches. our results have shown that the root-knot nematode resistant pathway is still partially suppressed at 9 days post inoculation in resistant cowpea root. There is indication that subtle variation of ROS concentration, induction of toxins and other defense related genes play a role in this unique resistance mechanism. Further functional analysis of these differentially expressed genes will help us to understand this intriguing plant-nematode interaction in a more precise manner.

Characterization of Heterorhabditis bacteriophora Response to Insect-Derived and Non-Biological Stimuli: Insights into Nematode Recovery and Released Proteins

Project description:Entomopathogenic nematodes (EPNs) are gaining significant popularity as biocontrol agents due to their ability to rapidly kill insect hosts. These nematodes produce bioactive molecules known as excreted/secreted products (ESPs) that help them overcome host immune defenses. Understanding the mechanisms that trigger ESP release and their composition is crucial for unraveling the factors underlying nematode pathogenicity and enhancing their effectiveness as biocontrol agents. Leveraging the insect parasitic nematode, Heterorhabditis bacteriophora, we describe an in vitro study focused on the recovery process of infective juveniles upon contact with selected activation materials. We evaluated three host-derived materials from Galleria mellonella larvae: live larvae, frozen larvae, and heat-inactivated homogenates; and two non-insect-derived materials: filtered water and phosphate-buffered saline. While none of the tested materials resulted in 100% IJ recovery, all induced ESP release. The ESPs were collected and analyzed using liquid chromatography-tandem mass spectrometry, resulting in the identification of 372 proteins. The highest number of proteins, approximately 200, were detected in response to heat-inactivated homogenate and phosphate-buffered saline treatment. Among the identified ESPs were proteins involved in immunomodulation and protease inhibition, both crucial for overcoming host defenses. Our findings highlight the complexity of nematode-host interactions and suggest avenues for improving biocontrol strategies through a deeper understanding of nematode activation and the released spectrum of ESPs.

Potato cyst nematode repsonses in potato

Project description:Sets of seven 2-week old potato plants carrying the nematode resistance gene H1, grown from tuber ‘chits’ in sandy loam at a constant temperature of 20 ºC and a light cycle of 16 hour light/8 hour dark, were each inoculated in the roots evenly with 2000 juveniles of the virulent potato cyst nematode Globodera pallida, or with the avirulent G. rostochiensis pathotype Ro1, or with water. Plants were manually watered throughout the duration of the experiment. 5, 17 and 33 days after inoculation, the roots were carefully washed and root tissue samples were individually flash-frozen in liquid nitrogen and stored at –80 ºC. Total RNA isolations were performed using the Qiagen RNeasy kit. All samples were treated with DNase. The experiment was replicated twice. Keywords: Direct comparison

Dissecting the genetic basis of an evolved acute heat stress and oxidative stress resistance in the nematode Caenorhabditis remanei reveals a complex and unique genetic architecture

Project description:Analysis of constitutive changes in gene expression patterns in populations of the nematode Caenorhabditis remanei experimentally evolved to either acute heat stress, acute oxidative stress or the lab environment. Results of this study demonstrate that acute heat stress and acute oxidative stress are complex traits with a unique genetic basis. Overall design: Pooled population RNA sequencing libraries were obtained from the ancestor population and three evolved populations: one biological replicate of the control selected population, one heat stress selected population and one oxidative stress selected population per replicate. Tissue was collected from populations of L1 larval worms.
