Abstract:
Clinical investigations prove that blue light irradiation
reduces the severity of psoriasis vulgaris. Nevertheless, the
mechanisms involved in the management of this condition remain
poorly defined. Despite the encouraging results of the clinical
studies, no clear guidelines are specified in the literature
for the irradiation scheme regime of blue light-based therapy
for psoriasis. We investigated the underlying mechanism of blue
light irradiation of psoriatic skin, and tested the hypothesis
that regulation of proliferation is a key process. We
implemented a mechanistic model of cellular epidermal dynamics
to analyze whether a temporary decrease of keratinocytes
hyper-proliferation can explain the outcome of phototherapy
with blue light. Our results suggest that the main effect of
blue light on keratinocytes impacts the proliferative cells.
They show that the decrease in the keratinocytes proliferative
capacity is sufficient to induce a transient decrease in the
severity of psoriasis. To study the impact of the therapeutic
regime on the efficacy of psoriasis treatment, we performed
simulations for different combinations of the treatment
parameters, i.e., length of treatment, fluence (also referred
to as dose), and intensity. These simulations indicate that
high efficacy is achieved by regimes with long duration and
high fluence levels, regardless of the chosen intensity. Our
modeling approach constitutes a framework for testing diverse
hypotheses on the underlying mechanism of blue light-based
phototherapy, and for designing effective strategies for the
treatment of psoriasis.

Project description:We investigated light dependent gene expression changes in the marine ochrophyte Nannochloropsis oceanica CCMP1779. These algae have several putative blue light photoreceptors but appear to lack red light photoreceptors. To study early light signaling in N. oceanica and avoid as much as possible secondary downstream events, we quantified gene expression changes in dark-adapted cells after a short blue or red light pulse. More genes were differentially expressed under blue than under red light. In addition, fold change in expression was smaller for the red light-treated samples. For example, the median fold change of induced genes was 3 for blue light and 2.5 for red light. Moreover, hierarchical cluster analysis showed that gene expression after red light treatment was more similar to the dark control than after blue light treatment.

Project description:Aureochromes represent a unique type of blue-light photoreceptors that possess a blue-light sensing flavin-binding LOV-domain and a DNA-binding bZIP domain. Therefore, in contrast to other photoreceptors, aureochromes are considered as light-driven transcription factors. As a member of the essential group of marine primary producers, the pennate diatom Phaeodactylum tricornutum possesses four aureochromes (PtAUREO1a, 1b, 1c, 2). We here show a dramatic change in the global gene expression pattern of P. tricornutum cells after a shift from red to blue light. About 75% of the genes show significantly changed transcript levels already after 10 and 60 min of blue light exposure, which includes genes of major transcription factors as well as other photoreceptors. Very surprisingly, in two independent PtAureo1a knockout lines, this light induced regulation of gene expression is almost completely abolished. Such a massive and fast transcriptional change depending on only one single photoreceptor is so far unprecedented. We hence conclude that PtAUREO1a plays a key role in light regulation.

Project description:The experiment investigates the consequences of Yjr141w downregulation on gene expression in yeast. The essential protein Yjr141w was depleted from cells using a tagged variant (Yjr141w-psd). The photo-sensitive degron (psd) module initiates degradation of the tag and its associated protein upon blue-light exposure. Altogether, we compared samples of the control yeast strain (ESM356-1) grown in darkness (3 arrays) and grown exposed to blue-light (wavelength 465 nm; light-flux 30 µmol m-2 s-1) with the Yjr141w-psd strain (YAL81) grown in darkness and grown exposed to blue-light.

Project description:In this study, we investigated the effects of light-induced fetal responses in avian species, specifically focusing on green monochromatic illumination (GMI). Broiler eggs were exposed to GMI either throughout the entire incubation period (Green) or specifically during the final three days before hatching (G3D), with control groups incubated in dark or white light conditions. Unbiased genome-wide analyses at day of hatch (DOH) revealed significant transcriptional changes in the hypothalamus of the G3D group, highlighting over 500 differentially expressed genes. In parallel with these transcriptional changes, our epigenetic analysis revealed that GMI exposure in the G3D group resulted in increased binding of phosphorylated CREB1 (pCREB1) and elevated levels of Histone H3 Lysine 27 acetylation (H3K27ac) at gene promoters linked to growth, appetite, and metabolism. Mechanistically, we found that pre-exposure to blue light, which disrupts green photoreceptor activity, reduced retinal green opsin levels and nullified the epigenetic modifications typically induced by GMI in the G3D group. Phenotypically, the G3D group exhibited increased post-hatch body weight at several stages, suggesting enhanced growth plasticity and metabolic efficiency, though these effects diminished with age. This study underscores the significant role of specific wavelengths and timing of light exposure in shaping developmental trajectories, and highlight the crucial role of epigenetics in shaping organismal responses to their environment.
