Proteome analysis reveals novel serum biomarkers for Henoch-Schönlein purpura in Chinese children


ABSTRACT: We performed proteomic profiling on serum samples from HSP patients and healthy children using an approach combining magnetic bead-based weak cation exchange and matrix-assisted laser desorption ionization-time of flight mass spectrometry. We next performed liquid chromatography-electrospray ionization-tandem mass spectrometry to identify the proteins and selected potential biomarker based on bioinformatics analysis of the STRING and GO dataset. 7 peptides showing the most significant changes in abundance across HSP patients and healthy children patients, were identified as peptide regions of ALB C4A TUBB FGA and EZR. Validation studies and statistical analysis showed that high serum C4A, ALB and low serum EZR levels, high serum C4A and IgA levels, and high serum D-dimer levels as independent risk factors for HSP, HSP nephritis (HSPN), and abdominal HSP, respectively
