Project description:Activation of the systemic and myocardial rennin-angiotensin-aldosterone system (RAAS) by hyperglycemia plays a critical role in the development of diabetic cardiomyopathy. To test the hypothesis that AngIV protects against diabetic cardiomyopathy via stimulation of AT4R and inhibition of overactive autophagy, diabetic mice were treated with low-, medium- and high-dose AngIV, AT4R antagonist divalinal, forkhead box protein O1 (FoxO1) inhibitor AS1842856 (AS) or their combinations. In vitro, cardiomyocytes were treated with different concentrations of glucose, low-, medium- and high-dose AngIV, divalinal, FoxO1-overexpression plasmid (FoxO1-OE), AS or their combinations. The results showed that AngIV treatment dose-dependently attenuated left ventricular dysfunction, remodeling, fibrosis, and myocyte apoptosis in diabetic mice. Besides, autophagy and FoxO1 protein expression enhanced by diabetes were dose-dependently suppressed by AngIV treatment. However, these cardioprotective effects of AngIV were completely abolished by divalinal administration. Bioinformatic analyses revealed that the differentially expressed genes were enriched in autophagy, apoptosis, and FoxO signaling pathways among control, diabetes, and diabetes+high-dose AngIV groups. Similar to AngIV, AS treatment ameliorated diabetic cardiomyopathy in mice. In vitro, AngIV inhibited collagen expression, apoptosis, overactive autophagy flux, and FoxO1 nuclear translocation induced by high glucose in cardiomyocytes. However, these protective effects of AngIV were completely blocked by the use of divalinal or FoxO1-OE, and these detrimental effects were reversed by the additional administration of AS. In summary, AngIV treatment dose-dependently attenuated left ventricular dysfunction and remodeling in a mouse model of diabetic cardiomyopathy, and the mechanism involved stimulation of AT4R, suppression of FoxO1 nuclear translocation and inhibition of FoxO1-mediated overactive autophagy.

The aim of this study was to examine the role of the leukocyte-derived enzyme Myeloperoxidase (MPO) in pathogenesis of anthracycline-induced Cardiomyopathy (AICM). We performed proteomics on whole heart tissue of MPO-deficient mice on C57BL/6J background and wildtype (WT) littermates seven days after intravenous injection of the anthracycline Doxorubicin (20 mg/kg bodyweight, dissolved in saline) or saline (NaCl).

Project description:Mesembryanthemum crystallinum (common ice plant) is one of the facultative halophyte plants, and it serves as a model for investigating the molecular mechanisms underlying its salt stress response and tolerance. Here we cloned one of homeobox transcription factor (TF) gene McHB7 from ice plant, which has 60% similarity with the Arabidopsis AtHB7. Overexpression of McHB7 in Arabidopsis (OE) showed that the plants had significantly elevated relative water content (RWC), chlorophyll content, superoxide dismutase (SOD) and peroxidase (POD) activities after salt stress treatment. Proteomics analysis identified 145 to be significantly changed in abundance, and 66 were exclusively increased in the OE plants compared to wild type (WT). After salt treatment, 979 and 959 metabolites were significantly increased and decreased in OE plants compared to the WT, respectively. The results demonstrated McHB7 can improve photosynthesis and increase the leaf chlorophyll content, and affect TCA cycle by regulating metabolites (e.g., pyruvate) and proteins (e.g., citrate synthase). Also, McHB7 modulates the expression of stress-related proteins (e.g., superoxide dismutase, dehydroascorbate reductase and pyrroline-5-carboxylate synthase B) to scavenge reactive oxygen species and enhance plant salt tolerance.

Project description:The aim of the experiment was to analyze key molecular changes using an unbiased, transcript-based approach in an experimental obesity model. Small expression RNA sequencing analysis was used to reveal expression changes. Male Long-Evans rats were given a control (CON) or high fat (20%) high sucrose (15%) diet (HFS) for 25 weeks. From week 16, the animals were injected. 0.25 mg · kg - 1 daily with selegiline (CON + S and HFS + S) or vehicle (CON, HFS). Left ventricular samples from all four groups (n = 4-5) were analyzed.

Project description:In this study, we have created a mouse model of methamphetamine cardiomyopathy that reproduces the chronic, progressive dosing commonly encountered in addicted subjects. We gradually increased the quantity of methamphetamine given to C57Bl/6 mice from 5 to 40 mg/kg over five months. At the fifth month, heart weight was increased, echocardiograms showed a dilated cardiomyopathy and survival was lower in males, with less effect in females. Interestingly, these findings correspond to previous observations in human patients, suggesting greater male susceptibility to the effects of methamphetamine on the heart.Transcriptional analysis showed changes in genes dysregulated in previous methamphetamine neurological studies as well as many that likely play a role in cardiac response to this toxic stress. We expect that a deeper understanding of the molecular biology of methamphetamine exposure in the heart will provide insights into the mechanism of cardiomyopathy in addicts and potential routes to more effective treatment.

Project description:The broilers were randomly allotted to four treatment groups (Con, DEX, P8, and DEX+P8 groups) with 10 replicates per group (10 broilers per replicate). Broilers in the Con and DEX groups were fed a basal diet. Broilers in the P8 and DEX+P8 groups were fed a basal diet containing 1 × 108 CFU/g P8. At 16 days of age, broilers in the DEX and DEX+P8 groups were injected with 3 mg/kg body weight DEX (200 μL), whereas broilers in the Con group were injected with an equal volume of saline.
