Project description:One of the major disfunctions that occurs in heat-stressed plants is enhanced protein damage and a consequent decline in cellular protein content. Creeping bentgrass (Agrositis stolonifera L.) is an economically important perennial grass species which is largely used on high value turf areas but experiences frequent damage due to heat stress. Several promising experimental lines of creeping bentgrass showed various tolerance levels to heat stress, with differential responses observed in physiological traits, total protein content and rates of protein degradation. The ubiquitin-proteasome system (UPS) plays a crucial role in the removal of damaged proteins, and there is a critical need to better understand the changes in protein accumulations and degradation via the UPS that occur during heat stress. Hence, we aimed to estimate change in global protein accumulations by performing gel-free proteomics as well as identify proteins that have been polyubiquitinated and targeted to the UPS pathway via polyubiquitin-omics in contrasting creeping bentgrass lines exposed to heat stress. It was found that heat-tolerant S11 729-10 was able to maintain less severe downregulation of proteins involved in the light reactions of photosynthesis, while enhancing the upregulation of antioxidant proteins, particularly during the later phase of stress. These contributed to its improved physiological performance including greater cell membrane integrity as well as healthier light harvesting components. Additionally, the faster turnover of key polyubiquitinated antioxidant proteins in S11 729-10 likely represents a critical mechanism for protecting against oxidative damage and enhancing tolerance under prolonged heat stress. This is the first time that the application of polyubiquitin-omics has been utilized in turfgrass. These findings provide deeper insights into protein metabolism underlying heat tolerance. Key stress-related traits or proteins identified in this study could be utilized to help develop new cultivars with enhanced tolerance to heat stress.

Project description:One of the eminent opportunities afforded by modern genomic technologies is the potential to provide a mechanistic understanding of the processes by which genetic change translates to phenotypic variation and the resultant appearance of distinct physiological traits. Indeed much progress has been made in this area, particularly in biomedicine where functional genomic information can be used to determine the physiological state (e.g., diagnosis) and predict phenotypic outcome (e.g., patient survival). Until now, ecology has lacked an analogous approach where genomic information can be used to diagnose the presence of a given physiological state (e.g., stress response) and then predict likely phenotypic outcomes (e.g., stress tolerance, fitness). We demonstrate that a compendium of genomic signatures can be used to classify the plant abiotic stress phenotype in Arabidopsis according to the architecture of the transcriptome, and then be linked with gene coexpression network analysis to determine the underlying signaling pathways and ultimately the genes governing the phenotypic response. Here, we release microarray data from an expression profiling study where plants were exposed to heat and drought alone, and in combination with each other. A direct loop design with 6 biological replicates for control, heat, drought, and combined heat and drought was performed. A schematic describing the design is provide as supplementary information.

Project description:To better understand the mechanisms that regulate the heat stress response in rice, we conducted a comparative analysis of transcriptome profile in panicles from two rice lines, heat-tolerant line 252 (HTL252) and heat-susceptible line 082 (HSL082) using rice Affymetrix GeneChip. In HTL252 panicles, 1538 differentially expressed genes (DEGs) genes with at least four-fold expression changes compared with the control under heat treatment were considered heat-responsive (HR). Of these DEGs, 522 genes were up-regulated while 1016 genes were repressed. Among DEGs in HSL082, 496 genes were induced and 1707 genes were repressed. Out of the 370 common DEGs found between HTL252 and HSL082, 129 genes were induced and 241 genes were repressed.

Project description:Heat shock response (HSR) is a cellular defense mechanism against various stresses. Both heat shock and proteasome inhibitor MG132 cause the induction of heat shock proteins, a distinct feature of HSR. To better understand the molecular basis of HSR, we subjected the mouse fibrosarcoma cell line, RIF-1, and its thermotolerant variant, TR-RIF-1 cells, to heat shock and MG132. We compared mRNA expressions using microarray analysis during recovery after heat shock and MG132 treatment. This study led us to group the 3,245 up-regulated genes by heat shock and MG132 into three families: genes regulated 1) by both heat shock and MG132 (e.g. chaperones); 2) by heat shock (e.g. DNA-binding proteins including histones); and 3) by MG132 (e.g. innate immunity and defense-related molecules).

Project description:Effects of heat priming applied to the first generation on tolerance of the successive generation to post-anthesis high temperature stress were investigated. Compared with the progeny of non-heat primed plants (NH), the progeny of heat-primed plants (PH) presented higher grain yield, leaf photosynthesis and activities of antioxidant enzymes and lower cell membrane damage under high temperature stress. In the transcriptome profile, 1430 probes showed obvious difference in expression between PH and NH. These genes were those of signal transduction, transcription, energy, defense, and protein destination and storage, respectively.
