Of these, 6 were similarily altered in the serum from the 263K infected hamsters Conclusion: These altered miRNA signatures may serve as the basis for non-invasive diagnostic assays for chronic wasting disease and may shed light on the pathogenesis of prion infection

Project description:Uremic pruritus (UP) is one of the common symptoms in patients with uremia, and its etiology and mechanism have not been fully understood. In view of the high incidence of UP, finding the specific changed proteins in blood of UP patients will help revealing the potential biological mechanisms of UP and explore biomarkers. In this study, LC-MS/MS based data independent acquisition (DIA) mode was used to analyze the serum samples of 54 UP patients (DKD-UP, HN-UP and GN-UP, n=18 for each), 18 uremic patients without pruritus (Negative) and 18 CKD patients without pruritus (CKD).

Project description:Primary Open-Angle Glaucoma (POAG) and Pseudoexfoliation Glaucoma (PEXG) are forms of glaucoma with distinct etiologies, but they share a common type of optic neurodegeneration that leads to irreversible loss of retinal ganglion cells, visual field and blindness if left untreated. The main objective in the present study was to explore whether differential-proteomic analysis of blood serum from patients with POAG or PEXG could lead to the identification of candidate biomarkers of glaucoma and provide clues to pathogenic mechanisms. A total of 202 blood samples from medically treated patients with POAG (n=73), PEXG (n=59), and cataracts (control, n=70) were collected and analyzed following a proteomic workflow that involved two phases: discovery and validation. In the discovery phase, differential-proteomics (2D-DIGE) methodology led to the identification of a panel of 35 proteins that were altered (i.e., over- or under-expressed) in serum samples of glaucoma patients. Functional pathway analysis suggested a potential involvement of these proteins in immunological and inflammatory pathways. In the validation phase, ELISA assays of the top-17-ranked proteins confirmed that most of them were over-expressed in the glaucoma groups. Stepwise discriminant analysis of the ELISA data resulted in a six-protein panel (APOA4, TF, C3, APOL1, IGHG2, C4A) that was used to generate multivariate predictive models. These models achieved a diagnostic efficiency of 81% (correct assignment), specificity of 83%, and sensitivity of 92%. Specifically, 100% efficacy was reached in the discrimination of the POAG group from control, and 86% efficacy in the discrimination of POAG from PEXG. Overall, these data demonstrate the discovery of a panel of serum biomarkers that could be used in large-scale multiplexed screenings for the diagnosis of POAG and PEXG patients. Furthermore, these biomarkers may have a role in the pathogenesis, and association to immune-inflammatory processes with glaucoma.

Methodology/Principal Findings: Serum samples were obtained from patients with clinical diagnoses of PD (n=117), MSA (n=31) and PSP/CBS (n=38) and 99 controls. Cytokine profiles of sera of patients and controls were analyzed with a semiquantitative human cytokine antibody array. In a next step, significantly altered cytokines were individually validated by immunoassays. The cytokine array revealed a significantly altered expression of 12 cytokines. Immunoassay validation confirmed a significant increase of PDGF-BB in PSP/CBS, MSA and PD and a decrease of Prolactin in PD (Kruskal-Wallis p<0.05). A multivariate analysis taking into account diagnoses anti-Parkinsonian treatment, sex and age revealed that PDGF-BB levels were influenced only by the diagnoses (p<0.001), whereas Prolactin levels were influenced only by anti-Parkinsonian treatment (p<0.001). These findings could be corroborated by a subgroup analysis in untreated patients. Conclusions/Significance: In our unbiased cytokine array screening approach we found PDGF-BB to be elevated in PSP/CBS, MSA and PD. Increased PDGF-BB levels might be of relevance in a model of molecular biomarkers for Parkinsonian syndromes. Project description:Exosomes were purified from 250 ul serum using ExoQuickTm. The presence of particles consistent in size with exosomes (60-150nm) was confirmed using a Nanosight LM10. miRNA was extracted from exosomes using an miRNeasy Serum/Plasma kit (Qiagen, #217184). miRNA was reversed transcribed using a TaqMan® microRNA Reverse Transcription Kit (Life technologies, #4366596). miRNA profiling was performed with a high throughput TaqMan® OpenArray® Human microRNA panel (Life technologies, #4461104). The panel consisted of probes for 754 human miRNAs that are based on miRNA sequences derived from Sanger miRBase v14. MegaplexTM Primer Human Pool A v2.1 and Human Pool B v2.0 or v3.0 The poor prognosis and rising incidence of oesophageal adenocarcinoma highlight the need for improved methods for detection of this cancer. Molecular biomarkers offer potential for this. The potential for circulating miRNAs as biomarkers in some other cancers has been shown, but circulating miRNAs have not been well characterized in oesophageal adenocarcinoma. This study investigated whether circulating miRNAs could be used to detect oesophageal adenocarcinoma.

Project description:The study is based on the identification of serum protein biomarkers for NT 1. A total of 27 serum samples were collected (16 NT 1 and 11 controls), the altered expression levels of low-abundant proteins (DEPs) associated with narcolepsy type 1 was investigated by comparing the proteomes of the two sample cohorts. DEPs that are revealed to be most related to NT 1 pathogenesis and have high levels of fold changes were further targeted and validated by LC-PRM-MS. Then, the effects of the DEPs were explored by system biology analysis.
