Excretory-secretory products of T. canis

Project description:Background: Toxocara canis is a significant zoonotic parasitic nematode with a worldwide distribution. While researches have been conducted on the excretory-secretory (ES) proteins of T. canis L3 larvae and the non-protein components of adult excretory-secretory products (ESPs), a comprehensive molecular characterization of adult-stage T. canis ES proteins remains lacking. Methods: The protein expression profiles of ES proteins were identified from female and male adult T. canis using Orbitrap Astral analyzer. Proteins with differential abundance (PDAs) between the sexes were analyzed, including subcellular localization, transcription factor, domain, GO annotation and KEGG enrichment analysis. The immune-interacting ES proteins were identified by co-immunoprecipitation (Co-IP) and mass spectrometry analysis. N-glycosite mapping in Brugia malayi female worms, male worms, and microfilariae

Project description:N-linked glycosylation is a critical post translational modification of eukaryotic proteins. N-linked glycans are present on surface and secreted filarial proteins that play a role in host parasite interactions. Examples of glycosylated Brugia malayi proteins have been previously identified but there has not been a systematic study of the N-linked glycoproteome of this or any other filarial parasite. In this study, we applied an enhanced N-glyco FASP protocol using an engineered carbohydrate-binding protein, Fbs1, to enrich N-glycosylated peptides for analysis by LC-MS/MS. We then mapped the N-glycosites on proteins from three host stages of the parasite: adult female, adult male and microfilariae. Fbs1 enrichment of N-glycosylated peptides enhanced the identification of N-glycosites. Our data identified 582 N-linked glycoproteins with 1273 N-glycosites. Gene ontology and cell localization prediction of the identified N-glycoproteins indicated that they were mostly membrane and extracellular proteins. Comparing results from adult female worms, adult male worms, and microfilariae, we find variability in N-glycosylation at the protein level as well as at the individual N-glycosite level. These variations are highlighted in cuticle N-glycoproteins and adult worm restricted N-glycoproteins as examples of proteins at the host parasite interface that are well positioned as potential therapeutic targets or biomarkers.

Male and female mouse heart miRNA sequencing

Project description:Purpose: The goal of this study is to identify the sex differential miRNA tanscriptome between male and female adult (4-6 wk. old) mouse hearts using small RNA-seq. Methods: miRNA profiles of male and female mouse hearts were generated by sequencing, n=4 for each sex, using Illumina HiSeq2500. The sequence reads were aligned to the mm10 reference genome using STAR via the bcbio-nextgen RNA-sequencing pipeline. Differential gene expression was determined by genwiz. Results: 11 miRNAs were differentially expressed between male and female mice [ P value <0.05, |log2(Fold Change)| > 0.5], with 9 more highly expressed in female hearts and 2 more highly expressed in male hearts.

A male-specific steroid hormone controls female reproductive biology in the malaria mosquito

Project description:Insects, unlike vertebrates, are generally believed to lack steroid hormones with functions predominantly associated with adult male biology. In the malaria mosquito Anopheles gambiae, the ecdysteroid 20-hydroxyecdysone (20E) appears to both control egg development in females and induce mating refractoriness and oviposition when sexually transferred by males. Here we show that these sex-specific functions are instead carried out by distinct steroids. We identify a male-specific oxidized form of 20E (3D20E) that upon sexual transfer switches off female mating receptivity, ensuring male paternity. Endogenous female 20E does not induce mating refractoriness, while it triggers oviposition in mated females when expression of a 20E-inhibiting kinase is repressed. 3D20E and 20E have different downstream targets, with 3D20E inducing expression of a tolerance factor that preserves female fitness during Plasmodium infection. The evolution of this male steroid has therefore not only shaped the mating biology of An. gambiae, but also impacted malaria transmission.
