Influence of Water Chemistry on Toxicogenomics of Chronic Lead Toxicity to the Fathead Minnow (Pimephales promelas)

Project description:Despite major successes in reducing the risks of lead (Pb) exposure over the past few decades, two issues of considerable importance remain unresolved: (1) how differences in water chemistry influence acute and chronic Pb toxicity, and (2) the elucidation of specific toxic mechanisms and modes of action (MOA). To more clearly define the water chemistry parameters mediating Pb toxicity we evaluated the effects of hardness (as CaSO4) and DOC (as humic acid (HA)) during chronic (150d) exposures to the fathead minnow (Pimephales promelas). Traditional toxicological endpoints were examined alongside gene expression analyses to help clarify the underlying mechanisms and MOA of Pb toxicity and to identify robust molecular markers of exposure and effect. Keywords: time course, chronic lead (Pb) exposure To analyze the gene expression responses to low-level Pb exposures fish were exposed +/- Pb in low ionic strength base water and fish collected at 2d, 4d, 10d, and 30d for microarray analysis. Equal amounts of RNA from all no lead controls were pooled (18 fish total for 2d & 4d exposures; 12 fish total for 10d & 30d exposures) as reference samples for hybridization with each of 3 separate biological replicate pools (6 fish total for 2d & 4d exposures; 4 fish total for 10d & 30d exposures) of RNA from age-matched, lead-exposed fish. Each pair of hybridizations was repeated with dye-swaps. Additional repeats were performed for various arrays to ensure quality of data obtained from initial hybridization.

Gene expression profiling and developmental outcomes reveals early toxicological mechanisms of lead effects in an early-life stage amphibian, Xenopus laevis

Project description:Lead (Pb) is a widespread environmental toxicant that can elicit a wide variety of adverse effects in aquatic species. In fish, exposure to Pb can induce oxidative stress and reduce antioxidant capacity and there is evidence that Pb-induced developmental abnormalities are the result of dysregulated metabolism pathways but to date, no studies have characterised the molecular mechanisms of lead toxicity in amphibians. To investigate this, we coupled transcriptomic responses and early-life stage toxicity testing to identify specific molecular mechanisms that drive the adverse effects of lead, in Xenopus laevis. Embryos were exposed to one of the concentrations (0, 70, 210 and 630 ng/L) of lead (II) nitrate starting from 2 days post fertilization (dpf) until 6 dpf and 21 dpf to examine the transcriptomic and apical responses, respectively. We observed common dysregulated pathways and with similar magnitude at medium and high lead concentrations, but a phenotypic response only with the highest lead level. RNA Seq analysis showed about ~250 differentially expressed genes that could be further categorized into KEGG pathways based on their involvement in key biological processes. The most dysregulated pathways were Glutathione metabolism, Cytochrome P450 (drug/ xenobiotic metabolism) and Steroid hormone biosynthesis. Developmental studies showed that tadpoles exposed to the highest Pb concentration (630 ug/L) had significantly decreased total length and higher proportion of developmental abnormalities. Therefore, evaluating gene expression responses can provide early indication of toxicological mechanisms of lead effects in amphibians. As with fish, dysregulated metabolic pathways are most likely the molecular mechanisms underlying lead toxicity in ELS amphibians and contribute to the altered growth and developmental abnormalities observed in this study.

Global Gene Expression Analysis Reveals Dynamic and Developmental Stage–Dependent Enrichment of Lead-Induced Neurological Gene Alterations

Project description:Background: The underlying genetic mechanisms specific to subtle neurological alterations associated with environmental lead (Pb) exposures have not been clearly elucidated. Objectives: The goal of this study was to identify novel gene targets and the underlying genetic mechanisms associated with developmental Pb neurotoxicity. Methods: We first exposed zebrafish embryos to a range of Pb concentrations throughout early development to establish relative toxicity. Using the data from that experiment, we exposed another group of zebrafish embryos to a sublethal dose of Pb (100 ppb) immediately after fertilization through 72 or 120 hr postfertilization (hpf). Global gene expression was then analyzed for molecular pathways and gene ontology enrichment, and Western blot analysis was performed to investigate the translation of gene expression changes to protein levels. Results: After 72 hpf, we identified 231 probes representing 90 nonredundant genes with well-established function or orthology to human genes as being altered by Pb exposure. This gene set was both confirmatory and novel in nature and was highly enriched for neurological development, function, and disease. Moreover, gene changes at this time point were correlated to altered protein levels. Alternatively, the gene set at 120 hpf did not share association with neurological development. Conclusions: Global gene expression alterations associated with developmental Pb exposure were dynamic and dependent on developmental stage. Gene expression alterations at the 72‑hpf time point were highly enriched with genes and molecular pathways associated with neurological development and disease. Mechanistic Insights into Lead Acetate-Induced Neurotoxicity Using RNA-Seq Analysis

Project description:Lead acetate (Pb-acet) is a neurotoxicant that significantly impacts human brain development. This study used LUHMES cells, a human embryonic stem cell-derived neuronal model, to explore its molecular toxicity mechanisms. RNA sequencing (RNA-seq) showed distinct gene expression changes due to Pb-acet exposure. Principal Component Analysis (PCA) highlighted significant gene expression alterations at high toxicant concentrations (Ld10) compared to low concentrations (Ld1) and controls. Pb-acet disrupted pathways related to ribosome function and RNA splicing, and activated endoplasmic reticulum (ER) stress pathways, including PERK, ATF6, and IRE1. It also upregulated genes linked to ER-associated degradation (e.g., SVIP) and apoptosis (Bcl2 and CHOP), and disrupted the ubiquitin ligase complex, indicating impaired protein homeostasis. This study identifies ribosomal processes and ER stress pathways as potential biomarkers of Pb-acet exposure, demonstrating the utility of LUHMES cells in neurodevelopmental toxicity and environmental biomarker research.

Exposure to the heavy metal lead induces DNA copy number alterations in zebrafish cells

Project description:DNA copy number variants (CNVs) are associated with the development of complex neurological diseases and disorders including autism spectrum disorder, schizophrenia, Alzheimer’s disease, and Parkinson’s disease. Exposure to multiple environmental chemicals including various heavy metals is suggested as a risk factor in these neurological diseases and disorders, but few studies have addressed if heavy metal exposure can result in spontaneous DNA copy number changes as a genetic mechanism contributing to these disease outcomes. In this study to further investigate the relationship between heavy metal exposure and spontaneous copy number alterations (CNAs), zebrafish fibroblast cells were exposed to the neurotoxicant lead (Pb). A crystal violet assay was first used to determine exposure concentrations with greater than 80% cell confluency. Then a zebrafish-specific array comparative genomic hybridization (CGH) platform was used to detect CNAs following a 72 hour Pb exposure (0.24, 2.4, or 24 μM). The Pb exposure resulted in 72 CNA amplifications ranging in size from 5 to 329 kb. No deletions were detected. CNAs resulted in 15 copy number alteration regions (CNARs), leaving 7 singlet CNAs. Two of the singlets were within high repeat genomic locations. The number of CNAs tended to increase in a concentration dependent manner. Several CNARs encompassed genes previously reported to have altered expression with Pb exposure, suggesting a mechanistic link. In addition, almost all genes are associated within a molecular network with the amyloid precursor protein (APP). Overall, these findings show that Pb exposure results in spontaneous CNAs that could serve as a mechanism driving adverse health outcomes associated with Pb toxicity including neurological disease pathogenesis for further study.

Lead exposure induced transgenerational developmental neurotoxicity by altering genome methylation in Drosophila

Project description:Heavy metal toxicity is a worldwide health problem. Lead exposure is of particular concern due to the adverse effects of low concentrations on cognitive development in children. Although the mechanism of lead neurotoxicity has been well studied, the analysis and molecular mechanism of the transgenerational effects of lead exposure-induced neurotoxicity are lacking. To address this, Drosophila, a powerful developmental animal model, was exposed to lead acetate. We found that Pb exposure during the developmental stage affected the neurodevelopment of F0 fruit flies, resulting in a loss of the correlation between the motor terminal area and muscle fiber area and the increased frequency of β-lobe midline crossing phenotype in Mushroom bodies. We also found that Pb exposure led to an increase in BRP expression, suggesting an increase in synaptic vesicle release sites and a decrease in synaptic vesicle protein SYN expression in F0 generation. This explained the results of our electrophysiological data that Pb exposure led to an increase in the amplitude of evoked excitatory junctional potential (EJP) and an increase in the frequency of spontaneous excitatory junctional potential (mEJP). Our results further confirmed that the developmental neurotoxicity of parental Pb exposure has a transgenerational effect. Neurodevelopmental defects, abnormalities in synaptic function, and repetitive behavior also were observed in the F3 offspring of F0 exposed to lead. Our Medip-seq analysis showed that Pb exposure altered the DNA methylation levels of many neurodevelopmentally associated genes (eg, hppy, nrg, baz, and spn) in the F3 offspring of the F0 generation with Pb exposure. Our findings suggest that epigenetic mechanisms may underlie the transgenerational inheritance of acquired phenotypic traits resulting from exposure to environmental factors.
