Vagus Nerve Stimulation enhances remyelination and decreases innate neuroinflammation in a Multiple Sclerosis rodent model.

Project description:This study investigated the effect of Vagus Nerve Stimulation (VNS) on innate neuroinflammation and remyelination in lysolecithin (LPC) induced demyelination, a preclinical model for Multiple Sclerosis (MS). In a first experiment (demyelination experiment), LPC was injected in the corpus callosum of 33 Lewis rats, inducing a demyelinated lesion, and rats were treated with either continuously-cycled VNS (cVNS) or one-minute per day VNS (1minVNS) or sham VNS, from two days before the injection until three days post-injection (dpi), when they were killed for immunohistochemistry and proteomics analysis. This timepoint corresponded with a demyelinated lesion and peak inflammation. In a second experiment (remyelination experiment), 13 rats were analogously treated with either cVNS or sham from two days before LPC injection until 11 dpi, when they were killed for tissue prelevation for immunohistochemistry and proteomics. This timepoints corresponded with partial remyelination of the lesion. For proteomics analysis, 20 rats were randomly selected, namely five cVNS and five sham rats of the demyelination experiment, and five cVNS and five sham rats of the remyelination experiment.

Transcriptome sequencing identified RNAs associated with renal fibrosis in UUO rat model (urine)

Project description:Renal fibrosis as the final outcome of many renal diseases, has always been paid more attention to by the researchers. To better understand whether lncRNAs could be a player in this process or be a biomarker for renal fibrosis diagnosis, we compared transcriptome sequencing data on renal tissues and urine respectively between UUO and shamed (Sham) rat model. Numerous genes including lncRNAs with significant changes in their expression were identified. 24 lncRNAs were up-regulated and 79 lncRNAs were down-regulated in the renal tissues of the UUO rats. 625 lncRNAs were up-regulated and 177 lncRNAs were down-regulated in urines of the UUO rats. Among the lncRNAs upregulated in renal tissue of UUO rats, 19 lncRNAs were predicted containing several conserved Smad3 binding motifs in the promoter. Among them, lncRNAs with putative promoter containing more than 4 conserved Smad3 binding motifs were demonstrated to be induced by TGF-β significantly in NRK-52E cells. We further confirmed that lncRNA TCONS_00088786 and TCONS_01496394 were regulated by TGF-β stimulation and also can influence the expression of some fibrosis-related genes through a feedback loop. Based on transcriptome sequencing data, bioinformatics analysis and qRT-PCR detection, we also demonstrated lncRNA in Urine are detectable and might be a novel biomarker of renal fibrosis. These data provide new information about the involvement of lncRNAs in renal fibrosis, indicating that they may serve as candidate biomarkers or therapeutic targets in the future.

Transcription profiling of rat skeletal muscle after fine needle stimulation

Project description:Fine needle stimulation also known as acupuncture is a traditional Chinese medical practice which causes relief of pain. We demonstrated that it caused neovascularization and enhanced recovery of blood perfusion in a ischemic portion of skeletal muscle in rats with hindlimb ischemia. Therefore we evaluated the effect of fine needle stimulation on skeletal muscle at gene expression level Experiment Overall Design: With 0.14mm diameter stainless steel needle, a hundred pricks were applied in a 100 square mili-meter region on addductor portion of the left rat hindlimb. Right hindlimb was left intact as control. 24 hrs after fine needle stimulation, skeletal muscle sample was resected from adductor muscle of fine needle stimulated left hindlimb and non-stimulated right hindlimbs, respectively. Then gene expression analysis with microarray was conducted on each skelatal muscle sample.

Urine Large Extracellular Vesicle LC-MS/MS Data from a Chronic Kidney Disease Rat Model

Project description:This study examined large renal tubule extracellular vesicles (LRT-EVs) isolated from urine of a 5/6 nephrectomy (5/6Nx) rat model that recapitulates many elements of human chronic kidney disease (CKD). Within weeks of 5/6Nx rats spontaneously exhibit proximal tubular damage including brush boarder shedding and the production of LRT-EVs. Analysis of the LRT-EV size and a lack of TUNEL staining in 5/6Nx rats, suggested LRT-EVs to be distinct from exosomes, microvesicles, and apoptotic bodies.

Species difference in gene expression effect of trichloroethylene (TCE) between mouse and rat (rat)

Project description:TCE is a non-genotoxic hepatocarcinogen in mouse, but not in rat or human. Extrapolation of data from laboratory animals to humans is difficult due to species-specific differences. To identify molecular pathways and biological changes responsible for species-specific differences in hepatocarcinogenesis, we analyzed gene expression profiles of livers from B6C3F1 mice and SD rats administered TCE by oral gavage once or repeatedly every 24 hrs for 14 days. Gene expression analysis revealed distinct clusters of transcriptional profiles in single- and repeated-dose mice and rats. Pathway analysis showed differences in biological pathways between single- and repeated-dose mice and rats. Activation of the MAPK signaling cascade and ubiquitin-proteasome inhibitory function, as well as inhibition of TGF-beta signaling, were specific to mice and suggest a role in hepatocyte proliferation. Although pathological analysis showed no evidence of apoptosis, gene expression analysis revealed changes in apoptosis-related genes. In addition to the previously reported suppression of apoptosis, results in repeated-dose mice showed that toxicity induced by TCE in turn induces apoptosis. Keywords: Response to chemical
