TMT-based quantitative proteomic analysis of Fusarium oxysporum f. sp. radicis-lycopersici treated with linalool

ABSTRACT: The Forl strain was cultured on PDA supplied with 0.8 mL/L linalool for 6 days at 25°C. The fungal strain on PDA supplied with only 0.1% Tween80 was cultured as the control. Three biological replicates were established for each treatment. An appropriate amount of mycelia (100 mg) was taken and ground in liquid nitrogen. Protein were extracted using buffer and subjected to HPLC-MS.

Quantitative proteomic analysis of Phytophthora nicotianae treated with nanomaterial-coated DDA

Project description:The Phytophthora nicotianae strain was cultured in liquid oat medium at 28°C for 48 hours, followed by treatment with nanomaterial-coated DDA for an additional 6 hours. Control groups included non-coated DDA and empty nanoparticles, respectively. Three biological replicates were established for each treatment. An appropriate amount of mycelia (100 mg) was collected and ground in liquid nitrogen. Proteins were extracted using lysis buffer and analyzed by high-performance liquid chromatography-mass spectrometry (HPLC-MS).

Genome-wide expression profiling in a Fusarium graminearum lacking FgVelB, a velvet complex gene

Project description:Using a 3'-tiling microarray covering the whole F. graminearum genes, we carried out genome-wide expression analyses of both a wild type and a FgVelB deletion strainn of F. graminearum at a sexual stage (3 days after perithecial induction on carrot agar) Our study is the first report elucidating regulatory pathways controlled by FgVelB in Fusarium graminearum. A total of 4 chips were used for microarray. Total RNAs were extracted from perithecia and/or ground mycelia formed on carrot agar cultures of a self-fertile wild-type and a self-sterile FgVelB-deleted strain of F. graminearum with two biological replicates.

Genome wide expression responses of Fusarium oxysporum wt and M-NM-^ThapX mutant to iron depleted and iron replete conditions.

Project description:Soilborne fungal pathogens cause devastating yield losses, are highly persistent and difficult to control. To culminate infection, these organisms must cope with limited availability of iron. Here we show that the bZIP protein HapX functions as a key regulator of iron homeostasis and virulence in the vascular wilt fungus Fusarium oxysporum. Deletion of hapX does not affect iron uptake, but causes derepression of genes involved in iron-consuming pathways, leading to impaired growth under iron-depleted conditions. F. oxysporum strains lacking HapX are reduced in their capacity to invade and kill tomato plants and immunodepressed mice. The virulence defect of M-NM-^ThapX on tomato plants is exacerbated by coinoculation of roots with a biocontrol strain of Pseudomonas putida, but not with a siderophore-deficient mutant, indicating that HapX contributes to iron competition of F. oxysporum in the tomato rhizosphere. These results establish a conserved role for HapX-mediated iron homeostasis in fungal infection of plants and mammals. Iron dependent gene expression in Fusarium oxysporum wt and M-NM-^ThapX mutant was measured 1 hour after shifting the mycelia to minimal medium with or without 50 M-NM-<M Fe2(SO4)3. Three independent experiments were performed.

Effect of salicylic acid on Fusarium graminearum, the major causal agent of fusarium head blight in wheat

Project description:Salicylic acid (SA) is one of the key signal molecules in regulating plant resistance to diverse pathogens. It is predominantly associated with resistance against biotrophic and hemibiotrophic pathogens, and triggering systemic acquired resistance (SAR) in Arabidopsis. However, whether and how SA directly affects Fusarium graminearum and how SA influences the defence efficiency of wheat against fusarium head blight (FHB) are still poorly understood. Previous experiments have shown that the growth of F. graminearum mycelia and the germination of spores were significantly inhibited, and eventually stopped by increasing amounts of SA in both liquid and solid media cultures. Co-inoculation of SA and Fg spores has led to reduced FHB symptoms in the very susceptible Triticum aestivum cultivar ‘Roblin’. To better understand the effect of SA on F. graminearum mycelial growth, we have compared the expression profiles of SA-treated and untreated F. graminearum liquid cultures after 8 and 24 h of treatment, using an F. graminearum custom-commercial microarray. The microarray analysis suggests that F. graminearum can metabolize SA through two pathways, the gentisate and catechol pathways that are present in many fungal species. Additional experiments have confirmed the capacity of F. graminearum to metabolize SA. Our results demonstrate that, although F. graminearum has the capacity to metabolize SA, SA has a significant and direct impact on F. graminearum through a reduction in efficiency of germination and growth at higher concentrations.
