Proteome Profiling of Endogenous and Potential S-nitrolysation in Colorectal Cancer


ABSTRACT: Background. As a common cancer with high incidence rate and mortality, colorectal cancer (CRC) is seriously threatening human health. Nitric oxide (NO) mediated protein S-nitrosylation (SNO) is of great significance in the occurrence and development of CRC, melanoma, breast cancer and other cancers. Materials and methods. In this study, the proteomic results of human cancer tissues, para-cancerous tissues, SW480, NCM460 and SW620, which were treated by biotin switch and magnetic beads enrichment technology, were analyzed by mass spectrometry (MS). Through the comparison and analysis of MS results and literatures, some proteins with potential significance were identified in CRC. Results. 105 peptides with SNO sites were screened from human tissues. 128 peptides with SNO sites were screened from SW480 and NCM460 cell lines. Through the cross validation of MS results and literatures, several SNO proteins were identified frequently in CRC, such as the nucleoside diphosphate kinase A (NDK A), desmocollin-1, protein S100-A8, Pyruvate kinase PKM, Glyceraldehyde-3-phosphate dehydrogenase (GAPDH), etc. Conclusion. We believed that NDK A, desmocollin-1, protein S100-A8, Pyruvate kinase PKM, GAPDH and other related proteins had important biological significance in CRC.
