Cerebellum PC O-GlcNAcylation TMT analysis


ABSTRACT: The total protein in the sample was extracted, part of the sample was taken out for protein concentration determination and SDS-PAGE detection, the other part was taken out for trypsin enzymolysis and labeling, and then equal amount of each labeled sample was mixed for chromatographic separation. Finally, the sample was analyzed by LC-MS/MS and data analysis. After quality assessment and preprocessing, expression level analysis and function analysis were conducted respectively. Functional annotation analysis of the identified proteins was performed using several common databases. GO analysis, Pathway analysis and interaction analysis were conducted for the screened differential proteins, and correlation analysis, expression pattern clustering heat map and venn analysis were conducted for the data of the comparison group. In addition, according to the data situation, study the correlation or the part of interest, select the key proteins and their functions or pathways, and conduct subsequent key research and verification direction.

Purkinje cells (PCs), as a unique type of neurons output from the cerebellar cortex, are essential for the development and physiological function of the cerebellum.
