Serum proteomics of patients with Kashin-beck disease

ABSTRACT: Kashin-Beck disease (KBD), a chronic and endemic degenerative chronic joint disease, which etiolo. In order to discover biomarkers for KBD, the serum DIA proteomics analysis was conducted to detect the differentially expressed proteins between KBD patients and healthy controls.

Project description:Arthritogenic alphaviruses including chikungunya, Mayaro, and Ross River viruses cause long-lasting musculoskeletal pain and inflammation. However, mechanisms driving chronic disease remain poorly understood. Here, we investigated joint-associated tissues in alphavirus-infected mice at a late stage of infection. Utilizing scRNA-seq, spatial transcriptomics, and flow cytometry we identified an accumulation of inflammatory macrophages in joint-associated tissues with elevated Tnf, Nlrp3, Il1b, and H2-Aa expression, and these cells harbored CHIKV RNA. Moreover, we identified accumulation of CD4+ T cells in joint-associated tissues, which express Ifng. Depletion of CD4+ T cells diminished MHC-II expression on joint macrophages, highlighting their potential role in inflammation. In addition, treatment with a small molecule inhibitor of CHIKV replication during chronic disease reduced viral RNA and joint inflammation, suggesting that viral RNA replication promotes chronic joint disease. Our data suggest that macrophages harbor replicating viral RNA and contribute to the sustained joint inflammation associated with chronic alphavirus disease.

Proteomic Screening Identifies Novel Biomarkers and Mechanisms of Synovial Joint Disease in a Canine Model of Mucopolysaccharidosis I

Project description:Mucopolysaccharidosis I is a lysosomal storage disorder characterized by deficient alpha-L-iduronidase activity, leading to abnormal accumulation of glycosaminoglycans in cells and tissues. Synovial joint disease is prevalent and significantly reduces patient quality of life. There is a critical need for improved understanding of joint disease pathophysiology in MPS I, including specific biomarkers to predict and monitor joint disease progression, and response to treatment. The objective of this study was to leverage the naturally-occurring MPS I canine model and undertake an unbiased proteomic screen to identify systemic biomarkers predictive of local joint disease in MPS I. Synovial fluid and serum samples were collected from MPS I and healthy dogs at 12 months-of-age, and protein abundance characterized using LC MS/MS. Stifle joints were evaluated postmortem using magnetic resonance imaging (MRI) and histology. Proteomics identified 40 proteins for which abundance was significantly correlated between serum and synovial fluid, including markers of inflammatory joint disease and lysosomal dysfunction. Elevated expression of three biomarker candidates, matrix metalloproteinase 19, inter-alpha-trypsin inhibitor heavy-chain 3 and alpha-1-microglobulin, was confirmed in MPS I cartilage, and serum abundance of these molecules was found to correlate with MRI and histological degenerative grades. The candidate biomarkers identified in this study have the potential to improve patient care by facilitating minimally-invasive, specific assessment of joint disease severity, progression and response to therapeutic intervention.

Inhibition of Hedgehog signaling in human osteoarthritic cartilage

Project description:Osteoarthritis (OA) is a common degenerative disease of the joint. Data from our lab indicates that Hedgehog (Hh) signaling is activated in human OA and murine models of OA (Lin et al., 2009, Nature Medicine). To identify Hh target genes, microarray analyses were performed to detect changes in gene expression when the Hh pathway was inhibited in human OA cartilage samples. Identifying Hh target genes in chondrocytes will elucidate key regulatory networks that govern chondrocyte homeostasis and provide novel therapeutic strategies for OA.

Proteomic Analysis of Tear Fluid from the Offspring of Keratoconus Patients Toward Novel Biomarkers and Pathways Associated with Corneal Biomechanical Weakness and Disease Susceptibility.

Project description:Keratoconus (KC) is a chronic and degenerative condition marked by pathological weakening, thinning, and protrusion of the corneal tissue. Corneal biomechanical weakness constitutes a hallmark of early keratoconus (KC) stages, which has underlined the importance of the study of biomechanical response in the early clinical screening of KC. In this line, the family history of KC is consistently identified as the leading risk factor for KC development, and the first-order pediatric relatives of KC patients are considered the most vulnerable population for KC development. On the other hand, tear fluid has become an attractive and accessible source that could be useful in identifying the molecular targets of several ocular diseases, such as corneal ectasias and more specifically KC disease. So, based on the vulnerability of first-order pediatric KC relatives to develop the disease, as well as the potential of tear fluid as a biomarker source, the purpose of this study was to analyze quantitatively and qualitatively the tear proteome of young KC offspring using micro–liquid chromatography coupled to tandem mass spectrometry, trying to get closer to the molecular drivers underlying the alterations in corneal biomechanical properties at at-risk stages of KC and to provide new insights into the molecular mechanisms involved in the earlier stages of the disease.
