The healthy adult mice running for 6 weeks as exercise model and sedentary mice as control were used to perform transcriptomic, proteomic, acetyl-proteomics, and metabolomics analysis. In addition, correlation analysis between transcriptome and proteome, and proteome and metabolome was conducted as well. This dataset is transcriptomic data.

Project description:Exercise training is a potent treatment of NAFLD and hepatic insulin resistance. Here we provide molecular information about the hepatic mitochondrial metabolism in mice when chronic overnutrition (high-energy diet (HED) for 6 weeks) is combined with exercise training. Training reduced the hepatic triacylglycerol content, fasting insulin, and reversed glucose intolerance. Training modified the hepatic mitochondrial proteome with a decrease in enzymes related to pyruvate metabolism and entry of acetyl-CoA into the TCA cycle. Transcriptome data revealed down-regulation of glucose oxidation and lipogenesis. The mitochondrial respiratory capacity of trained HED-fed mice is increased despite reduced content of complex I. Training decreased diacylglycerol species and JNK phosphorylation, both of which can induce insulin resistance. Increased mitochondrial mass and oxidative capacity of the trained muscle further unburdens the liver from substrate overload. Together, when high fat and carbohydrate intake in mice is accompanied by exercise, the decline of mitochondrial function and insulin resistance can be prevented by modification of mitochondrial acetyl-CoA metabolism.

Project description:Previous research reports have shown that curcumin and exercise have good anti-tumor effects. However, there is currently a lack of relevant research reports on the combined effect of physical exercise with curcumin supplementation on cancer and its mechanisms. This study constructed an anti breast tumor mouse model under the combined effects of curcumin treatment and swimming exercise. Transcriptomics and metabolomics techniques were used to screen differentially expressed genes and metabolites, evaluate the anti-cancer effect, and analyze the molecular regulatory mechanisms related to metabolism. Through the mouse phenotype observation, including the detection of tumor appearance, tumor imaging in vivo, and HE staining results of pathological sections, curcumin administration and exercise intervention combined treatment has more obvious inhibitory effect on breast cancer than a single treatment. The combined effect treatment group had a total of 445 differentially expressed genes, including 154 upregulated genes and 291 downregulated genes; Functional enrichment analysis showed that calcium signaling pathway, Wnt signaling pathway, PI3K Akt signaling pathway and IL-17 signaling pathway significantly participated in the anti breast cancer process of curcumin combined exercise. The results of inter group differential metabolite analysis showed that the combined effect of curcumin and exercise involves two unique pathways: amino sugar and nucleotide sugar metabolism, as well as amino acid biosynthesis. The metabolic pathways of amino sugars and nucleotide sugars include chitosan, D-glucosamine 6-phosphate, L-fucose, and N-acetyl beta-mannosamine; The biosynthetic pathway of amino acids includes DL isoleucine, DL tyrosine, and homocysteine. Finally, through the joint analysis of transcriptome and metabolomic data, it was found that the most significantly correlated genes among the positive ion mode differential metabolites under the combined exercise of curcumin were catechins, including 8 correlated genes Cpeb2, Grem1, Nid1, Samd5, Hsph1, Glis3, Mapk6, and Phldb2; The most significant negative ion mode differential metabolite is 1-myristyl-2-hydroxy-sn-glycerol-3-phosphate ethanolamine, which includes 27 correlated genes Mmp10, Dock4, Dpysl3, Tnfsfm13, Etf1, Dzip1, Scara5, Plcg2, Dusp18, Zmat3, Marf1, Tox4, Tmem119, Tep1, Cybc1, AW549877, Dynap, Hhip, Tpm3, Zc3hav1l, Sema5a, Txlng, Lrrc55, Pprrx1, Rtl8b, Adam19, and Frrs1.

Project description:The pleiotropic transcription factor Yin Yang 1 (YY1) regulates cellular metabolism in a stage- and tissue-dependent manner. However, its role in cardiac energetic regulation remains unclear. We found that cardiac-specific YY1 knockout (cYY1 KO) mice exhibited cardiac dysfunction after two weeks of swimming exercise, suggesting an impaired adaptive cardiac response. Further analysis revealed that exercise-induced upregulation of transcription factor networks controlling cardiac metabolism, including peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α), was blunted in cYY1 KO mice. Consistently, citrate synthase activity, an indicator of mitochondrial content, was reduced in these mice. In response to transaortic constriction (TAC), cYY1 KO mice developed significantly exacerbated heart failure. RNA sequencing showed downregulation of metabolic genes and pathways, including PGC-1α, in TAC cYY1 KO mice hearts. Additionally, mitochondrial complex protein levels and complex I activity were reduced, consistent with suppressed metabolic gene transcription. Previously, we observed increased YY1 expression in hearts subjected to both exercise and TAC. However, while cardiac metabolism was enhanced with exercise, it was impaired with TAC. We found that p300 binding to YY1 increased during exercise, whereas histone deacetylase 3 (HDAC3) binding to YY1 increased in TAC hearts. These findings suggest that YY1-mediated transcriptional regulation of metabolic genes is modulated by the epigenetic regulators p300 and HDAC3.

Project description:The hippocampal proteins obtained from both exercised (moderate-intensity running for 6 weeks) and sedentary mice were separately analyzed for Liquid chromatography–tandem mass spectrometry (LC-MS/MS) identification with three replications. Acetylome data identified 3876 acetyl sites and 1764 acetylated proteins, which constituted 31% of the whole hippocampal proteome (totally 5725 proteins identified). In addition, 1305 proteins with 2769 acetyl sites were quantified. When defining the cutoffs for the fold change in abundance of 1.5 and P value < 0.05, 272 acetyl sites on 252 proteins were differentially regulated between exercise and control mice. Analysis of the subcellular localization showed that these differentially acetylated proteins (DAPs) exert different functions in multiple compartments, including the cytoplasm, mitochondria, nucleus and plasma membrane. The 19 up-regulated acetylated proteins mainly resided in mitochondria, while the 253 down-regulated DAPs were most abundant in cytoplasm, and the annotation of molecular functions were significantly enriched in catalytic activity and binding, respectively. Gene Ontology (GO) analysis showed that DAPs were primarily correlated with cytoskeleton, myelin sheath and axons in the cellular component category; and actin cytoskeleton organization, regulation of protein polymerization or depolymerization, and hexose/glucose metabolic process were significantly enriched in the biological process category. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that DAPs were involved in central carbon metabolism, neurodegeneration diseases, synapses, and various signaling pathways. Among them, chronic exercise induced significant alterations in phototransduction, carbon-related metabolism (carbohydrate digestion and absorption, glycolysis/gluconeogenesis, fructose and mannose metabolism, and pyruvate metabolism) and Hippo signaling pathway. Meanwhile, 21 proteins were significantly expressed, which were enriched in the pathway of complement and coagulation cascades.

Project description:To explore the effects of moderate intensity exercise on protein of lactylation in mouse muscle tissue metabolism. The healthy adult mice running for 6 weeks as exercise model and sedentary mice as control were used to perform transcriptomic, proteomic, lactylation-proteomics, and metabolomics analysis. In addition, correlation analysis between transcriptome and proteome, and proteome and metabolome was conducted as well. In this study, 159 lactate sites of 78 proteins were identified to be differentically regulated by moderate intensity exercise. Enrichment analysis showed that lactated proteins Mtatp8, Atp5mg and Atp5po exhibited ATP hydrolysis activity. They are involved in biological processes such as mitochondrial transmembrane transport, and Mtatp8, Atp5mg and Atp5po participate in Oxidative phosphorylation and Thermogenesis pathways. The lactation levels of Mtatp8, Atp5mg and Atp5po proteins in exercise group were significantly decreased, while their protein levels were significantly increased. The combined analysis of proteomics and metabolomics showed that the Oxocarboxylic acid metabolism pathway and Sphingolipid signaling pathway had significant changes under the influence of moderate intensity exercise. Our results showed that moderate intensity exercise has a certain effect on the lactylation level of mice, possibly by reducing lactylation levels of Mtatp8, Atp5mg and Atp5po and increasing the expression of their protein levels, thereby regulating the Oxidative phosphorylation pathway and participating in energy metabolism. 2-Oxocarboxylic acid metabolism pathway and Sphingolipid signaling pathway need to be further explored.

Project description:Background: Exercise mimetics is a proposed class of therapeutics that specifically mimics or enhances the therapeutic effects of exercise. Muscle glycogen and lactate extrusion are critical for physical performance. The mechanism by which glycogen and lactate metabolism are manipulated during exercise remains unclear. This study aimed to assess the effect of miR-92b on the upregulation of exercise training-induced physical performance. Methods: Adeno-associated virus (AAV)-mediated skeletal muscle miR-92b overexpression in C57BLKS/J mice, and global knockout of miR-92b mice were used to explore the function of miR-92b in glycogen and lactate metabolism in skeletal muscle. AAV-mediated UGP2 or MCT4 knockdown in WT or miR-92 knockout mice was used to confirm whether miR-92b regulates glycogen and lactate metabolism in skeletal muscle through UGP2 and MCT4. Body weight, muscle weight, grip strength, running time and distance to exhaustion, and muscle histology were assessed. The expression levels of muscle mass-related and functionrelated proteins were analysed by immunoblotting or immunostaining. Results: Global knockout of miR-92b resulted in normal body weight and insulin sensitivity, but higher glycogen content before exercise exhaustion (0.8538 ± 0.0417 vs 1.043 ± 0.040, **P=0.0087), lower lactate levels after exercise exhaustion (4.133 ± 0.2589 vs 3.207 ± 0.2511, *P=0.0279), and better exercise capacity (running distance to exhaustion, 3616 ± 86.71 vs 4231 ± 90.29, ***P=0.0006; running time to exhaustion, 186.8 ± 8.027 vs 220.8 ± 3.156, **P=0.0028), as compared to those observed in the control mice. Mice skeletal muscle overexpressing miR-92b (both miR-92b-3p and miR-92b-5p) displayed lower glycogen content before exercise exhaustion (0.6318 ± 0.0231 vs 0.535 ± 0.0194, **P=0.0094), and higher lactate accumulation after exercise exhaustion (4.5 ± 0.2394 vs 5.467 ± 0.1892, *P=0.01), and poorer exercise capacity (running distance to exhaustion, 4005 ± 81.65 vs 3228 ± 149.8, ***P＜0.0001; running time to exhaustion, 225.5 ± 7.689 vs 163 ± 6.476, **P=0.001). Mechanistic analysis revealed that miR-92b-3p targets UDP-glucose pyrophosphorylase 2 (UGP2) expression to inhibit glycogen synthesis, while miR-92b-5p represses lactate extrusion by directly target monocarboxylate transporter 4 (MCT4). Knockdown of UGP2 and MCT4 reversed the effects observed in the absence of miR-92b in vivo. Conclusions: This study revealed regulatory pathways, including miR-92b-3p/UGP2/glycogen synthesis and miR-92b-5p/MCT4/lactate extrusion, which could be targeted to control exercise capacity.
