Project description:Pathogenesis of colorectal cancer (CRC) is associated with alterations in gut microbiome. Previous studies have focused on the changes of taxonomic abundances by metagenomics. Variations of the function of intestinal bacteria in CRC patients compared to healthy crowds remain largely unknown. Here we collected fecal samples from CRC patients and healthy volunteers and characterized their microbiome using quantitative metaproteomic method. We have identified and quantified 91,902 peptides, 30,062 gut microbial protein groups, and 195 genera of microbes. Among the proteins, 341 were found significantly different in abundance between the CRC patients and the healthy volunteers. Microbial proteins related to iron intake/transport; oxidative stress; and DNA replication, recombination, and repair were significantly alternated in abundance as a result of high local concentration of iron and high oxidative stress in the large intestine of CRC patients. Our study shows that metaproteomics can provide functional information on intestinal microflora that is of great value for pathogenesis research, and can help guide clinical diagnosis in the future.

This cross-sectional study aimed to compare the proteomic signatures of periodontitis patients using full-mouth periodontal sampling with those obtained from serum and saliva samples. This study analysed proteome profiles from these biological sources in three systemically healthy, non-smoking patients with stage III, and grade C periodontitis.

Project description:The composition of the salivary microbiota has been reported to differentiate between patients with periodontitis, dental caries and orally healthy individuals. Thus, the purpose of the present investigation was to compare metaproteomic profiles of saliva in oral health and disease. Stimulated saliva samples were collected from 10 patients with periodontitis, 10 patients with dental caries and 10 orally healthy individuals. Samples were analyzed by means of shotgun proteomics. 4161 different proteins were recorded out of which 1946 and 2090 were of bacterial and human origin respectively. The human proteomic profile displayed significant overexpression of the complement system and inflammatory mediators in periodontitis and dental caries. Bacterial proteomic profiles and functional annotation were very similar in health and disease. Data revealed multiple potential salivary proteomic biomarkers of oral disease. In addition, comparable bacterial functional profiles were observed in periodontitis, dental caries and oral health, which suggest that the salivary microbiota predominantly thrives in a planktonic state expressing no characteristic disease-associated metabolic activity. Future large-scale longitudinal studies are warranted to reveal the full potential of proteomic analysis of saliva as a biomarker of oral health and disease.

Project description:Periodontitis and hypertension often occur as comorbidities. In the face of severe periodontitis patients with hypertension, periodontists often have to give priority to control the patient's blood pressure before surgical treatment, and how to use drugs also brings a lot of inconvenience for clinical treatment. In the stage of periodontal basic treatment, it is a problem that needs to be solved to treat periodontal inflammation and control blood pressure at the same time, so as to achieve “co-treatment of comorbidities”. To this issue, we proposed a controlled-release composite hydrogel approach with dual antibacterial and anti-inflammatory activities as a resolution for pathogen-originated periodontitis. Specifically, chitosan (CS) with inherent antibacterial properties was crosslinked with antimicrobial peptide (AMP) modified polyethylene glycol (PEG) to form the dual antibacterial hydrogel (CS-PA). Subsequently, curcumin nanoparticles (CNP) were encapsulated in the hydrogel to impart anti-inflammatory activities. In the mouse model of periodontitis complicated with hypertension, CS-PA/CNP was applied to gingival sulcus, found that it produced a perfect therapeutic effect on periodontitis and hypertension at the same time. The antibacterial experiments showed that CS-PA/CNP had an excellent inhibitory effect on a variety of periodontal pathogens, demonstrated the potential regulatory ability of the microbiota. In addition, cytological studies revealed the molecular mechanism that CS-PA/CNP enhanced antioxidant capacity of macrophages through the glutathione metabolism pathway, while enhancing their anti-inflammatory capacity. In conclusion, CS-PA/CNP has demonstrated its superior therapeutic effect and potential clinical translational value in the treatment of periodontitis and hypertension, which also serves as a drug delivery platform to provide combinatorial therapeutic options for periodontitis with complicated pathogenesis.

Project description:Periodontitis is increasingly linked to diverse brain disorders, yet causal mechanisms remain elusive. Here we demonstrate that ligature-induced oral dysbiosis in mice is sufficient to perturb central function. Six weeks of periodontitis produced anxiety-like behavior and motor deficits, accompanied by microglial depletion, reduced neuronal activity and region-selective transcriptional down-regulation in the frontal cortex. Pharmacological microglial ablation phenocopied, and glucocorticoid-receptor blockade rescued, these abnormalities, implicating microglia and activation of the hypothalamic–pituitary–adrenal axis. 16S rRNA gene sequencing revealed significant shifts in oral and gut microbiota that were partially normalized by a broad-spectrum antibiotic cocktail. Antibiotics alone elevated corticosterone but did not affect microglia or behavior, indicating that dysbiosis and glucocorticoids act synergistically on the brain dysfunction. Antibiotic treatment restored microglial density and behaviors in ligature mice, despite plasma corticosterone levels remaining elevated and comparable to those in antibiotic-treated controls. Our findings suggest oral dysbiosis as a tractable driver of neuroimmune dysfunction and redefine periodontitis as a systemic disorder with direct consequences for brain health.
