Project description:Monkeys were treated with a single i.v 200 mg/kg dose of hPara.09 or Anti-gD (Isotype control). Brain was harvested on days 4 and 28, and RNA was extracted from the cerebral cortex. The following groups are included: Group 1: Day 4 gD 200mg/kg n=6; Group 2: Day 4 hPara.09 200 mg/kg n=6; Group 3: Day 28 gD 200mg/kg n= 6; Group 4: Day 28 hPara.09 200 mg/kg n=6 (24 animals total). To assess expression heterogeneity between brain samples, 2 different frontal cortex punches were taken from each monkey, resulting in 48 total samples.

Project description:To examine irreversible changes in the developing brain following seizures, juvenile inbred mice were intraperitoneally injected with kainate and nicotine. Experiment Overall Design: Kainate and nicotine were dissolved in sterile saline, and intraperitoneally injected into postnatal day 23 males at a dose of 15 mg/kg and 10 mg/kg, respectively. Control mice received a comparable injection of saline. The injection was repeated three times at 2-hour intervals in the daytime, and after one week, the P30 survivors were sacrificed for sample preparation. Neocortices and hippocampi were isolated from mice that had received the repeated injections of saline, kainate, and nicotine (n=6, 5, and 6 mice, respectively).

Project description:Aim: We investigated the effect of acute and chronic glucocorticoid exposure on the hepatic transcriptome and its dependency on active androgen signaling. Methods: Adult male mice were used in the experiments. For the chronic exposure, mice were implanted with CORT (20mg) or vehicle pellets and administered ENZA (40mg/kg/day) or solvent via oral gavage. For the acute exposure, mice were pre-treated with ENZA (40mg/kg/day) or solvent every 12 hours for 24 hours via oral gavage and subsequently subcutaneously injected with 3mg/kg CORT or vehicle. Livers of all animals were dissected after 3 hours and snap-frozen. Next, total RNA was isolated and send for 100bp paired-end sequencing by BGI.

Project description:MgSO4 is given to prevent preterms neurological deficits. We have investigated proper effects and hypoxia-ischemia interfering effects of MgSO4 on transcription in neonate mice at day 5 (a brain development stage mimicking preterm maturity), at neuroprotection dose (600 mg/kg). Pools of RNA from 6 forebrains (3 males and 3 females) were used for total RNA extraction at 5 times post single MgSO4 injection (1.5H, 3H, 6H, 12H, 24H). Separately, MgSO4 effects were assessed post hypoxia-ischemia (3H and 12H). Co-hybridization of MgSO4-treated mRNA (labelled Cy5) and control tissues mRNA (labelled Cy3) were performed on Agilent (design 026655) micro-arrays in 4-7 replicates. Co-hybridization of saline injected (Labelled Cy3) or MgSO4 injected (Labelled Cy5) hypoxia-ischemia (according to Rice Vannucci (RV) procedure) exposed mice brains were performed on Aglient (design 026655) microarrays in triplicates.

Project description:F344 rats were divided into 4 groups: vehicle control, DEN, DEN+CLO and CLO. After one week of basal diet, rats belonging to the groups DEN and DEN+CLO underwent intraperitoneal injection of DEN (30mg/kg body weight) dissolved in NaCl 9‰. The two other groups were injected with NaCl 9‰ alone. At 30mg/kg, DEN is non-necrogenic thus only exhibiting initiating properties. Twelve days after injection, diet from rats belonging to the groups CLO and DEN+CLO was changed for diet containing 5000ppm CLO for up to 608 days. Series of 5 rats from each group were necropsied at days 18, 46, 102, 264, 377, 447 (reverse phase from day 377: no more CLO in the diet for rats belonging to the DEN+CLO group), and for the CLO and Control group 524, and 608 days after the injection of DEN or saline. Project description:Zebrafish transgenic lines Tg(fabp10a:dsRed), Tg(hand2:EGFP) and Tg(kdrl:ras-mCherry) in AB wild-type background were anesthetized with MS-222 and adult females were injected intraperitoneally with 500 mg/kg thioacetamide (TAA) or sterile water as a control 6 times over the course of 2 weeks. We have characterized transcriptomic profiles of FACS-isolated hepatocytes (dsRed+), stellate cells (EGFP+) and liver endothelial cells (mCherry+) from fishes treated with TAA or sterile water. Cells negative for the fluorescence were used as a control.

Project description:In this study we investigated whether nicotinamide mononucleotide (NMN) could protect against Doxorubicin-induced cardiotoxicity and physical dysfunction in vivo. To assess the short- and long-term toxicity, two Doxo regimens were tested, acute and chronic. In the acute study, C57BL6/J (B6) mice were injected intraperitoneally (i.p.) once with Doxo (20 mg/kg) and NMN (180 mg/kg/day, i.p.) was administered daily for five days before and after the Doxo injection. In the chronic study, B6 mice received a cumulative dose of 20 mg/kg Doxo administered in fractionated doses for five days. NMN (500 mg/kg/day) was supplied in the mice’s drinking water beginning five days before the first injection of Doxo and continuing for 60 days after.

Project description:Global gene expression profiling reveals a potential anti-melanoma effect of SQ-diEG in B16F10 lung colonization mice model. Squalene (SQ) was considered as a promising natural agent in anti-cancer treatment due to its strong antioxidant and anti-inflammatory activity; however, its pharmacological value has been largely underestimated because of its poor solubility and bioavailability. To adress this problem, a novel amphiphilic SQ derivative which bearing ethylene glycol oligomers was synthesized and was used as a permeation enhancer in this study to check its potential effect on anti-melanoma metastasis using B16F10 lung colonization mice model. Total 40 six years old male C57BL/6J mice were randomly divided into five groups (8 mice per group), including the control "(-)B16F10 injected and water treated"; the model "(+) B16F10 injected and water treated"; the positive control "(+) B16F10 injected and DTIC treated (Dacarbazine, 70 mg/kg/day)"; the SQ-diEG treatment "(+) B16F10 injected and SQ-diEG treated (5 mg/kg/day)"; the SQ-diEG treatment "(+) B16F10 injected and SQ-diEG treated (25 mg/kg/day)". After one week of acclimatization, B16F10 were murine melanoma cells line which intravenously injected (200000 tumor cells in 100 μl PBS, 0.2 ml/mouse) through the tail vein of mice. Mice was orally administrated with Dacarbazine (DTIC, positive control, 70mg/kg), and SQ-diEG (5mg/kg and 25mg/kg) for 20 days. On the last day of administration, mice was sacrificed and its lung was washed and collect for the further RNA extraction. Finally, the mice microarray gene expression profiling was conducted for two biological replicates.
