Carnosol alleviating C26 induced Cancer Cachexia

ABSTRACT: Shotgun proteomics using LC-high resolution tandem mass spectrometry was applied to check the protein expression profiles of C2C12 myotubes in control group (treated with solvent control 0.1% DMSO), C26 medium group (treated with conditioned medium of C26 tumor cells for 48 h), CS group (treated with car-nosol at 20 μM for 48 h) and C26+CS group (treated with conditioned medium of C26 tumor cells in the presence of 20 μM carnosol for 48 h). Samples of three independent experiments were collected.

Project description:Proteins secreted into the conditioned medium by human monocytic cell line, THP-1, or human buffy coat-derived monocytes polarised to either an M1 (incubation for 48 h with 100 ng/mL LPS and 20 ng/mL IFN-gamma) or M2 (incubation for 48 h with 20 ng/mL IL-4) phenotype. The conditioned medium was enriched for anionic molecules (including proteoglycans) by anion exchange chromatography prior to analysis.

Gene expression in rat hippocampal CA1 during the extinction of contextual fear memory

Project description:Adult rats (male Lister hooded) were pre-exposed for 3 d for 10 min /d (24 hr interval) to two different novel contexts. On day 4 rats received a single unsignaled scrambled footshock (0.5 mA for 2 s) 2 min into a 3min exposure to one of the two contexts. Rats were either returned to the CS conditioned context (CS+, EXT group) or to the other (CS-, NOR group) context for 10 min, 48h after contextual fear conditioning. Rats were housed in the home cages at all times outside the conditioning and re-exposure components. The novel context chosen to be the conditioned context was counterbalanced across the experimental groups. Rats were killed by CO2 inhalation 2 hours after CS- or CS+ exposure. CA1-enriched dorsal hippocampus was immediately dissected on ice before rapid freezing and storage at -80oC. The CS+ group represents rats that underwent extinction of contextual fear memory.

Bulk RNA sequencing of cytotoxic T lymphocytes of OT1 mice

Project description:We found that MC38 tumor-conditioned medium(TCM) treatment significantly impacted multiple pathways including cholesterol metabolism pathway of CTLs. Further analysis of cholesterol metabolism pathway, we observed that Cholesterol-25-hydroxylase(Ch25h) expression was significantly downregulated after treated with TCM compared to treated with fibroblasted-conditioned medium(FCM). In the meanwhile, we found that activating transcription fator 3(Atf3) was signifcantly upregulated after TCM treatment. Finally, we investigated the Atf3-Ch25h axis regulating CTLs function in tumor context.

Expression data from human acute lymphoblastic leukemia cell line SupB15 and its drug resistant subpopulations SupB15-MR

Project description:SupB15 cells are highly sensitive to Mitoxantrone treatment. To obtain drug resistant subpopulations, SupB15 cells were incubated in normal medium or BMSC derived conditioned medium, treated with 10nM of Mitoxantrone for 6 days. SupB15 cells in normal medium could not survive. a small population of SupB15 cells in conditioned medium group survived the treatment and ecpanded, termed as SupB15-MR. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes during this process.
