Project description:While RNA silencing is crucial for plant resistance against viruses, the cellular connections between RNA silencing and antiviral responses in plants remain poorly understood. In this study, we aim to investigate this relationship by examining the subcellular localization of small RNA loading and viral replication in Arabidopsis. Our findings reveal that Argonaute 2 (AGO2), a key component of RNA silencing, loads small RNAs at the endoplasmic reticulum (ER)-chloroplast membrane contact sites (MCSs). We identify a chloroplast-localized protein, RNA helicase 3 (RH3), which interacts with AGO2 and facilitates the loading of small RNAs into AGO2 at these MCSs. Furthermore, we discover that MCSs serve as replication sites for certain plant viruses. RH3 also promotes the loading of viral-derived small RNAs into AGO2, thereby enhancing plant antiviral resistance. Overall, our study sheds light on the roles of RH3 in RNA silencing and plant antiviral defenses, providing valuable insights into the cytobiological connections between RNA silencing, viral replication, and antiviral immunity.

This protein was not characterized so far. It is localized in chloroplasts, influences chloroplast biogenesis and chlorophyll biosynthesis and its role decreases with the plant age.

Project description:A pluripotent inducible cell line was generated from Arabidopsis to address the limitations of traditional in planta dicot systems and to develop a well-controlled experimental setup where the individual regulatory check points of chloroplast development could be defined. Following light exposure, this cell line was shown to differentiate into photosynthetically active cells with functional chloroplasts providing an experimental system with a temporal gradient of chloroplast development. Using this unique cell line from the dicot Arabidopsis we could demonstrate that the development from a proplastid to a functional chloroplast, and thereby the establishment of photosynthesis, is dependent on a regulatory mechanism involving two distinct phases. First, light exposure triggers an initial change in gene expression, metabolite profile, chlorophyll accumulation and plastid structure. Secondly, a second signal, most likely triggered by activation of the chloroplast, is required for full transition to a functional chloroplast and the shift from heterotrophic to photoautotrophic metabolism.

Project description:Chloroplasts are the metabolically most active compartment of mature leaf cells. Their proteins are involved in essential cellular processes, such as photosynthesis, nitrogen fixation, and fatty acid synthesis. For this, chloroplast proteins have to associate with other fellow organellar proteins to form functional units, react to changing environmental conditions, or optimize efficiency of biochemical reactions. We here investigated chloroplast protein-protein interactions by a combination of complexome profiling and cross-linking mass spectrometry (CX-MS). Different detergents and MS settings were tested for developing a new workflow, which was found to produce data of improved quality when comparted to standard complexome profiling approaches. This procedure was applied to chloroplasts acclimated to increasing light intensities to investigate the role of protein-protein interactions in the adaption to these conditions.

Project description:The lipid-derived signal jasmonate (JA) controls growth-defense balance by modulating the abundance of JAZ proteins that repress MYC transcription factors. In Arabidopsis, the JAZ-MYC regulon also promotes JA-induced degreening of detached leaves under conditions of dark-induced starvation. These observations, together with the finding that JA biosynthesis and signaling mutants do not exhibit obvious defects in natural senescence, have confounded a satisfactory explanation of the physiological significance of JA-induced chlorosis. Using intact Arabidopsis plants grown under normal laboratory conditions, we manipulated JAZ abundance to investigate JA-induced degreening as a component of the leaf growth-defense syndrome. A jaz decuple mutant (jazD) lacking most JAZs exhibited strong growth-defense tradeoffs without visible symptoms of senescence. Further depletion of JAZ by treatment of jazD with coronatine recapitulated the major hallmarks of senescence, including the loss of chlorophyll and photosynthetic capacity, turnover of chloroplast proteins and lipids, and transition of chloroplasts to gerontoplasts. The data highlight the regulation of senescence by jasmonate signaling along the growth-defense continuum.
