Proteomics analysis of Microbacterium sediminis YLB-01

ABSTRACT: Microbacterium sediminis YLB-01, a marine microbacterium tolerant to pressure and low temperature, was subjected to high pressure (accompanied by low temperature) and proteomic analysis was carried out to explore the mechanism of its adaptation to the high-pressure environment of the deep sea.

Adaptations to high pressure of Nautilia sp. strain PV-1, a piezophilic Epsilonproteobacterium isolated from a deep-sea hydrothermal vent

Project description:Physiological and gene expression studies of deep-sea bacteria under pressure conditions similar to those experienced in their natural habitat are critical to understand growth kinetics and metabolic adaptations to in situ conditions. The Epslilonproteobacterium, Nautilia sp. strain PV1, was isolated from hydrothermal fluids released from an active deep-sea hydrothermal vent at 9°N on the East Pacific Rise. Using a high pressure/high temperature continuous culture system we established that strain PV-1 has the shortest generation time of all known piezophilic microorganisms and we investigated its protein expression pattern in response to different hydrostatic pressures. Proteomic analyses of strain PV-1 grown at 200 Bars and 5 Bars showed that pressure adaptation is not restricted only to stress response or homeoviscous adaptation, but that it is more diversified and protein specific, with a fine and variegated regulation of enzymes involved even in the same metabolic pathway. As previously reported, proteins synthesis, motility, transport and energy metabolism are all affected by pressure, although to different extents. In strain PV-1, low pressure condition seems to activate the synthesis of phage-related proteins and an overexpression of enzymes involved in central carbon metabolism.

Analysis of physiological and gene expression changes related to Vibrio harveyi adaptation in the time of global warming

Project description:Vibrio species represent one of the most diverse genera of marine bacteria known for their ubiquitous presence in natural aquatic systems. Several members of this genus including Vibrio harveyi are receiving increasing attention lately because they are becoming a source of health problems, especially for some marine organisms widely used in sea food industry. To learn about adaptation changes triggered by V. harveyi during its long-term persistence at elevated temperatures, we studied adaptation of this marine bacterium in sea water microcosms at 30 oC that closely mimicks the upper limits of sea surface temperatures recorded around the globe.

Effect of heat shock at 500 atm and pressure shock to 500 atm on Methanocaldococcus jannaschii

Project description:Growth and transcriptional profiles of the barophilic methanarchaeon Methanocaldococcus jannaschii were studied at temperatures up to 98C and pressures up to 500 atm. Application of 500 atm of hyperbaric pressure shifted the optimal growth temperature upwards, and heat shock from 88C to 98C at 500 atm resulted in termination of growth. Pressure shock of M. jannaschii from 7.8 to 500 atm over 15-min, the first pressure upshift reported for a barophile, did not accelerate growth. Transcriptional profiles indicated a similar pressure response under growth and heat shock at 500 atm and pressure shock to 500 atm suggesting that the commonly affected genes are important for high-pressure adaptation. Factorial microarray design allowed de-convolution of the interacting effect of elevated pressure and heat shock on expression profiles, thus suggesting genes that may contribute to the organism’s survival in the turbulent in situ conditions of deep-sea hydrothermal vents. Gene expression profile in the pectoralis muscle of penguin before and after acclimatisation to marine life

Project description:In the present study, we were interested in gene expression changes in the pectoralis muscle of juvenile king penguins during the transition from terrestrial to marine life. Strictly terrestrial during their first year after hatching, king penguin chicks must then depart to sea to reach nutritional emancipation and pectoralis muscle is largely involved in penguin adaptation to the marine environment. To compare these transcriptomic profiles, we realized heterologous hybridization on Affymetrix GeneChip Chicken Genome Arrays, as the chicken is the closest model species for which microarrays are available. The development of a new algorithm, MaxRS, allow us to determine differentially expressed genes implicated in energetic metabolism or involved in cellular defense against reactive oxygen species and associated injuries. We compared muscle sample biopsy from 4 penguin juveniles captured just before they undergone their first immersion to cold water (named NI for Never Immersed) and 3 penguin juveniles that had completly accomplished their acclimation to marine life (named SA for Sea Acclimated).

Induction of DAN/TIR yeast cell wall mannoprotein genes in response to high hydrostatic pressure and low temperature

Project description:Global transcriptional profiles of Saccharomyces cerevisiae were studied following changes in growth conditions to high hydrostatic pressure and low temperature. These profiles were quantitatively very similar, encompassing 561 co-upregulated genes and 161 co-downregulated genes. In particular, expression of the DAN/TIR cell wall mannoprotein genes, which are generally expressed under hypoxia, were markedly upregulated by high pressure and low temperature, suggesting the overlapping regulatory networks of transcription. In support of the role of the mannoproteins in cell wall integrity, cells acquired resistance against treatment with SDS, Zymolyase and lethal level of high pressure when preincubated under high pressure and low temperature. Revealing the moleclar mechanisms of temperature acclimation and adaptation in marine diatoms with transcriptional responses to sub- and supra-optimal growth temperatures

Project description:Background: Ocean temperatures are projected to increase over the coming century, with dramatic consequences for the marine biosphere. Diatoms are important contributors to marine primary production and the ocean carbon cycle, yet the molecular mechanisms that regulate their acclimation and adaptation to temperature are poorly understood. Method: Here we use a transcriptomic approach to identify the molecular mechanisms associated with temperature acclimation and adaptation in closely related colder- and warmer-adapted diatom species. Results: We find contrasting patterns of differential expression at sub- and supra-optimal temperatures across the two species, which may be due to adaptive changes in baseline expression. Frontloaded and divested pathways indicate protein processing machinery, membrane structure, and the balance between temperature-independent photosynthesis and temperature-dependent metabolism are key elements of adaptation to temperature changes. Conclusions: Our findings suggest that transcriptional frontloading and divestment may provide a framework to interpret diatom acclimation and adaptation to temperature and success under future warming.
