Sericin Enhances Cryopreserved Sperm Quality in Chengde Hornless Black Goats by increasing Glutamine metabolism

Project description:<p>Cryopreservation of Chengde polled goat semen plays a critical role in conserving genetic resources, enhancing the utilization efficiency of superior breeding rams, and advancing artificial insemination techniques. However, spermatozoa are vulnerable to oxidative stress during the freezing process, which can significantly compromise sperm motility. This study demonstrated that supplementation of the semen cryoprotectant with 0.6% sericin significantly improved post-thaw sperm viability to 65.25% in Chengde hornless goats, while concurrently reducing both the sperm abnormality rate and intracellular ROS levels. Integrated TMT proteomics and LC/MS metabolomics identified 256 differentially expressed proteins and 109 differential metabolites between sericin supplementation and frozen control groups. Functional analysis revealed significant enrichment of differential metabolites, such as glutamine, in the alanine, aspartate, and glutamate metabolism pathway, concomitant with marked upregulation of antioxidant proteins including LRP8, GSTM3, and SIRT2. Thus, sericin enhances cryotolerance primarily by improving sperm viability, reducing oxidative damage, and sustaining energy metabolism. These findings establish the molecular foundation for optimizing goat semen cryopreservation and advancing artificial insemination technology.</p>

Maternal gestational nutrition perturbs offspring sperm epigenome in sheep [ncRNA-seq]

Project description:There is a growing body of evidence that inadequate maternal nutrition during gestation can have immediate and life-long effects on offspring. However, little is known about the reproductive effects of maternal gestational nutrition in offspring males. Here, using a sheep model of poor maternal nutrition (restricted- or over-feeding) during gestation, we found that poor maternal gestational nutrition does not affect semen characteristics (i.e. volume, sperm concentration, pH, sperm motility, sperm morphology) and scrotal circumference in offspring. However, by evaluating associations between poor maternal gestational nutrition and altered small non-cording RNAs (sncRNAs) and DNA methylation in offspring sperm, we demonstrated that poor maternal gestational nutrition alters sperm sncRNA composition and expression. Whole genome bisulfite sequencing further identified genomic regions with increased or decreased DNA methylation in sperm in response to poor maternal gestational nutrition. These findings imply that maternal diet-induced epigenetic errors can accumulate in sperm to worsen developmental outcomes of future generations.

Influence of the two different cryopreservation methods to inflammation of dental pulp cells

Project description:Dental pulp cells of cryopreserved teeth (slow and rapid speed) were examined with microarray for screening which gene is involve in the inflammation process during the cryopreservation process. Intact caries-free, freshly extracted premolars (n=6) were collected from 3 patients for microarray assay analysis. They were classified as control and cryopreserved groups. Cryopreserved groups were divided into rapid freezing and slow freezing group.

Cell freezing protocol suitable for ATAC-Seq on motor neurons derived from human induced pluripotent stem cells

Project description:We describe a protocol for freezing neuronal cells that is compatible with ATAC-Seq, producing results that compare well with those generated from fresh cells. We developed our protocol on a disease-relevant cell type, namely motor neurons differentiated from induced pluripotent stem cells from a patient affected by spinal muscular atrophy. We found that while flash-frozen motor neurons are not suitable for ATAC-Seq, the assay is successful with slow-cooled cryopreserved samples. Using this method, we were able to isolate high quality, intact nuclei, and we verified that epigenetic results from fresh and cryopreserved motor neurons agree quantitatively.

Proteomic analysis of fresh, equilibrated, and frozen/thawed turkey semen in relation to the extracellular fluid proteome

Project description:The cryopreservation of avian semen without significant loss of fertilizing ability is of practical interest to the poultry industry and for the conservation of genetic biodiversity. However, the unique physiological features make avian spermatozoa highly sensitive to damage caused by cryopreservation. The proteomic changes in turkey (Meleagris gallopavo) semen throughout the cryopreservation process have never been investigated previously. Therefore, the purpose of the present study was to evaluate the proteomic changes of extracellular fluid (ECF)of semen during cryopreservation at three stages (fresh, equilibrated, and frozen/thawed semen) using 2-dimensional difference in-gel electrophoresis (2D-DIGE) coupled with matrix-assisted laser desorption/ionization mass spectrometry (MALDI TOF/TOF). Six protein spots were significantly enriched in ECF proteome after equilibration compared to fresh semen. Twenty seven protein spots were enriched in ECF after cryopreservation compared to fresh semen. Twenty six protein spots were significantly enriched in ECF proteome after cryopreservation compared to equilibrated semen. The proteins enriched in ECF after cryopreservation were mainly related to NADH metabolic and glycolytic process.
