The targets study of HDCA in ameliorating NAFLD induced by high-calories diet by multi-omics technology


ABSTRACT: HDCA supplementation ameliorated diet-induced NAFLD in C57 wild type mice. In attempt to comprehensively understand the molecular mechanism occurring during the protective effect of HDCA, transcriptomic and proteomic profiles of mouse liver were first analyzed utilizing RNA-Sequencing and Tandem Mass Tag-based quantitative proteomic approach, respectively. Then, we screened for potential HDCA binding proteins by using biotin-labeled HDCA and human proteome microarray.

Nonalcoholic fatty liver disease (NAFLD) is usually characterized with disrupted bile acid (BA) homeostasis. However, the exact role of certain BA in NAFLD is poorly understood. Here we show levels of serum hyodeoxycholic acid (HDCA) decrease in both NAFLD patients and mice, as well as in liver and intestinal contents of NAFLD mice compared to their healthy counterparts.
