Ejection of mitochondria and TNF-α activation mediate hypertonicity-induced adipocytes dedifferentiation

ABSTRACT: Adipocyte extracellular vesicles and particles (EVs) proteomic profiling. In this study, we focus on the EVs proteomes changes induced by hypertonicity in 3T3-L1 and differentiated adipocytes. The source cells were cultured with isotonic/hypertonic medium for 48 hours, and EVs were isolated from the conditional medium using sequential ultracentrifugation. A label-free MS strategy was employed to achieve proteome identification and quantification.

Background
Recent studies demonstrated that elevated osmolarity could induce adipocyte dedifferentiation, representing an appealing procedure to generate multipotent stem cells. Here we aim to elucidate the molecular mechanisms that underlie osmotic induction of adipocyte reprogramming.
Methods
To induce dedifferentiation, the 3T3-L1 or SVF adipocytes were cultured under the hypertonic pressure in 2% PEG 300 medium. Adipocyte dedifferentiation was monitored by aspect ratio measure ...[more]
