Project description:Copper tolerance and sulfite tolerance are two well-studied phenotypic traits of Saccharomyces cerevisiae. The genetic bases of these traits are derived from allelic expansion at the CUP1 locus and reciprocal translocation at the SSU1 locus, respectively. Previous work identified a negative association between sulfite and copper tolerance in S. cerevisiae wine yeasts. Here we probe the relationship between sulfite and copper tolerance and show that an increase in CUP1 copy number does not impart copper tolerance in all S. cerevisiae wine yeast. Bulk-segregant QTL analysis was used to identify variance at SSU1 as a causative factor in copper sensitivity, which was verified by reciprocal hemizygosity analysis in a strain carrying 20 copies of CUP1. Transcriptional and proteomic analysis demonstrated that SSU1 over-expression did not suppress CUP1 transcription or constrain protein production but suggested that SSU1 overexpression induced sulfur limitation during exposure to copper. Finally, an SSU1 over-expressing strain exhibited increased sensitivity to moderately elevated copper concentrations in sulfur-limited medium, demonstrating that SSU1 over-expression burdens the sulfate assimilation pathway. Over-expression of MET 3/14/16, genes upstream of H2S production in the sulfate assimilation pathway increased the production of SO2 and H2S but did not improve copper sensitivity in an SSU1 overexpressing background. We conclude that copper and sulfite tolerance are conditional traits in S. cerevisiae and provide evidence of the metabolic basis for their mutual exclusivity. These findings suggest an evolutionary basis for the extreme amplification of CUP1 observed in some yeasts.

Project description:The gene expression profile of E. coli K-12 MG1655 grown in minimal medium supplemented with elevated copper concentrations (as copper-glycine) has been analysed using whole genome oligonucleotide microarrays. “Pool” RNA was isolated from five independently grown 50ml DMA cultures of E. coli K-12 MG1655. “Test” RNA was isolated from three independently grown 50ml DMA cultures of E. coli K-12 MG1655, for each growth condition (no added CuSO4, or supplemented with either 0.75mM or 2mM copper glycine). A sample of each culture was taken at mid to late exponential growth phase at OD600 0.8 (measured using a Pharmacia Biotech Ultrospec 2000 in a cuvette with a path length of 10mm). The cell viability and fermentation activity after freeze-thaw were dramatically decreased due to freeze-thaw injury. Because freeze-thaw injury involves complex phenomena, the mechanisms of it are not fully understood. We attempted to analyze the mechanisms of freeze-thaw injury by indirect gene expression analysis during post-thaw incubation after freeze-thaw treatment using DNA microarray profiling. The results showed that a high frequency of the genes involved in the homeostasis of metal ions were up-regulated depending on the freezing period. The phenotype of the deletion mutants of the up-regulated genes extracted by indirect gene expression analysis was assessed. The deletion strains of the MAC1 and CTR1 genes involved in copper ion homeostasis exhibited freeze-thaw sensitivity, suggesting that copper ion homeostasis is required for freeze-thaw tolerance. Supplementation with copper ions during post-thaw incubation increased intracellular superoxide dismutase activity. Inverse correlated with intracellular superoxide dismutase activity, intracellular levels of reactive oxygen species were decreased. Moreover, cell viability increased by supplementation with copper ions under specific assessment conditions. This study suggested that insufficiency of copper ion homeostasis may be one of the causes of freeze-thaw injury.

Project description:Analysis of genome-wide gene expression patterns in response to copper in WT and COPR mutant in the cyanobacterium Synechocystis sp PCC 6803. Here we have used microarrays to interrogate the global responses to copper additions at non-toxic (0.3 micromolar) and toxic concentrations (3 micromolar) of copper in WT and in a mutant in the copR gene. Addition of the non-toxic copper concentration stimulated the metabolism and induced the switch in the use from cyitochrome c6 to plastocyanin. In contrast, high copper addition induced a stress response and affected the metabolism of several other metals. This included repression of ribosomal, photosynthetic and metabolic genes and induction of chaperones and antioxidant enzymes genes. Finally CopRS seems to control only the expression of the copMRS and copBAC operons as all other genes that were differentially expressed by copper seemed to be unaffected in copR strains.

Project description:In frozen dough baking technology, baker’s yeast Saccharomyces cerevisiae encounter freeze-thaw injury. After thawing, dramatically decrease in cell viability and fermentation activity is caused by freeze-thaw injury. The freezing period is critical factor in freeze-thaw injury, thus we focused and investigated time-dependent gene expression profiles in recovery process from freeze injury. First, changes in gene expression profiles in S. cerevisiae in recovery process from freeze-thaw injury were analyzed using a DNA microarray. The results showed the genes which were involved in homeostasis of metal ions were time-dependent up-regulated 2-fold or more in a series. Then we examined whether these genes were related to tolerance in freeze-thaw injury by using deletion strain. The results showed that deletion of MAC1, CTR1, and PCA1 genes which involved in copper ion transport exhibited freeze-thaw sensitivity in compared with wild type. These genes are involved in copper ion uptake to a cell under a copper deficiency condition or in copper ion homeostasis, suggesting that it may be related between freeze-thaw injury and copper ion transport. To determine the effect of supplementation of copper ion on cells after freeze-thaw treatment, cell viability, intracellular superoxide dismutase (SOD) activity, and intracellular levels of reactive oxygen species (ROS) were examined by various copper ion condition medium. The results showed that intracellular SOD activity was increased and intracellular levels of ROS were decreased by supplementation of copper ion, but there was no significant difference in cell viability. These results of the present study may suggest that copper ion concentration in yeast cell after freeze-thaw treatment is important to recovery from freeze-thaw injury due to redox control of intracellular levels of ROS, but copper ion did not directly affect cell viability.

Project description:Copper is both an essential metal for life, but is also toxic at high concentrations. In mammalian cells, two copper transporters are known, one localized at the plasma membrane (CTR1) and one localized in late endosomes-lysosomes (CTR2). In order to gain insights on the possible influence of the importing pathway on cellular responses to copper, two copper challenges were compared, one with copper ion, which is likely to use preferentially CTR1, and one with a copper-polyacrylate complex, which will likely to be internalized via the endosomal pathway and therefore use preferentially CTR2. A model system consisting in the J774A1 mouse macrophage system, with a strong endosomal/lysosomal pathway, was used. In order to gain wide insights into the cellular responses to copper, a two-dimensional gel-based proteomic approach was used. The proteomic results were validated by targeted experiments, and showed differential effects of the import mode on general cellular physiology parameters such as the reduced glutathione content, the actin cytoskeleton or the mitochondrial potential, but also on more specialized macrophage functions such as phagocytosis or inflammatory responses.

Project description:Our hypothesis is that copper modulates the activity of multiple intracellular signal transduction pathways to affect transcription. We have previously shown that copper activates transcription through both metal- and oxidative stress-responsive signal transduction pathways. Since the global molecular mechanisms underlying copper toxicity have not been well elucidated in humans, we have profiled transcriptome changes in HepG2 cells exposed to 100, 200, 400 and 600 uM copper for 4, 8, 12 and 24 hours using a human oligonucleotide microarray. Differentially expressed genes were identified, and integrated into biological and functional pathways through Gene Ontology analysis. Global gene expression profile was overlaid onto biomolecular interaction networks and signal transduction cascades using pathway mapping and interactome identification. Total RNA was extracted from the stress-treated yeast cells by using a hot phenol method. Poly(A)+ RNA was enriched from total RNA by using an Oligotex dT30 (Super) mRNA purification kit (Takara Bio, Ohtsu, Japan). cDNA synthesis, cRNA synthesis, and labeling were performed according to the Affymetrix user’s manual (Affymetrix, Santa Clara, USA). Biotinyated cRNA was fragmented and then used as a probe.Affimetrix Yeast Genome 2.0 arrays (Affymetrix) were used as DNA microarrays. All experiments were done in triplicate independently.

Project description:Selenoprotein P is a hepatokine which is essential for maintaining systemic selenium homeostasis and is used as serum biomarker for the selenium status in humans. In addition to selenium, also copper homeostasis is mainly regulated by the liver coordinating systemic distribution of copper bound to ceruloplasmin or excreting surplus copper into the bile. Circulating selenium and copper concentrations are most often negatively correlating, e.g. during aging copper levels are increasing while selenium levels are decreasing. Based on these results, we addressed the question of how both trace elements interfere with each other using the liver-derived cell line HepG2. We observed that copper treatment resulted in a substantial increase of intracellular selenium concentrations. In parallel, extracellular SELENOP concentrations were drastically reduced while SELENOP accumulated within the cells. The same observation was made when using primary murine hepatocytes. Indeed, SELENOP was one of the proteins most strongly downregulated by copper in an untargeted secretome approach. Accumulation of hepatic copper is a characteristic of Wilson’s disease. Accordingly, SELENOP levels were decreasing in the serum of LPP rats, a model of Wilson’s disease starting at disease onset. Also, Wilson’s disease patients showed reduced serum SELENOP concentrations when circulating copper concentrations were low. This positive correlation between copper and ceruloplasmin and SELENOP was also observed in a GWAS analysis of EPIC-Potsdam samples identifying SNP rs11708215 as a modulating factor. Our data indicate that under conditions of a suboptimal selenium supply combined with a high copper intake a functional selenium deficit can become even worse because peripheral tissues such as the brain depend on SELENOP for their selenium supply.
