Project description:Mesenchymal stromal cells-derived small extracellular vesicles (MSC-sEVs) have recently attracted considerable attention because of their therapeutic potential in various immune diseases. We previously reported that MSC-sEVs could exert immunomodulatory roles in allergic airway inflammation by regulating group 2 innate lymphoid cell (ILC2) and dendritic cell (DC) functions. Therefore, this study aimed to investigate the therapeutic effects of MSC-sEVs on mature DC (mDC)-ILC2 interplay in allergic rhinitis (AR). Here, we isolated MSC-sEVs from induced pluripotent stem cells (iPSC)-MSCs using anion-exchange chromatography for the generation of sEV-mDCs. sEV-mDCs were co-cultured with peripheral blood mononuclear cells (PBMCs) from patients with AR or purified ILC2s. The levels of IL-13 and GATA3 in ILC2s were examined by flow cytometry. Bulk RNA-sequence for mDCs and sEV-mDCs was employed to further probe the potential mechanisms, which were then validated in the co-culture systems.

Project description:Bone marrow mesenchymal stromal cells (MSCs) regulate homeostasis and trafficking of cells of the blood lineage. In response to traumatic injury or infection, MSCs are believed to mobilize from the bone marrow, but it is largely unknown how egress into circulation impacts MSC function. Here we show that biomechanical forces associated with trafficking of MSCs from the bone marrow into the vasculature contribute uniquely to genetic signaling that reinforces MSC repression of immune cell activation. Laminar wall shear stress (LSS) typical of fluid frictional forces present on the lumen of arterioles stimulates increases in antioxidant and anti-inflammatory mediators, as well as an array of chemokines capable of immune cell recruitment. Importantly, LSS promotes a signaling cascade through COX2 that elevates prostaglandin E2 (PGE2) biosynthesis, permitting MSCs to suppress immune cell activation in the presence of inflammatory cues. Pharmacological inhibition of COX2 depleted PGE2 and impaired the ability of MSCs to block tumor necrosis factor-α (TNF-α) production, supporting a key role for PGE2 in the MSC immunomodulatory response to LSS. Preconditioning of MSCs by LSS ex vivo was an effective means of enhancing therapeutic efficacy in a rat model of traumatic brain injury, as evidenced by decreased numbers of apoptotic and M1-type activated microglia in the hippocampus and by retention of endogenous MSCs in the bone marrow. We conclude that biomechanical forces provide critical cues to MSCs residing at the vascular interface which influence MSC immunomodulatory and paracrine functions, thus providing unique opportunities for functional enhancement of MSCs used in therapeutic applications.

Project description:Mesenchymal stem cell-derived exosomes (MSC-Exos), noted for their minimal immunogenicity and robust immunomodulatory potential, have emerged as key regulators of immune cell fate. Nevertheless, a comprehensive understanding of their global impact on human NK cell proliferation and effector function remains elusive. In this study, we employed a standardized ex vivo expansion and exosome-priming system to systematically evaluate the regulatory effects of MSC-Exos on peripheral blood-derived NK cells via flow cytometry, cytotoxicity assays, single-cell transcriptomics, and proteomic profiling. Proteomic profiling identified an enrichment of FcγR-related signaling components within MSC-Exos, suggesting a potential role in enhancing antibody-dependent cellular cytotoxicity (ADCC).Collectively, our findings demonstrate that MSC-Exos bolster NK cell expansion and effector maturation through coordinated immuno-metabolic reprogramming, offering a strategic approach to enhancing the therapeutic efficacy of NK cell-based immunotherapies.

Here, we show a novel function for dedicator of cytokinesis 2 (DOCK2) in regulating extra-hematopoietic immune function of three independent stromal cell sources: induced pluripotent stem cells (iPSC)-derived mesodermal stromal cell (iPS-MSC), severe combined immunodeficiency (SCID) patient-derived fibroblasts and CRISPR/Cas9 knockout cells (iPS-MSCDOCK2KO). We first reprogrammed human mesenchymal stromal cells (MSC) into iPSC before differentiating the iPSCs back into MSC. Immature iPS-MSCs lacked immunosuppressive potential. Successive phenotypic maturation facilitated immunomodulatory function, while maintaining clonogenicity, comparable to parental MSCs. Sequential RNA-seq displayed time-dependent immune-related gene expression eventually resembling parental MSCs. SCID patient-derived fibroblasts harboring bi-allelic DOCK2 mutations also showed significantly reduced immunomodulatory capacity compared to non-mutated fibroblasts. CRISPR/Cas9-mediated DOCK2 knockout in healthy iPSCs resulted in significantly reduced immunomodulatory capacity, reduced F-actin stress-fibers, and a disturbed subcellular localization of CDC42 activation. We provide first evidence for extra-hematopoietic immunomodulation by the guanin exchange factor DOCK2. This suggests that persisting immune disease after successful blood stem cell transplantation in SCID patients could in part be due to loss-of-function DOCK2 mutations.

Project description:Human clinical trials in type 1 diabetes (T1D) patients are underway using mesenchymal stem cells (MSC) without prior validation in a mouse model for the disease. In response to this void, we characterized bone marrow-derived murine MSC for their ability to modulate immune responses in the context of T1D, as represented in non-obese diabetic (NOD) mice. In comparison to NOD-, BALB/c-MSC express higher levels of the negative costimulatory molecule PD-L1 and promote a shift toward Th2-like responses in treated NOD mice. In addition, transfer of MSC from resistant strains (i.e. NOR or BALB/c), but not from NOD mice, conferred disease protection when administered to prediabetic NOD mice. The number of BALB/c-MSC trafficking to the pancreatic lymph nodes of NOD mice was higher than in NOD mice provided autologous NOD-MSC. Administration of BALB/c-MSC resulted in reversal of hyperglycemia in 90% of NOD mice (p=0.002). Transfer of autologous NOD-MSC imparted no such therapeutic benefit, and in fact soft tissue and visceral tumors were uniquely observed in this setting (i.e. no tumors were present with BALB/c- or NOR-MSC transfer). These data provide important preclinical data supporting the basis for further development of allogeneic MSC-based therapies for T1D and potentially, other autoimmune disorders. To generate MSC, BM mononuclear cells were isolated from the femurs and tibiae of at least 5 mice in order to minimize cell variability. Cells are seeded in flasks at a concentration of 10x106/25 cm2 in M10 medium (DMEM medium [Cambrex, East Rutherford, New Jersey] containing 10% fetal calf serum [HyClone, Logan, Utah], 1% penicillin-streptomycin, and 1% L-glutamine [both from Cambrex]). To examine MSC in an inflammatory setting, 7.5x105 NOD- or BALB/c-MSC/well were cultured for 48h in 6-well plates in M10 medium containing 10 ng/ml recombinant murine IL-1beta (Peprotech, Rocky Hill, NJ). To ensure that our cultured cells had multipotent potential, we tested MSC P4 cultures for their ability to undergo differentiation into chondrocytes, osteocytes, and adipocytes as previously published.10 Chondrogenic differentiation was induced by 50 ug/ml ascorbic acid and 1 ng/ml TGF?-1 (Peprotech), osteogenic differentiation was induced by 50 ug/ml ascorbic acid, 10 mM sodium alpha-glycerophosphate, and 10-8 M dexamethasone, and adipogenic differentiation was induced by 10-7 M dexamethasone and 6 ng/ml insulin. MSC were analyzed for expression at passage 4 (P4). We examined the gene expression profile of NOD- (samples #1 to 4) and BALB/c-MSC (samples #5 to 8), obtained from normoglycemic NOD mice and age-matched BALB/c mice. A striking concordance in the pattern of gene regulation across the entire screen was observed when BALB/c-MSC genes were compared to the NOD-MSC 4 different culture of NOD derived MSC were compared to 4 differente cell culture of Balb/c derived MSC, Balb/c cell coltures were considered reference samples.

Project description:Bone marrow-derived mesenchymal stromal cells (MSC(M)) are promising immunomodulatory therapeutics for osteoarthritis (OA); however, mixed clinical efficacy has been reported. We present new 24-month follow-up data from our previous clinical trial investigating single intra-articular injection of autologous MSC(M) in knee OA, with analysis of responders and non-responders to probe differential baseline disease features and MSC(M) basal fitness. Analysis of patient-reported outcome measures (PROMs) showed responders had greater OA severity at baseline. Lower baseline levels of angiogenic/inflammatory synovial fluid biomarkers correlated to improved PROMs. A gene panel of putative immunomodulatory critical quality attributes (CQAs) distinguished MSC(M) donors classified as responders versus non-responders. Furthermore, an algorithm was developed to rank MSC(M) donors based on in vitro monocyte/macrophage polarization readouts and clinical efficacy. Lastly, microRNA-sequencing of MSC(M) revealed 69 microRNAs associated with responder status. Our data provide evidence for specific clinical phenotype features and biomarkers to stratify OA patients, as well as CQAs for quantifying basal immunomodulatory fitness of MSC(M) in knee OA.
