Mass spectrometry data of colon cancer cell SW480 treated with Caffeic acid phenyl ethyl

ABSTRACT: Caffeic acid acetophenate exhibits anti-tumor activity. We employed proteomics to investigate the targets of caffeic acid acetophenate in human colon cancer cells. The experimental group consisted of SW480 human colon cancer cells treated with 10μM caffeic acid phenethyl for 24 hours, while the control group was treated with an equivalent volume of DMSO. Non-standard quantitative mass spectrometry was utilized to analyze differentially expressed proteins between the two groups and identify the specific target of caffeic acid acetophenate on colon cancer cells.

Project description:Transcriptional profiling of human colon cancer SW480 cells comparing control untreated SW480 cells with cells stably transfected with LRP16 treated with or without etoposide (50 μM) for the indicated periods (0,1hour, 3hours).An exploratory microarray analysis was performed with mRNA extracted from clutured SW480 cells transfected with LRP16 or control plasmid that were treated with or without etoposide. Total RNA of colon cancer cells stably transfected with vector control and LRP16 treated with or without etoposide (50 μM) for the indicated periods was isolated and purified using RNeasy Kit (Qiagen, Hilden, Germany). Integrity of RNA was assessed by using an Agilent BioAnalyser 2100 (Agilent Technologies).

Project description:Cancer stem-like cells are defined as small population of cancer cells which has, higher tumor-initiating ability, self-renewing ability and differentiation ability. In this experiment, transcription profile of cancer stem-like cells derived from human colon cancer line cell SW480 was investigated. Cancer stem-like cells were isolated as side population (SP) cells by Hoechst33342 dye staining from human colon cancer line cell SW480. Non-cancer stem-like cells were isolated as main population (MP) cells. RNAs were isolated from SP cells and MP cells derived from SW480 cells and array experiment was perfromed in dye swaped fashion.

Project description:STAT3 is determined to play a crucial role in JAK/STAT signaling pathway. The mutations and copy number alteration of STAT3 can cause cancers and other diseases. The 317-567 amino acid domain of STAT3 protein contributes to binding with genomic DNA for gene activation. However, the impact of the mutations within DNA binding domain of STAT3 on the genomic binding pattern is not fully understood. Stat3 was deleted the 400-411 amino acid residue of \\"NNGSLSAEFKHL\\" using CRISPR-CAS9 in human colon cancer cells SW480. The cells were performed STAT3 ChIP-seq to investigate the genome-wide binding pattern of STAT3 between wild type and mutant STAT3 of SW480.

Transcription profiling of effected genes by compound BIO,3G4 and knockdown of MED23

Project description:Drug discovery, inhibition of WNT pathway and identification of molecular targets. Prostate EPT1cells were treated with 6-bromoindirubin-3ΓÇÖ-oxime(BIO) or DMSO for 24 hours, colon cancer SW480 cells were treated with compound 3G4 for 24 and 72 hours, SW480 cells were transfected with MED23 knockdown shRNA vector for 48 hours. Cells were harvested and total RNA was extracted using Qiazol method followed by Agilent DNA microarray analysis.
