Proteome profiling of BV2 cells with altered ALDH2 enzyme activity


ABSTRACT: Aldehyde dehydrogenase 2 family member (ALDH2) is a key mitochondrial dehydrogenase that functions in the detoxification of acetaldehyde, and endogenous 4-hydroxy-2-nonenal (4-HNE) and malondialdehyde generated by polyunsaturated fatty acid peroxidation. Whether ALDH2 activity altered microglia cell roles in CNS is unclear. The aim of this study was to reveal proteins expression changes induced by altered ALDH2 activity. After bioinformatic analysis, we detected a total of 3077 confident proteins. differentially expressed proteins were filtered with adj. P value <0.05 and expression fold change <0.83 or >1.20. Reactome pathway analysis revealed dysregulated metabolism of lipids and metabolism.

In the ALDH2 rs671 variant, a guanine changes to an adenine, resulting in a dramatic decrease in the catalytic activity of the enzyme. Population-based data are contradictory about whether this variant increases the risk of Alzheimer's disease.
