Label-free quantitative proteomics analysis of ASPC-1 and H358

ABSTRACT: A total of one million AsPC-1 and H358 cells were treated with DMSO or 100 nM MCB-36 for 12 hours, after which the cells were collected by centrifugation and stored at -80°C until further analysis. Proteins were reduced, alkylated, and purified by chloroform/meth anol extraction prior to trypsin digestion. The search parameters included tryptic digestion with up to two missed cleavages allowed, carbamidomethylation (C) as a fixed modification, and oxidation (M) and acetylation (protein N-terminal) as variable modifications. Peptide identification was carried out with an initial precursor ion mass tolerance of 10 ppm and a fragment mass tolerance of 0.02 Da.
