RiBi project double mutants

Project description:The RiBi project investigated the role of chromatin factor Spt6 in ribosome biogenesis. In this experiment we performed transcription profiling of Saccharomyces cerevisiae single and double deletion mutants involved in ribosome assembly. The single mutants examined in this experiment were: arx1-deletion, dbp7-deletion, mrt4-deletion, zuo1-deletion and spt6 -140 mutant. The double mutants were spt6-140 mutant plus arx1-deletion; spt6-140 mutant plus dbp7-deletion; spt6-140 mutant plus mrt4-deletion; spt6-140 mutant plus rsa1-deletion; and spt6- 140 mutant plus zuo1-deletion. The experiment was performed using a dye-swap design where a single wildtype culture was used as a common reference pool.

The human Whole miRNOme project version 1

Project description:This project analyzes peripheral blood profiles of controls and patients of 14 different diseases, all collected, measured, and analyzed using exactly the same SoP. Since miRNAs are known to be valuable diagnostic markers we asked whether respective patterns of patients can be detected in peripheral blood samples rather than in biopsies. The project aimed at an impoved understanding of complex profiles rather than single markers. Thus, a high-throughput technique was necessary, profiling all known miRNAs integratively and combining different diseases to achieve a high degree of specificity.

Project description:For the GST pull-down of DRG ganglion, the GST- or Tmc7-GST-associated beads were further incubated with cell lysates derived from DRG ganglion. The pulled-down proteins were then separated by SDS-PAGE gel electrophoresis and subsequently stained with silver staining Kit. Finally, the target molecular weight bands and undenatured beads were analyzed by proteomic analysis, respectively.

Oxygen-dependent interaction between FBXL5 and CIA targeting complex regulates iron homeostasis

Project description:The iron sensing protein FBXL5 is the substrate adaptor for a SKP1-CUL1-RBX1 E3 ubiquitin ligase complex that regulates the degradation of iron regulatory proteins (IRPs). Here we describe a mechanism of FBXL5 regulation involving its interaction with the cytosolic Fe-S cluster assembly (CIA) targeting complex comprised of MMS19, FAM96B, and CIAO1. We demonstrate that the CIA targeting complex promotes the ability of FBXL5 to degrade IRPs. In addition, the FBXL5-CIA targeting complex interaction is regulated by oxygen tension displaying a robust association in 21% O2 that is severely diminished in 1% O2 and contributes to O2-dependent regulation of IRP degradation. Together, these data identify a novel oxygen-dependent signaling axis that links IRP-dependent iron homeostasis with the Fe-S cluster assembly machinery.
