Proteomics Analyses of Mycelium and Cysts of Phytophthora capsici

Project description:In this project, we have performed a comparative analysis of the proteomes of the mycelium and cysts of P. capsici. The identified proteins were functionally classified, which provides the basis for future research on these developmental stages of P. capsici and closely related species. Many of the proteins enriched in either the mycelium or cysts were found to contribute specific functions in these two life stages. Proteins enriched in mycelium were essentially associated with vegetative growth, whereas those enriched in cysts were primarily related to development-specific processes associated with the initiation of infection. The data provide a better understanding of the metabolic pathways underlying the mycelial and cyst developmental stages of P. capsici and point to potential targets of anti-oomycete compounds at each of these stages.

BCR repertoire of B cells with distinct developmental timings

Project description:B cells are known to have different properties and BCR repertoires depending on the time of development. Our objective is to investigate the BCR repertoire of B cells across embryonic, neonatal, and adult stages, particularly in cells with a RAG2 expression history. We focus on sequencing and analyzing the immunoglobulin heavy chain (IGH) genes of these cells to understand their BCR diversity and specificity. Additionally, we explore the relationship between B-1a cells and bone marrow IgM+ plasmablasts/plasma cells, aiming to shed light on the development and function of B-1a cells in the immune system.

Developmental transcriptomics of the model caterpillar fungus Cordyceps militaris

Project description:The RNA-Seq technique was used to investigate the differentially transcribed genes of the caterpillar fungus Cordyces militaris. It was calculated that >63% of total annotated genes were expressed, including >55% of putative pathogen-host interaction genes. Overall, the fruiting-body development and mycelium growth by C. militaris activated high expression levels of genes in association with translation and post-translational modifications. Otherwise, there were many differentially expressed genes when the fungus was grown on insect pupae to different stages or in liquid medium. In general, the transcriptional profiles were more similar with each other during fungal fruiting-body developmental stages than during growth in a liquid culture.

Genome-wide gene expression analysis of Fusarium graminearum isolate PH-1 in spores and mycelium

Project description:Using RNA-Seq data from the mycelium of Fusarium graminearum, we found weakly expressed regions on all of the four chromosomes that exactly matched with non-conserved regions. Comparison of gene expression between two different developmental stages (spores and mycelium) showed that the expression of genes in conserved regions is stable, while gene expression in non-conserved regions is much more influenced by developmental stage. Examination of gene expression in two developmental stages (spores and mycelium). Three biological replicates were performed for each stage.

Saprolegnia parasitica proteome

Project description:In this project, we made a comprehensive quantitative proteomic analysis of four defined developmental stages of S. parasitica (mycelium, primary cysts, secondary cysts and germinated cysts) to gain greater insight into the types of proteins specifically linked to each stage. A total of 2423 unique proteins were identified using qualitative (gel-based) and quantitative (iTRAQ, isobaric tags for relative and absolute quantitation) approaches. Of these, 358 proteins, associated with various biological processes, were found to be significantly enriched between different life cycle stages of S. parasitica. The transcript abundance of several cyst and mycelium enriched proteins was also checked by quantitative real-time PCR. This is the first large scale proteomic analysis on Saprolegnia and the data from this study will enhance our current knowledge about this pathogen by identifying biological processes key to each developmental stage.
